Mit Terpenen als H‐Donatoren konnte die Cyangruppe katalytisch (Pd/Kohle) zur Methylgruppe hydriert werden. Bei analogen Hydrierungen von veresterten Mandelsäurenitrilen entstanden Äthylbenzole; hier wurde zusätzlich der Acyloxyrest durch Wasserstoff ersetzt.
Die friiher beschriebenen Hydrierungen von Nitrilen mit gebundenem Wasserstoff und Pd/ Kohle gelingen bei ausreichend hohcr Temperatur auch mit elementarem Wasserstoff. cr-Cyan-zimtsaureester werden in cr-Methyl-hydrozimtsaureester iibergefiihrt, aus denen das pharmakologisch wichtige 2-Amino-I -phenyl-propan und dessen Derivate gut zuganglich sind. Relativ schwer gelingt die Hydrierung von R-CN zu R-CHJ, wenn R ein unsubstituierter, langkettiger aliphatischer Rest ist. Es wird versucht, den Mechanismus der Umsetzungen zu klaren.Mit Al-p-Menthen, a-Phellandren oder Limonen und Pd/Kohle werden Cyanreste in Methylgruppen umgewandelt I ) , wobei der Stickstoff als NH3 austritt. Zusammenfassung Für die Labordauerkultur von Dermatophyten wird ein Keratin‐Agar beschrieben, auf dem die Laborstämme ihre Korrespondenz zu Naturstämmen weniger schnell verlieren. Die Keratin‐Passage führt zu einer Aktivierung der morphologischen Mannigfaltigkeit der Dermatophyten. Der Flagellat Trichomonas vaginalis muß alle 48–72 Stunden in frisches Nährmedium überimpft werden, um ihn am Leben zu halten. Fügt man aber dem Pferdeserum‐Thio‐glycollate‐Medium 2 % Aminoessigsäure hinzu, kann im selben Medium ohne überimpfen der Flagellat 4–8 Wochen lebend gehalten werden. Versagerquote bei 10 %. Eine weitere Kulturmöglichkeit besteht darin, das Pferdeserum durch 12 % Hühnerei‐Eiweiß zu ersetzen. Hierin konnte Trichomonas vaginalis 2–4 Wochen ohne überimpfen lebend gehalten werden. Versagerquote bis zu 25 %. Summary A keratin‐agar for maintenance of dermatophytes is described. Using this medium the laboratory strains do not loose their correspondence to natural strains so quickly. The keratin‐passage causes activation of morphological manifoldness of the dermatphytes. The preparation of a culture medium for Trichomonas vaginalis by addition of 2 % aminoacetic acid to horseserum‐thioglycollate‐medium is described. After inoculation at the bottom of the culture the flagellate can be kept alive for 4–8 weeks at 37° C without subinoculation. The failure rate is about 10 %. The replacement of the horseserum in the medium by 12 % fresh hen's egg albumen gives a further possible culture method for the protozoon. The flagellate could be kept alive for 2–4 weeks in this medium. The failure rate may be up to 25 %. Resumen Para el mantenimiento de los cultivos de dermatofitos en el laboratorio se describe un agar‐creatina sobre el cual estos pierden menos rápidamente su correspondencia con las cepas naturales. El pasaje por creatina lleva a una activación de las múltiples características de los dermatofitos. Para mantener viva a la Tricomona vaginal se la debe reinocular en un medio de cultivo fresco cada 48–72 horas. Si se agrega 2 % de ácido aminoacético a este medio de cultivo de suero de equino y tioglicolate, se la puede mantener viva 4–8 semanas sin reinoculación. Porcentaje de fracasos alrededor del 10 %. Otra posibilidad está dada por el reemplazo del suero de equino por 12% de albúmina de huevo de ave. Así se pudo mantener viva a la Tricomona vaginal 2–4 semanas sin reinoculación. Porcentaje de fracaso alrededor del 25 %.
Zusammenfassung Es wird ein neues Testverfahren zur Prüfung auf Fungizidie beschrieben. Die Pilze werden dabei direkt im Reaktionskolben mit Glaskugeln fein zerschlagen, so daß die zu testende Substanz den Pilz gut erreicht und ein eventueller, störender, konterkarierender Mikroluftfilm eliminiert wird. Dem Reaktionskolben werden in zeitlichen Abständen mittels Platinöse Proben entnommen und mit diesen Proben ohne Waschvorgang Kulturröhrchen mit speziellem, flüssigem Nährmedium inoculiert. Im flüssigen Nährmedium kann nach spätestens 72 Stunden das submers wachsende Mycel beobachtet werden, wenn der Pilz nach der Behandlung noch lebt. Nach einigen Tagen kann dann der Pilz an der Oberfläche des flüssigen Nährmediums wachsen. Summary A new method for testing fungicidal (not fungistatic!) qualities is described. The microfungi are finely broken down by shaking with small glass pearls directly in the reaction flask. This ensures, that the substance which is being tested comes into good contact with the fungi, and that a possible disturbing layer of air is avoided. Samples are taken from the reaction flask at intervals with an platinum‐wire loop. Culture tubes containing special liquid nutrient medium are inoculated with the taken samples. If the fungi are still alive after this treatment, then a submerged growing mycelium can be observed within 3 days in the liquid nutrient medium. After some days the fungi may also grow on the surface of the liquid medium. Resumen Se describe un nuevo método para testificar cualidades fungicidas. Para ello el hongo es destruido mediante la agitación del tubo de ensayo que lo contiene junto con pequeñas bolitas de vidrio. La sustancia a investigar entra así en contacto directo con el hongo y se evitan posibles entorpecedoras microcapas de aire. Del tubo de ensayo se extraen en intérvalos de tiempo con un asa de platino muestras que son inoculadas sin lavado previo en tubos con medio de cultivo líquido especial. En este medio de cultivo líquido se puede ver a las 72 hs. el crecimiento sumergido del micelio si el hongo ha sobrevivido el tratamiento. Pasados unos dias el hongo puede crecer en la superficie del medio de cultivo.
