GENOWEFA BONCZAR, MONIKA WSZOŁEK, MARIA WALCZYCKA, ANETA ŻEBROWSKA, KRZYSZTOF MACIEJOWSKI WPŁYW WYBRANYCH CZYNNIKÓW NA AKTYWNOŚĆ WODY I JAKOŚĆ MIKROBIOLOGICZNĄ MIĘKKICH SERÓW Z MLEKA OWCZEGO S t r e s z c z e n i e Materiał badawczy stanowiło mleko owcze i miękkie sery podpuszczkowe. W mleku oznaczono zawartość: suchej masy, tłuszczu, związków azotowych ogółem oraz kwasowość miareczkową, pH i gęstość. Mleko podzielono na dwie części -jedną część spasteryzowano w temp. 72 °C, pozostałej nie pasteryzowano. Do obu części mleka dodano podpuszczkę (o mocy 1:1000). Po uzyskaniu skrzepu i pokrojeniu uformowano sery. Po 24 h jeden ser z mleka pasteryzowanego i jeden z surowego poddano soleniu w solance o stężeniu 16 % przez 1 h. Podobnie po jednym serze z mleka surowego i jednym z pasteryzowanego zaparzano w wodzie w temp. 70 °C przez 3 min.Świeże sery -surowe i pasteryzowane poddano analizie, która obejmowała oznaczenie zawartości suchej masy, pH, aktywności wody, parametrów tekstury, a także badania mikrobiologiczne, dotyczące oznaczenia ogólnej liczby drobnoustrojów, liczby bakterii z grupy coli, liczby bakterii kwaszących oraz liczby pleśni i drożdży. Po 14 dniach przechowywania w temp. 4 °C w serach oznaczano zawartość suchej masy, aktywność wody, pH, parametry tekstury i przeprowadzono badania mikrobiologiczne. Wyniki opracowano statystycznie.Stwierdzono, że pasteryzacja wpłynęła na obniżenie badanych parametrów w serach, statystycznie istotnie w przypadku pH, spoistości i żujności oraz liczby drobnoustrojów (ogólnej, coli i drożdży). Solenie istotnie zredukowało aktywność wody w serach. Liczba drobnoustrojów (ogólna, coli i drożdży) zmniejszyła się nieznacznie, lecz w sposób statystycznie nieistotny. Zaparzanie spowodowało istotny statystycznie wzrost zawartości suchej masy oraz parametrów tekstury: twardości, spoistości, sprężystości i żujności. Współczynniki korelacji między aktywnością wody i pozostałymi analizowanymi parametrami serów zmieniały się w zależności od zastosowanego czynnika doświadczalnego. Jej wartość świadczy o dostępności wody dla mikroorganizmów, a tym samym o możliwości ich rozwoju, co wpływa na jakość produktów i ich trwałość. Spośród produktów mleczarskich mleko w proszku odtłuszczone charakteryzuje się aktywnością wody 0,254 i trwałością wynoszącą 6 miesięcy, podczas gdy aktywność wody sera twarogowego wynosi 0,955, a jego trwałość 48 h [4]. Gdy aktywność wody wynosi powyżej 0,95 w mleku i produktach mleczarskich rozwijają się bakterie z rodzaju Pseudomonas, Escherichia, Proteus, Shigella, Klebsiella Clostridium perfringens i niektóre drożdże, od 0,91 -0,95 Salmonella, Clostridium botulinum, Serratia, Lactobacillus, Pediococcus, Bacillus i niektóre pleśni...
Przedstawiono wyniki badań dotyczące czynników warunkujących zawartość cholesterolu i produktów jego utleniania (PUCh) w wyrobach mleczarskich. Opisano wpływ gatunku i rasy zwierząt, okresu laktacji, sposobu żywienia oraz stanu zdrowotnego wymienia na poziom cholesterolu w mleku surowym. Uwzględniono wpływ czynników technologicznych: zwłaszcza obróbki termicznej i homogenizacji mleka, dodatku kultur starterowych o zróżnicowanym składzie oraz warunków przechowywania na poziom cholesterolu w produktach mleczarskich. Scharakteryzowano czynniki decydujące o powstawaniu PUCh w wyrobach mleczarskich. Cholesterol jest związkiem nierozpuszczalnym w wodzie, dlatego też w płynach ustrojowych transportowany jest jako składnik lipoprotein-globularnych cząstek w kształcie miceli, zbudowanych z hydrofobowego rdzenia, w którego skład wchodzą triacyloglicerole i estry cholesterolu, otoczony powłoką amfipatyczną, zbudowaną z białka, fosfolipidów i cholesterolu [19]. Ze względu na właściwości fizyczne i funkcjonalne, lipoproteiny dzieli się na klasy:
Z mleka krowiego poddanego pasteryzacji i zaszczepionego kulturami starterowymi DVS: YC-180 (Lactobacillus delbrueckii subsp. bulgaricus oraz Streptococcus thermophilus), YO-MIX 207 (Lactobacillus delbrueckii subsp. bulgaricus, Streptococcus thermophilus, Lactobacillus acidophilus, Bifidobacterium lactis), LA-5 (Lactobacillus acidophillus) i BB-12 (Bifidobacterium bifidum) wyprodukowano napoje fermentowane. W mleku i wyprodukowanych z niego 1-i 14-dniowych napojach fermentowanych oznaczono zawartość tłuszczu, pH i kwasowość miareczkową, a także zawartość cholesterolu metodą enzymatyczną z użyciem oksydazy cholesterolowej. Mleko pasteryzowane zawierało średnio 20,0 mg/100 g cholesterolu, a jego tłuszcz-5,64 mg/g. W wyprodukowanych jednodniowych napojach fermentowanych z udziałem szczepionek YC-180, YO-MIX 207, LA-5 i BB-12 stwierdzono statystycznie istotną (p ≤ 0,05) redukcję poziomu cholesterolu, w porównaniu z mlekiem, odpowiednio do [%]: 49,4, 41,3, 44,8 i 45,9 początkowej jego zawartości w 100 g mleka i odpowiednio do [%]: 54,9, 44,3, 46,3 i 48,0 w 1 g tłuszczu mleka. Kultury starterowe wykazały zróżnicowanie pod względem zdolności do zmniejszania zawartości cholesterolu, przy czym największe-szczepionka YO-MIX 207, a najmniejsze-YC-180. Podczas 14-dniowego przechowywania napojów w warunkach chłodniczych zaobserwowano dalsze zmniejszenie poziomu cholesterolu w napojach i w ich tłuszczu, średnio o około 15 %.
Celem pracy było określenie wpływu pasteryzacji (jej braku, pasteryzacji: w temp. 72 °C/15 s, w temp. 85 °C/15 s, w temp. 95 °C/10 min) i homogenizacji mleka (jej braku, homogenizacji przy 6 MPa) na zawartość cholesterolu w mleku i wyprodukowanych z niego serach podpuszczkowych typu bundz. Stwierdzono, że oba procesy wpłynęły statystycznie istotnie (p ≤ 0,05) na zawartość cholesterolu zarówno w mleku, jak i w serach. Pasteryzacja spowodowała zwiększenie zawartości cholesterolu w mleku. W serach nastąpiło natomiast zmniejszenie jego poziomu i było ono proporcjonalne do wzrostu temperatury pasteryzacji. W mleku surowym i w serach z niego wyprodukowanych zawartość cholesterolu wynosiła odpowiednio: 14,41 i 115,61 mg/100 g, natomiast po pasteryzacji w temp. 95 °C/10 min odpowiednio: 19,34 i 55,01 mg/100 g. Proces homogenizacji mleka surowego wpłynął na zmniejszenie poziomu cholesterolu zarówno w mleku, jak i w serach, odpowiednio do: 5,55 i 88,48 mg/100 g. Po analizie równoczesnego wpływu obu czynników wykazano, że wraz ze wzrostem temperatury pasteryzacji poziom cholesterolu w serach w większym stopniu zmniejszał się wtedy, gdy były one wyprodukowane z mleka homogenizowanego.
