Leptin administered intracerebroventricularly (icv) evokes the resuscitating effect in haemorrhage-shocked rats, and the action is associated with activation of the histaminergic system. The aim of the present study was to examine the possible role of the histamine H3 receptor in leptin-evoked cardiovascular effects.
Hemorrhagic shock has a potential to be life-threatening when it is not treated. The main causes of hemorrhagic shock involve: (1) forces causing injury; and (2) diseases that can cause hemorrhage., Therefore, due to the causes of hemorrhagic shock and the life-threatening potential, the search for new methods of shock treatment is extremely valuable to the modern medicine. The aim of this study was to investigate the influence of clobenpropit in the model of hemorrhagic shock. The experiments were conducted in 110 adult male Wistar rats weighing between 205 and 470g. 1, 2 and 5 μmol/kg of intravenous H3 receptors reverse agonists, clobentropit, and/or 1, 5 and 10 μmol/kg H3 receptor agonist, imetit, were used as general anesthetics. Irreversible hemorrhagic shock was induced by the paused bleeding until the mean arterial pressure (MAP) lowered to the level of 20–25 mmHg. It was proved that, in cases of critical hypotension, clobenpropit triggered a dose-dependent increase of: systolic blood pressure (SBP), diastolic blood pressure (DBP), MPA and heart rate (HR) of rats with critical hypotension. The most significant changes in hemodynamic parameters were achieved by administrating dosages of 2 mmol/kg. This resulted in the survival rate increase to up to 100%. However, imetit did not trigger any hemodynamic changes nor an increase in SBP, DBP, MAP or HR. Furthermore, it was found that the premedication with prazosin, yohimbine, 6-hydroxydopamine and the vasopressin V1a receptor antagonist blocked the effects of clobenpropit. Additionally, premedication with propranolol, captopril and ZD 7155 did not cause any significant changes in the measured hemodynamic parameters. In conclusion, after an intravenous injection clobenpropit, the inverse agonist of H3 histamine receptors/agonist of histamine receptors H4, causes a resuscitating effect on rats in hemorrhagic shock. Moreover, such effect is based on the effector mechanisms of sympathetic nervous system and vasopressin.
Activation of the central histaminergic system leads to the reversal of experimental haemorrhagic shock, whereas neuropeptide Y (NPY) administered intracerebroventricularly (icv) induces a depressor effect in haemorrhagic hypotension. Since histaminergic neurons of the tuberomammillary nucleus receive input from neurons producing NPY localized in the caudal magnocellular nucleus of the hypothalamus, the aim of the study was to examine (1) the cardiovascular effects of the NPY type 1 (Y 1) receptor antagonist in haemorrhagic shock and (2) the possible involvement of the histaminergic system in this action.

Nie wykazano korelacji pomiędzy niekorzystnymi skutkami a zawartością amin w żywności. Można przypuszczać, że stężenie histaminy powyżej 100 mg/kg w produktach spożywczych nie jest zalecane. Dzięki ścisłym reżimom technologicznym podczas produkcji żywności częstość zatruć pokarmowych histaminą, wynikających z zanieczyszczeń bakteryjnych, jest sporadyczna.
Wstęp:Pobudzenie ośrodkowego układu histaminergicznego wywołuje działanie resuscytacyjne u szczurów we wstrząsie krwotocznym. Skoro podawana dożylnie bombezyna wykazuje podobne działanie, a receptory bombezynowe są obecne w strukturach ośrodkowego układu nerwowego, odpowiedzialnych za regulację czynności układu krążenia, celem pracy było zbadanie wpływu zablokowania receptorów bombezynowych na efekty krążeniowe wynikające z podania do komory bocznej mózgu (icv) histaminy we wstrząsie krwotocznym u szczurów.Materiał i metody:Badania przeprowadzono u szczurów, samców szczepu Wistar w znieczuleniu ogólnym (ketamina 100 mg/kg + ksylazyna 10 mg/kg, dootrzewnowo). Zastosowano model odwracalnego wstrząsu krwotocznego ze średnim ciśnieniem tętniczym (MAP) 30–35 mmHg. Niezwłocznie po zakończeniu krwawienia podawano icv antagonistów receptorów bombezynowych/0,9% roztwór NaCl, a 5 min później tą samą drogą histaminę (50 nmol) bądź 0,9% roztwór NaCl.Wyniki:Utrata krwi prowadziła do obniżenia ciśnienia tętna (PP), częstości rytmu serca (HR) i przepływu krezkowego krwi (MBF). Histamina wywoływała działanie presyjne ze wzrostem PP i MBF. Efekty działania histaminy były częściowo hamowane przez [D-Phe12,Leu14]-bombezynę i BIM 23042, odpowiednio nieselektywnego antagonistę receptorów bombezynowych oraz selektywnego blokera receptorów bombezynowych typu 1 (BB1). W grupach kontrolnych nie stwierdzono wpływu antagonistów receptorów bombezynowych na mierzone parametry układu krążenia.Wnioski:Endogenne peptydy bombezynopodobne, działając poprzez receptory BB1, mogą wpływać na wywoływane przez ośrodkowo działającą histaminę działanie resuscytacyjne we wstrząsie krwotocznym u szczurów.
