Acetaminophen, or paracetamol is an over-the-counter analgesic generally used by all groups of people, including pregnant women. The present investigation aims to elucidate the effects of acetaminophen on the development of zebrafish Danio rerio in which embryogenesis is ex utero. Developing eggs (n = 30) were exposed to different doses (0, 1, 5, 10, 50 and 100 microg L(-1)) of the drug in triplicate and observations were made hourly until gastrulation and once in 24 h thereafter for seven consecutive days. Acetaminophen induced anomalies at different levels of development in a dose-dependent manner, causing impairment in (1) the early development, (2) hatching, (3) organogenesis (by altering the rate of apoptosis), (4) larval growth and morphometry, (5) tail and tail-fin formation, (6) pigmentation and (7) larval behavior and survival. The results of the present study clearly reveal that acetaminophen interfered with the normal embryonic development, growth, behavior and survival of D. rerio larvae.

An important limiting factor in assisted reproduction treatment success rates is oocyte quality. In spite of improved results through several important innovations, the pregnancy rate per collected oocyte remains far too low. In order to improve this situation, it is necessary to learn more about fundamental factors modulating follicular development patterns. FSH is known to be the driving force for follicle development, but it is not yet understood how its multifarious functions are controlled and modulated. Evidence is accumulating that FSH glycoforms may be the key to this mystery. Intact follicle culture is a useful tool for the clarification of the actions of the different isoforms because the follicle unit is maintained and allowed to develop through several critical stages. Additionally important is the availability of the oocyte for functional evaluation. Because of these features, relationships can be uncovered that are not revealed with single cell test systems. The results so far obtained with this system suggest that follicle development pattern and oocyte quality is strongly influenced by FSH glycoform range, and that the requirements of the follicle may shift during progress through different stages of development. More studies are required, but these findings already suggest that the physiological shifts of circulating FSH glycoforms may indeed be important, and that attention should be paid to the glycoform distribution of exogenously applied FSH.

Tartrazine (TTZ) is an azo dye used as a colorant in food products, drugs, and cosmetics. The present study evaluates the impacts of TTZ on embryonic development of zebrafish ( Danio rerio). Laboratory-raised D. rerio embryos (n = 20/concentration) were exposed to graded dilutions of TTZ (0, 0.1, 1, 2, 3, 4, 5, 10, 20, 30, 40, 50, 75, and 100 mM) from gastrulation stage (5.25 hours postfertilization [hpf]) until hatching and developmental trajectory was traced up to day 7. The no observed effect concentration (NOEC), median lethal concentration (LC), median effective concentration (EC), and teratogenic index (TI) were calculated. Exposure of embryos to < 10 mM TTZ had no effects; 20 to 30 mM TTZ caused tail bending, cardiac and yolk sac edema in 50% of larvae; in 30 to 50 mM TTZ-exposed embryos the heart rates declined along with the above mentioned deformities, causing mortality within 96 to 144 hpf; development ceased completely at 75 to 100 mM concentration. The NOEC and LC were recorded at 5 and 29.4 mM dose, respectively. The EC values for heart rate, cardiac edema, tail bending, and hatching success were at 59.60, 53.81, 98.28, and 58.97 mM with TI quotient 0.49, 0.54, 0.29, and 0.49, respectively. We conclude that TTZ is not embryo toxic/teratogenic for zebrafish embryos up to a dose level of 10 mM concentration.
