Bacillus thuringiensis (Bt) é um bacilo Gram positivo produtor de bacteriocinas que tem ação bacteriolítica ou bacteriostática sobre microrganismos patogênicos. As bacteriocinas são peptídeos antimicrobianos que atuam contra bactérias Gram positivas e negativas e diferem entre si quanto a composição de aminoácidos, biossíntese, transporte e modo de ação. Nos alimentos, as bacteriocinas podem ser encontradas naturalmente como produtos da microbiota normal ou introduzida. O objetivo desse projeto foi isolar estirpes de Bacillus thuringiensis a partir de amostras de solos do Distrito Federal e verificar a produção de bacteriocinas com potencial de inibição de microrganismos. Foram isoladas e caracterizadas nove estirpes de Bacillus thuringiensis (Bac, P4, B01, B02, B03, B04, B05, B06 e B07). Cada estirpe foi testada quanto ao potencial de inibição por meio de bioensaios de confronto por estrias e por Well Difusion contra bactérias patogênicas (Serratia marcescens, Escherichia coli e Proteus sp.) e contra as estirpes de Bt isoladas. Os bioensaios por estrias foram realizados como triagem para a seleção das estirpes de Bt produtoras de bacteriocinas. Os ensaios foram mantidos a 28 °C por 24 h a 72 h. Após a seleção das estirpes de Bt com potencial de inibição, realizou-se o bioensaio por Well Difusion para a confirmação da melhor estirpe de Bt produtora de bacteriocinas. Os bioensaios foram mantidos a 28 °C por 24 h seguindo-se a medição dos halos. A confirmação da produção de bacteriocina foi realizada por uma cinética de crescimento durante 48 h, com coletas realizadas em intervalos de tempo determinados. A estirpe de Bt denominada Bac apresentou o melhor potencial de inibição sobre as demais estirpes de Bt e a cinética de produção de bacteriocinas mostrou que esse peptídio é produzido e secretado em momentos específicos do crescimento da estirpe de Bt. Vale ressaltar que essa bacteriocina não inibiu as bactérias patogênicas testadas, indicando a necessidade de ampliar os testes contra outras espécies de bactérias patogênicas. Sendo assim, bacteriocinas produzidas por estirpes de B. thuringiensis podem ser utilizadas na inibição do crescimento de microrganismos, indicando um potencial de uso biotecnológico dessa proteína, visando o desenvolvimento de produtos de interesse biomédico
