Phenolic compounds, among them isopropyl caffeate, possess antioxidant potential, but not without toxicity and/or adverse effects. The present study aimed to evaluate the antioxidant activity and toxicity of isopropyl caffeate through in silico, in vitro and in vivo testing. The results showed that isopropyl caffeate presents no significant theoretical risk of toxicity, with likely moderate bioactivity: GPCR binding, ion channel modulation, nuclear receptor binding, and enzyme inhibition. Isopropyl caffeate induced hemolysis only at the concentrations of 500 and 1000 μg/ml. We observed types A and O erythrocyte protection from osmotic stress, no oxidation of erythrocytes, and even sequestrator and antioxidant behavior. However, moderate toxicity, according to the classification of GHS, was demonstrated through depressant effects on the central nervous system, though there was no influence on water and food consumption or on weight gain, and it did present possible hepatoprotection. We conclude that the effects induced by isopropyl caffeate are due to its antioxidant activity, capable of preventing production of free radicals and oxidative stress, a promising molecule with pharmacological potential.

ResumoCissus sicyoides L. (cipó-anil, insulina vegetal), pertence à família Vitacea, é muito empregada pela população para o tratamento de diabetes. O objetivo desta pesquisa foi realizar uma triagem fitoquímica do extrato hidroalcóolico das folhas de C. sicyoides para utilizá-lo como fonte primária na realização do ensaio toxicológico com Artemia salina L. Foram realizados testes fitoquímicos do extrato hidroalcoólico das folhas de C. sicyoides para pesquisa dos metabólitos secundários e foi avaliado a atividade toxicológica utilizando cistos de A. salina para quantificar a concentração letal média (CL50). O extrato apresentou esteróides, taninos, flavonóides e saponinas, compostos que já haviam sido encontrados em outros estudos dessa espécie. Apresentou alta toxicidade frente à A. salina, reportando Concentração Letal Média (CL50) de 930,7 g mL -1 , servindo de parâmetro para a realização de estudos posteriores em modelos animais com a pretensão de alcançar mais resultados para um uso seguro da planta da C. sicyoides.Palavras-chave: Artemia salina, ecotoxicologia, etnobotânica, plantas medicinais, fitoquímica. It is much used by the population for the treatment of diabetes. IntroduçãoDesde os primórdios, os recursos naturais, em especial as plantas, têm sido um dos elos entre o homem e a natureza, pois os homens pré-históricos já faziam uso das plantas para amenizar o sofrimento dos males físicos que lhes acometiam (Sharma et al., 2014). O homem primitivo, no meio de tentativas e erros, teve de adquirir conhecimentos e através destes e observar quais as plantas poderiam ser utilizadas como alimento, medicamento e ainda destacar as que fossem venenosas ou perigosas. Com o passar do tempo, o poder curativo das plantas, tornou-se muito importante para ser esquecido e o homem começou a sistematizar o seu uso (Allen, 2016).

The ethanolic extract of the leaves of Cissampelos sympodialis showed great pharmacological potential, with inflammatory and immunomodulatory activities, however, it showed some toxicological effects. Therefore, this study aims to verify the toxicological potential of alkaloids of the genus Cissampelos through in silico methodologies, to develop a method in LC-MS/MS verifying the presence of alkaloids in the infusion and to evaluate the toxicity of the infusion of the leaves of C. sympodialis when inhaled by Swiss mice. Results in silico showed that alkaloid 93 presented high toxicological potential along with the products of its metabolism. LC-MS/MS results showed that the infusion of the leaves of this plant contained the alkaloids warifteine and methylwarifteine. Finally, the in vivo toxicological analysis of the C. sympodialis infusion showed results, both in biochemistry, organ weights and histological analysis, that the infusion of C. sympodialis leaves presents a low toxicity.

Objetivo: Investigar a substância GA-4, quanto a ser um potente candidato a antidepressivo, além de identificar seu possível mecanismo de ação através de docking. Métodos: Foram realizados ensaios psicofarmacológicos in vivo, devidamente autorizado pelo Comitê de ética no uso de animais, utilizando camundongos Swiss, sendo através de testes do campo aberto nas doses de 50, 100 e 200 mg/Kg, analisando os parâmetros: Rearing, Grooming, Número de cruzamentos, micções e bolos fecais. Além do teste do nado forçado, sendo avaliado o tempo de imobilidade dos animais nas mesmas doses do teste de campo aberto. A modelagem molecular foi realizada através de programas computacionais. Resultados: Os resultados mostraram que a substância GA-4 apresenta atividade antidepressiva, tendo em vista que reduziu o tempo de imobilidade no teste do nado forçado e não alterou a atividade locomotora no teste de campo aberto. No estudo de docking, a substância GA-4 teve afinidade com o receptor α2- adrenérgico e se ligou no mesmo sítio de ativação da ioimbina, mostrando ser um possível antagonista dos receptores α2- adrenérgico Conclusão: Diante dos resultados obtidos, podemos concluir que a substância GA-4 é um potencial candidato a medicamento antidepressor. Possivelmente por antagonizar os receptores α2- adrenérgico.
