The use of reference genes is required for relative quantification in gene expression analysis and since the stability of these genes could be variable depending on the experimental design, it has become indispensable to test the reliability of endogenous genes. Therefore, this study evaluated 10 reference candidate genes in two different experimental conditions in order to obtain stable genes to be used as reference in expression studies related to scrotal hernias in pigs. Two independent experiments were performed: one with 30 days-old MS115 pigs and the other with 60 days-old Landrace pigs. The inguinal ring/canal was collected, frozen and further submitted to real-time PCR analysis (qPCR). For the reference genes stability evaluation, four tools were used: GeNorm in the SLqPCR, BestKeeper, NormFinder and Comparative CT. A general ranking was generated using the BruteAggreg function of R environment. In this study, the RPL19 was one of the most reliable endogenous genes for both experiments. The breed/age effects influenced the expression stability of candidate reference genes evaluated in the inguinal ring of pigs. Therefore, this study reinforces the importance of evaluating the stability of several endogenous genes previous their use, since a consensual set of reference genes is not easily obtained. Here, two sets of genes are recommended: RPL19, RPL32 and H3F3A for 30-days MS115 and PPIA and RPL19 for the 60 days-old Landrace pigs. This is the first study using the inguinal ring tissue and the results can be useful as an indicative for other studies working with gene expression in this tissue.
White Striping (WS) has been one of the main issues in poultry production in the last years since it affects meat quality. Studies have been conducted to understand WS and other myopathies in chickens, and some biological pathways have been associated to the prevalence of these conditions, such as extracellular calcium level, oxidative stress, localized hypoxia, possible fiber-type switching, and cellular repairing. Therefore, to understand the genetic mechanisms involved in WS, 15 functional candidate genes were chosen to be analyzed by quantitative PCR (qPCR) in breast muscle of normal and WS-affected chickens. To this, the pectoral major muscle (PMM) of 16 normal and 16 WS-affected broilers were collected at 42 days of age and submitted to qRT-PCR analysis. Out of the 15 genes studied, six were differentially expressed between groups. The CA2, CSRP3, and PLIN1 were upregulated, while CALM2, DNASE1L3, and MYLK2 genes were downregulated in the WS-affected when compared to the normal broilers. These findings highlight that the disruption on muscle and calcium signaling pathways can possibly be triggering WS in chickens. Improving our understanding on the genetic basis involved with this myopathy might contribute for reducing WS in poultry production.
No oeste do estado de Santa Catarina, a agricultura é a atividade econômica predominante, visando à produção de milho, soja e produtos destinados à pecuária. Para manter tais produções, este setor utiliza intensamente insumos químicos, como fertilizantes e agrotóxicos, causando poluição nos ecossistemas. Dentre os resíduos químicos, os agrotóxicos são os mais tóxicos, constituídos por uma grande variedade de compostos, que afetam os organismos animais e vegetais. Este estudo objetivou avaliar a toxicidade do herbicida glifosato na concentração de 65 µg/L e o dobro desta concentração 120 µg/L, nos ovários do peixe Danio rerio. Os peixes permaneceram expostos às concentrações do herbicida glifosato por 15 dias para o cálculo do Índice Gonadossomático (IGS) e massa corporal. Para o teste de micronúcleos os peixes ficaram expostos por 3 dias às diferentes concentrações de glifosato. Não houve diferença estatística significativa na massa corporal dos grupos experimentais avaliados. Houve diferença estatística significativa (p<0,05) na massa dos ovários do grupo controle quando comparado com os grupos expostos a diferentes concentrações de glifosato. Em relação ao IGS houve diferença significativa entre as diferentes concentrações de glifosato e também em relação ao grupo controle. Porém a concentração de 120 µg/L apresentou maior diferença em relação à concentração de 65 µg/L (p<0,05). A frequência de micronúcleos e IGS dos peixes expostos ao glifosato, em ambas as concentrações, foram estatisticamente significativas em relação ao grupo controle. Portanto concluiu-se que o glifosato tem um potencial genotóxico em ambas as concentrações e pode causar danos a reprodução dos peixes. Palavras-chave: Poluição. Micronúcleos. Genotóxico. Os herbicidas estão entre os insumos químicos mais utilizados na agricultura e apresentam ação toxicológica para as plantas. Dentre os herbicidas existentes, o Glifosato e a sua formulação comercial Roundup são os mais utilizados devido à eficácia no controle de ervas daninhas. Entretanto, estudos sugerem que estes herbicidas podem provocar distúrbios á organismos aquáticos, principalmente o Roundup, que além do Glifosato ativo, apresenta na composição componentes surfactantes, que provavelmente aumentaria o seu potencial toxicológico para os seres vivos. Desta forma, o presente estudo teve como objetivo comparar o potencial toxicológico do herbicida Glifosato e do Roundup em peixes Danio rerio. Para isso, exemplares de D.rerio permaneceram em aquários expostos a concentração de 65 ?g/L de cada herbicida diluído em água. Foram realizadas contagens de micronúcleos e calculado o Índice Gonadossomático (IGS) e verificado os estágios de maturação gonadal. Os resultados demonstraram que os peixes expostos aos herbicidas, apresentaram aumentos significativos na frequência de micronúcleos, bem como no IGS e maior ocorrência de ovários com estágios em maturação e maduros. Não foram verificadas diferenças significativas na comparação dos herbicidas, exceto um aumento na frequência de ovários maduros nas fêmeas expostas ao Glifosato. Observou – se que mesmo em pequenas concentrações, supostamente os herbicidas poderiam ter ocasionado efeitos nocivos para os peixes D.rerio, podendo induzir efeitos clastogênicos nos eritrócitos, assim como, distúrbios reprodutivos, aumentando a preocupação sobre os impactos que estes herbicidas podem acarretar sobre o ciclo de vida destes organismos.
