Small non-coding RNAs (sncRNAs) are indispensable for proper germ cell development, emphasizing the need for greater elucidation of the mechanisms of germline development and regulation of this process by sncRNAs. We used deep sequencing to characterize three families of small non-coding RNAs (piRNAs, miRNAs, and tRFs) present in Sus scrofa gonads and focused on the small RNA fraction present in both male and female gonads. Although similar numbers of reads were obtained from both types of gonads, the number of unique RNA sequences in the ovaries was several times lower. Of the sequences detected in the testes, 2.6% of piRNAs, 9% of miRNAs, and 10% of tRFs were also present in the ovaries. Notably, the majority of the shared piRNAs mapped to ribosomal RNAs and were derived from clustered loci. In addition, the most abundant miRNAs present in the ovaries and testes are conserved and are involved in many biological processes such as the regulation of homeobox genes, the control of cell proliferation, and carcinogenesis. Unexpectedly, we detected a novel sncRNA type, the tRFs, which are 30–36-nt RNA fragments derived from tRNA molecules, in gonads. Analysis of S. scrofa piRNAs show that testes specific piRNAs are biased for 5′ uracil but both testes and ovaries specific piRNAs are not biased for adenine at the 10th nucleotide position. These observations indicate that adult porcine piRNAs are predominantly produced by a primary processing pathway or other mechanisms and secondary piRNAs generated by ping-pong mechanism are absent.
Abstract:The surface modification of titanium alloys is an effective method to improve their biocompatibility and tailor the material to the desired profile of implant functionality. In this work, technologically-advanced titanium alloys-Ti-15Mo, Ti-13Nb-13Zr and Ti-6Al-7Nb-were anodized in suspensions, followed by treatment in alkali solutions, with wollastonite deposition from the powder phase suspended in solution. The anodized samples were immersed in NaOH or KOH solution with various concentrations with a different set of temperatures and exposure times. Based on their morphologies (observed by scanning electron microscope), the selected samples were investigated by Raman and X-ray photoelectron spectroscopy (XPS). Titaniate compounds were formed on the previously anodized titanium surfaces. The surface wettability significantly decreased, mainly on the modified Ti-15Mo alloy surface. Titanium alloy compounds had an influence on the results of the titanium alloys' surface modification, which caused the surfaces to exhibit differential physical properties. In this paper, we present the influence of the anodization procedure on alkali treatment effects and the properties of obtained hybrid coatings.
In this paper, the surface modification of titanium alloys Ti-15Mo, Ti-13Nb-13Zr, and Ti-6Al-7Nb is presented as a material for dental implants. The conditions of the plasma electrolytic oxidation process and alkali treatment were designed in this way to enhance the biological properties of the surface of promising Ti alloys. The differences in their surface morphology and, consequently, in their biological properties were discussed. The bioactivity of the samples was examined in vitro using simulated body fluid, and Saos-2 osteoblast cells. On all the samples, characteristic apatite particles were formed. However, compared to as-ground, natively-oxidized bare alloys, the plasma electrolytic oxidation (PEO)-modified surface of the Ti-13Nb-13Zr alloy showed the highest cytocompatibility for Saos-2 osteoblast cells, and a beneficial gain of cytocompatibility was also achieved in the treated sample of Ti-6Al-7Nb. In contrast, the modification of the Ti-15Mo alloy did not influence the adhesion and proliferation of osteoblast cells.
This article concerns research on washing effectiveness of a high-pressure washing device. Three types of investigations were carried out: Laboratory tests on washing of crushed-stone and gravel aggregate, as well as pilot-scale tests on crushed-stone aggregate. Laboratory tests were conducted for four changeable parameters and pilot-scale investigations for three parameters. All tests were performed based on factorial experiment procedure. For each type of material there was built a model based on multiple regression method. Results of investigations showed that operating pressure in the washer has the highest effect on obtained washing effectiveness, measured as a degree of dust impurities’ removal from feed. The second parameter with the highest impact on washing process effectiveness was the maximum particle of feed, and its relationship to the obtained washing effectiveness appeared inversely proportional. For the crushed-stone aggregate the susceptibility of washing also appeared significant in models.
In the present work, deposition of poly(sebacic anhydride) PSBA loaded by amoxicillin, cefazolin, or vancomycin on a previously anodized Ti‐15Mo surface is presented. PSBA loaded by the drug was deposited so as not to lose the functionality of the porous oxide layer microstructure. The morphology was evaluated using scanning electron microscopy, surface roughness, and wettability. The drug concentration was evaluated using high‐performance liquid chromatography. It was determined that the drugs were loaded into coatings in the range of 35.2–122.87 μg/cm2 of Ti sample. The drugs released more than 16% after 0.5 hr of the hybrid coating immersion in artificial saliva. After 3 days, the PSBA coatings were degraded by 51.3 mol %, and after 7 days by 77.8 mol %, which makes it possible to load the material by different, biologically active substances. An antimicrobial investigation of Staphylococcus aureus (DSM 24167) and Staphylococcus epidermidis (ATCC 700296) confirmed the activity of the hybrid layers against the pathogens. Hybrid layer with vancomycin best inhibits the adhesion of the bacteria, whereas coatings with amoxicillin and cefazolin showed a much better bactericidal activity. In this article, the difference in the obtained results is discussed, as well as the possibility of the application of this functional material in biomedicine.
