Rei der Darstellung und Bearbeitung des o -N i t r o b e n z y l -m e t h y l -1; e t o n s , das urspriinglich zu einer anderen Untersuchung herangezogen wurde, fie1 die Tatsache auf, daB rnit Mydroxylamin-Chlorhydrat zwei v e rs c h i e d e n e O x i m e erhalten warden, obwohl von vornherein in diesem I:alle rnit Raumisomerie nicht gerechnet werden konnte. Es ist zwar bekannt, da13 die Oxime der unsymmetrisch gebauten Benzophenone, des Benzoins und des B e n d s in stereoisomeren Formen auftreten. Dagegen wurde Rauniisomerie bei den Oximen des Acetophenons, des Propiophenons usw., des Benzylmethyl-ketons oder des Desoxy-benzoins nicht beobachtet, also iiberall dort, WO die Ketogruppe direkt rnit einem Alkyl (oder einem phenyl-substituierten Alkyl) verbunden ist. Aus diesen Tatsachen heraus neigte man zur Ansicht, da13 die Oxime der gemischt fett-aromatischen Ketone in der Regel nur in einer Form auftreten. Wahrend also die Gruppierung C,H,. CO. CH,, CsH5. CO. CH,. CH, usw. das Auftreten stereoisomerer Formen zu verhindern scheint, andern sich die Verhaltnisse, wenn z. B. das Acetophenon in der Seitenkette bromiert wird: denn das w -B r o ma c e t o p h e n o nl), C,H,. CO. CH,Br, vermag stereoisomere Oxime zu liefern,). Andererseits ist sowohl beim Desoxy-benzoin, C,H,. CH, . CO. C,H,, wie bei seinem Nitrierungspunkt, dem o-Nitro-desoxybenzoin, C,H,(NO,) . CH,. CO. C,H5 s), nur je ein Oxim bekannt; um so auffallender ist daher die Tatsache, da13 die Einfiihrung der o-standigen Nitrogruppe in das Benzyl-methyl-keton wiederum das Auftreten zweier stereoisomerer Formen bedingt.Zunachst ein Wort iiber die Reindarstellung des der Untersuchung zugrunde liegenden Ketons : Erstmals wurde das o-h'itrobenzyl-methyl-keton von A. v. B a e y e r und J ackson,) durch Nitrierung von Phenyl-aceton erhalten. Die genannten Porscher legten jedoch keinen Wert auf seine Isolierung, sondern verarbeiteten es sofort in unreinem Zustande auf a-Methylindol. Der Reindarstellung des o-h'itro-ketons stellten sich zunachst Schwjerigkeiten in den Weg insofern, als bei der Nitrierung rnit iiberschussiger rauchender Salpetersaure (mit berechneter hlenge Nitriersaure verlauft der ProzeB in der Hauptsache in ganz anderer Richtung, wie aus dem experimentellen Teil ersichtlich) eine ganze Reihe der verschiedensten Nitroprodukte entsteht, aus denen das o-Nitrobenzyl-methyl-keton weder durch fraktionierte Krystallisation noch durch Destillation (selhst im Hochvakuum) infolge der Zersetzlichkeit vor allem der hoher nitrierten Produkte isoliert werden konnte. Erst die B e h a n d l u n g d e s X i t r i e r g e m i s c h e s i n a l k o h o l i s c h e r Losung rnit H y d r o x y l a m i n ergab eine Methode, uni das g e s u c h t e K e t o n i n G e s t a l t s e i n e s O x i m s q u a n t i t a t i v v o n d e n a n d e r e n S u b s t a n z e n z u t r e n n e n , ein Weg, der sich vielleicht auch bei anderen Gelegenheiten zur Isolier...
Attempts were undertaken to develop cyclic β‐casomorphin‐5 analogs with improved opioid activity profiles by deletion of the glycine residue in position 5, leading to analogs structurally related to the opioid peptide morphiceptin (H‐Tyr‐Pro‐Phe‐Pro‐NH2). The tetrapeptide sequence Boc‐Tyr(tBu)‐D‐Xaa‐Phe‐Yaa‐OH (Xaa = Lys, Orn, A2bu; Yaa = Pro in L‐ or D‐configuration) was used to study the influence of ring size and chirality on the yield of cyclization between the ω‐amino group of Xaa and the C‐terminal carboxyl group. In all cases the cyclization reaction was performed under identical experimental conditions to allow a direct comparison with regard to yield and homogeneity. The reaction products were purified by crystallization and liquid chromatography, and were characterized by HPLC, TLC, electrospray mass spectrometry and 1H‐NMR spectroscopy. In none of the reactions performed with the cyclization precursors containing proline in the L‐configuration could a cyclic monomer be detected, and the cyclodimer (7–9) was the exclusive product in each case. Cyclodimerization was also the favored reaction in the attempted formation of the 11‐membered ring of the cyclic [D‐A2bu2, D‐Pro4]‐morphiceptin analog 12, since only traces of the monomer were found. In the case of both the [D‐Lys2, D‐Pro4]‐analog 10 and the [D‐Orn2, D‐Pro4]‐ analog 11, the cyclomonomer/cyclodimer ratio was about 80: 20. The cyclic monomers 10 and 11 showed high opioid activity in the μ‐receptor‐representative guinea pig ileum assay (IC50= 2–5 nM) and in the δ‐receptor representative mouse vas deferens assay (IC50= 50–60 nM), whereas the potency of the cyclodimers was 2–3 orders of magnitude lower in both in vitro bioassays.
