Enol Ethers, XV l) Synthesis and Structure of [n](2.4)PhloroglucinophanesEnol ethers of cyclic ketones 1 react with malonyl dichloride (2) in ether by double acylation t o yield [n](2.4)phloroglucinophanes, their tautomers or derivatives (acetales, ethers) resp.; after non aqueous workup the bicyclic compounds 5,6, after workup in water the bicyclic compounds 7 -1 0 resulted. The different product formation can be explained by the known relations of structure and binding for unsaturated bicyclic systems and metacyclophanes, resp. From the benzoic structures of the metacyclophanes 9f and l o g it can be concluded, that even [8] -and [7]metacyclophanes are relatively free of strain. For the cleavage of the methyl aryl ethers 6d and 6 h an addition elimination mechanism could be established with H2"O. Crystal structure determinations were carried out for 7b, d and 10h.In der vorstehenden ArbeitIa' haben wir eine neue Synthese von Phloroglucin bzw. Phloroglucinderivaten, ausgehend von Ketonenolethern durch Umsetzung mit Malonyldichlorid beschrieben. Diese cyclisierende Diacylierung laBt sich auch auf Enolether cyclischer Ketone anwenden, wobei [n](2.4)Phloroglucinophane bzw. deren Tautomere oder Derivate (Acetale, Ethet) entstehen, woriiber in der vorliegenden Arbeit berichtet wird. Aufgrund der Tautomeriemoglichkeiten sowie der Ringspannungen in den bicyclischen Phloroglucinophanen waren interessante Ruckschlusse auf Struktur und Reaktionsverhalten von Metacyclophanen in Abhangigkeit von der RinggroBe zu
Enolether 2 lassen sich rnit aktivierten Saurechloriden wie den Chloressigsaurechloriden 1 a und 1 b, Cyanacetylchlorid (6c) oder Malonylchloriden 6a, b unter milden Bedingungen in guten Ausbeuten acylieren. Die Abhangigkeit der Produktbildung und der Ausbeuten vom Acylierungspotential der eingesetzten Saurechloride wird aufgezeigt. Die erhaltenen potentiellen PKetoaldehyde 4,5 bzw. 1,3,5-Tricarbonylverbindungen 8 sind wertvolle Zwischenprodukte fur die Synthese von Heterocyclen. Enol Ethers, XIII') Acylation of Enol Ethers with Reactive Carbonyl ChloridesEnol ethers 2 are acylated under mild conditions with activated acyl chlorides, e.g. chloroacetyl chlorides la, b, cyanoacetyl chloride (6c), and malonic acid chlorides 6a, b in good yields. The products formed and the yield depend on the acylation potential of the acyl chlorides. The potential p-keto aldehydes 4,5, and 1,3,5-tricarbonyl compounds 8 obtained are valuable intermediates for heterocyclic synthesis.Enamine und Enolether haben als Derivate von Carbonylverbindungen besonders bei der gezielten C -C-Verknupfung groRe Bedeutung in der Synthese erlangt. In der praparativen Anwendung erganzen sich die beiden Verbindungsklassen; die nucleophileren Enamine z. B. sind besonders gut Alkylierungen, Acylierungen und Carboxylierungen zuganglich 3), wahrend die weniger reaktiven Enolether rnit den Acetalen von Aldehyden und Ketonen sowie mit Orthoameisensaureester in guten Ausbeuten unter C -C-Bindungskniipfung reagieren4s5). In der praparativen Anwendung erganzen sich Enolether und Enamine auch insofern vorteilhaft, als sie in Abhangigkeit von den zugrundeliegenden Carbonylverbindungen von sehr unterschiedlicher Stabilitat und Zuganglichkeit sind. Ketone lassen sich meist bequem in relativ stabile Enamine uberfiihren, wahrend Aldehydenarnine wegen ihrer Empfindlichkeit nur schlecht zu handhaben sind6a). Enamine des Acetaldehyds z. B. lassen sich nur unter volligem AusschluR von Feuchtigkeit und Luftsauerstoff als wenig stabile Verbindungen darstellen6b). Umgekehrt verhalt es sich dagegen rnit der Zuganglichkeit und Stabilitat von Enolethern. Aldehydenolether sind leichter zuganglich und auch stabiler als Enolether von Ketonen; Vinylether -die Enolether des Acetaldehyds -sind sogar grofitechnische Produkte.Bekanntlich gelingt die Acylierung freier Aldehyde mit Carbonsaurederivaten wegen der bevorzugten Selbstkondensation der Aldehyde -basisch bzw. sauer katalysierte Aldolreaktion -0
The effects of aralkylation of selected oligonucleotides by a bulky chemical carcinogen, 7,12-dimethylbenz(a)anthracene (after activation) have been studied. The aralkylation involves the base adenine, designated A* at the modification site, in the center of synthetic heptameric, nonameric and pentadecameric oligonucleotides; complementary strands lacking any modification were also synthesized. The products were studied by UV melting curves and CD spectral techniques. Duplex formation was modified by such aralkylation of a central base in the oligomers. The extent of duplex formation was found to depend on chain length as follows: no evidence was found for duplex formation of the heptamer d(GTCA*GAC) + d(GTCTGAC); the nonamer, d(GTGCA*ATCC) + d(GGATTGCAC), appears to form a duplex at high salt concentrations and reduced temperature; the pentadecamer, d(CCGCT-GCGA*TCCGGC) + d(GCCGGATCGCAGCGG), forms a duplex at low salt concentration and room temperature, but its melting temperature is lower than that of the nonalkylated parent system. CD-spectra for the duplexes formed by the nonamer or pentadecamer are indicative of a right-handed helical conformations. On phosphordiesterase digestion it appears that the aralkylated adenine and the base on its 5'-side act as "stops" for enzymatic digestion from either direction. We suggest, from model building, that this inhibition of phosphodiesterase activity is the result of the steric bulk and disposition of the polycyclic aromatic hydrocarbon. We further suggest that unusual base pairing (mismatching), such as A...A, which would lead to an AT transversion, may be favored by the bulkiness of the aromatic group.
(Trimethylsilyl)enolether von Aldehyden 1 und nichtcyclischen bzw. cyclischen Ketonen 4 bzw. 10 lassen sich mit Malonyldichlorid bei Raumtemperatur bzw. bereits bei -60°C acylieren. Hierbei entstehen nach waDrigcr Aufarbeitung aus den 1 -(Trimethylsiloxy)alkenen I die bislang nur schwer zuganglichen 6-unsubstituierten 4-Hydroxy-2H-pyran-2-one 3 und aus den l-(Trimethylsiloxy)cycloalkenen 10 die 5,6-Alkylen-uberbruckten CHydroxy-2H-pyran-2-one 11. Die Acylierung von 2-(Trimethylsiloxy)-l-propen (4a) und 3-(Trimethylsiloxy)-2-penten (4 b) fiihrt zu Gemischen aus 4-(Trimethylsiloxy)-2H-pyran-2-onen 5 (Hauptprodukte) und Phloroglucin-tris(trimethylsily1ethern) 6 (Nebenprodukte). Enol Ethers, XVlI') Acylation of Trimethylsilyl Enol Ethers with Malonyl Dichloride - Synthesis of 4-Hydroxy-2H-pyran-2-onesTrimethylsilyl enol ethers of aldehydes 1, acyclic and cyclic ketones 4 and 10, respectively, are acylated by malonyl dichloride (2) at room temperature or even at -60°C. After aqueous workup, the 6-unsubstituted 4-hydroxy-2H-pyran-2-ones 3 are obtained from 1 -(trimethylsi1oxy)alkenes 1, whereas the 5,6-alkylene-bridged 4-hydroxy-2H-pyran-2-ones I1 are formed from 1-(trimethy1siloxy)cycloalkenes 10. Acylation of 2-(trimethylsiloxy)-l-propene (4a) and 3-(trimethylsiloxy)-2-pentene (4 b) results in mixtures of 4-(trimethylsiloxy)-2H-pyran-2-ones5 (main products) and phloroglucinol tris(trimethylsily1) ethers 6 (by-products).Mit den Enolethern von Aldehyden werden bei der Umsetzung mit Malonyldichlorid ausschlieBlich 3-Oxopentansaurederivate erhalten 3), die sich mit konz. Die Acylierung von Ketonenolethern mit Malonyldichlorid fiihrt dagegen in Abhangigkeit von der Struktur der eingesetzten Enolether und den Reaktionsbedingungen zu Gemischen aus 5-Alkoxy-3-oxo-4-hexensaureestern bzw. den durch Cyclisierung daraus resultierenden 4-Hydroxy-2H-pyran-2-onen und Phloroglucin-m~noalkylethern~).
