It is well-known that Amazon tropical forest soils contain high microbial biodiversity. However, anthropogenic actions of slash and burn, mainly for pasture establishment, induce profound changes in the well-balanced biogeochemical cycles. After a few years the grass yield usually declines, the pasture is abandoned and is transformed into a secondary vegetation called "capoeira" or fallow. The aim of this study was to examine how the clearing of Amazon rainforest for pasture affects: (1) the diversity of the Bacteria domain evaluated by Polymerase Chain Reaction and Denaturing Gradient Gel Electrophoresis (PCR-DGGE), (2) microbial biomass and some soil chemical properties (pH, moisture, P, K, Ca, Mg, Al, H + Al, and BS), and (3) the influence of environmental variables on the genetic structure of bacterial community. In the pasture soil, total carbon (C) was between 30 to 42 % higher than in the fallow, and almost 47 % higher than in the forest soil over a year. The same pattern was observed for N. Microbial biomass in the pasture was about 38 and 26 % higher than at fallow and forest sites, respectively, in the rainy season. DGGE profiling revealed a lower number of bands per area in the dry season, but differences in the structure of bacterial communities among sites were better defined than in the wet season. The bacterial DNA fingerprints in the forest were stronger related to Al content and the Cmic:Ctot and Nmic:Ntot ratios. For pasture and fallow sites, the structure of the Bacteria domain was more associated with pH, sum of bases, moisture, total C and N and the microbial biomass. In general microbial biomass in the soils was influenced by total C and N, which were associated with the Bacteria domain, since the bacterial community is a component and active fraction of the microbial biomass. Results show that the genetic composition of bacterial communities in Amazonian soils changed along the sequence forest-pasture-fallow.

The Amazon rainforest is well known for its rich plant and animal diversity, but its bacterial diversity is virtually unexplored. Due to ongoing and widespread deforestation followed by conversion to agriculture, there is an urgent need to quantify the soil biological diversity within this tropical ecosystem. Given the abundance of the phylum Verrucomicrobia in soils, we targeted this group to examine its response to forest-to-pasture conversion. Both taxonomic and phylogenetic diversities were higher for pasture in comparison to primary and secondary forests. The community composition of Verrucomicrobia in pasture soils was significantly different from those of forests, with a 11.6% increase in the number of sequences belonging to subphylum 3 and a proportional decrease in sequences belonging to the class Spartobacteria. Based on 99% operational taxonomic unit identity, 40% of the sequences have not been detected in previous studies, underscoring the limited knowledge regarding the diversity of microorganisms in tropical ecosystems. The abundance of Verrucomicrobia, measured with quantitative PCR, was strongly correlated with soil C content (r = 0.80, P = 0.0016), indicating their importance in metabolizing plant-derived carbon compounds in soils.

Bacteria and fungi are the most active decomposers of organic materials in soil. They directly affect plant nutrient availability, and chemical and physical properties of soils. This investigation aimed at quantifying the effect of several organic materials on microbial activity of a Rhodic Eutrudox. Soil samples were incubated over a period of 91 days with the following organic materials: cattle manure (CM), earthworm humus (HM), and city sewage sludge from Barueri (BA) and Franca (FR). The activities of cellulase, protease and urease enzymes, the soil microbial carbon content (by fumigation-extraction method) and the exothermal effect were evaluated. Experimental design was randomized and arranged as factorial scheme five treatments × seven samplings with five replications. Organic materials promoted oscillations in microbial carbon (mg C g -1 soil), and enzyme activities -cellulase (µg glucose g -1 soil 24 h ) e no efeito térmico total (J g -1 solo), na seguinte ordem decrescente: (1) 655.90; 49.68; 24.55; 477.90; 367.70; 32.76; 10.66; 426.29; 273.40; 18.77; 19.35; 368.00; (4) 261.00; 17.05; 9.02; 202.79; (5) controle. Correlações foram obtidas entre o carbono microbiano e as enzimas celulase e protease (r = 0.54; p < 0.001), e entre o efeito exotérmico e a protease (r = 0.34; p < 0.005). A atividade de enzimas esteve diretamente relacionada à microbiota do solo, refletindo, dessa forma, no efeito térmico total. SUMMARYEnzymatic activity is an important property for soil quality evaluation. Two sequences of experiments were carried out in order to evaluate the enzymatic activity in a soil (Rhodic Eutrudox) amended with cattle manure, earthworm casts, or sewage sludges from the municipalities of Barueri and Franca. The activity of commercial enzymes was measured by microcalorimetry in the same soil samples after sterilization. In the first experiment, the enzyme activities of cellulase, protease, and urease were determined in the soil samples during a three month period. In the second sequence of experiments, the thermal effect of the commercial enzymes cellulase, protease, and urease on sterilized soil samples under the same tretaments was monitored for a period of 46 days. The experimental design was randomized and arranged as factorial scheme in five treatments x seven samplings with five replications. The treatment effects were statistically evaluated by one-way analysis of variance. Tukey´s test was used to compare means at p ≤ ≤ ≤ ≤ ≤ 0.05. The presence of different sources of organic residues increased the enzymatic activity in the sampling period. Cattle manure induced the highest enzymatic activity, followed by municipal sewage sludge, whereas earthworm casts induced the lowest activity, but differed from control treatment. According to estimates of the intergovernmental panel on climate change, the continued use of oil based-energy is responsible for more than two thirds of the anthropogenic emissions of greenhouse gases in the atmosphere. In Brazil, the National Program for use and biodiesel production has looked for the diversification of feedstock for biodiesel production. Among several alternative sources of energy, microalgae biomass shows great potential to be used as raw material for producing biodiesel. Rich in lipids and fatty acids, the oil yield per hectare in some strains of microalgae is considerably higher than the most conventional oilseed crops such as palm, Jatropha, soybean and sunflower. The commercial production triggered strong interest at the 1960s, with the development of a series of technologies to cultivate microalgae in open ponds and photobioreactors. Industrial or agricultural wastes such as vinasse previously treated in anaerobic digesters, for example, can be recycled and reused through the cultivation of microalgae, besides the application in the fertirrigation of sugar cane crop. This would also qualify the cultivation of microalgae as a clean development mechanism to reduce the levels of greenhouse gases.
