-This study was conducted to investigate the antioxidative activity and cytotoxicity of fermented Allium victorialis extract. The results were as follows; The total polyphenol content of A. victorialis extract was 2.63 mg/g, and that of fermented A. victorialis extract was 1.65 mg/g which decreased a little by fermentation. The total flavonoid content of A. victorialis extract was 57.77 mg/g, and that of fermented A. victorialis extract was 62.27 mg/g, and this could increase a little from fermentation. Electron donating ability of A. victorialis extract was lower than vitamin C(97.71%), but before fermentation it was 82.29% and after fermentation it became 82.40%. Nitrite scavenging ability of A. victorialis extract before and after fermentation showed lower numerical value than that of butylated hydroxytoluene(BHT) at pH 2.5 but that of A. victorialis extract expressed higher than that of BHT. Superoxide dismutase-like activity showed relatively low level, 15%. Nitrite production increased by A. victorialis extract but was inhibited after fermentation. Methyl diamphetamine (MDA) content was inhibited with increased concentration of A. victorialis extract compared with H2O2 treatment but there was not any difference before and after fermentation. Therefore, production of lipid peroxide(LPO) was inhibited by A. victorialis extract. Cell viability of fibroblast cell was tend to slightly decrease with increased concentration of A. victorialis extract, but not different with control.
Objectives : The objective of this research was to investigate the antibacterial and antioxidant activities of bio-fermented Sanguisorbae Radix extract. Methods : The Sanguisorbae Radix extract was fermented by Streptococcus thermophilus, and their products was tested for antibacterial activity against pathogenic microorganisms namely, Bacillus cereus, Bacillus subtilis, Staphylococcus aureus, Escherichia coli, Vibrio parahaemolyticus and Salmonella typhimurium by paper disc diffusion method and the antioxidant activities of extract was evaluated by five different assays as electron donating ability(EDA), superoxide dismutase(SOD)-like activity, polyphenol, flavonoid contents and nitrite scavenging ability. Results : The bio-fermented Sanguisorbae Radix extract was safe from heat. Antibacterial activity of fermented Sanguisorbae Radix extract appeared relatively highly against Bacillus cereus and Staphylococcus aureus and didn't show any difference. EDA in comparison to Vitamin C showed over 90% activity at about the same time of Sanguisorbae Radix extract expressed highly. SOD activity showed 15% in fermentation before and after. The nitrite scavenging ability of Sanguisorbae Radix extract before and after fermentation showed higher numerical value over 70% in pH 2.5 than that of butylated hydroxytoluene(BHT). But SOD activity, EDA and nitrite scavenging ability were not different between the Sanguisorbae Radix extract before and after fermentation. Total polyphenol content expressed over about 20 mg/g, and that of the Sanguisorbae Radix extracts was increased than that of the fermented Sanguisorbae Radix extracts. Conclusions : The results suggest the usefulness of developing functional materials using antioxidant active Sanguisorbae Radix extract was fermented by Salmonella typhimurium with high polyphenol contents and nitrite scavenging ability.
: This experiment was carried out in order to collect the basic data on the standardization of the manufacturing process of Rehmannia glutinosa Libosch. var. purpurea Makino drying. By the drying methods of R. glutinosa, the content of water, inorganic components, reducing sugar, catalpol and benzo[α]pyrene were investigated. The water content was 15.6~17.2% when R. glutinosa was dried by cold-warm air moisture absorption drying method (CAMAD) at 60℃ during 6 days. Among of the inorganic components of R. glutinosa the K content was the most followed by P, Na, Ca and Mg. The reducing sugar content of R. glutinosa by the hot air drying method (HAD) was much more than that by the CAMAD. The catalpol content of R. glutinosa was not different by the drying temperature when it was dried by the CAMAD. The catalpol content of the large size tuber (about 50.0 g/unit) showed a tendency to increase from 60℃ until 70℃ drying temperature, but that of the small size tuber(about 4.0 g/unit) was decreased as being a trend as the drying temperature high when R. glutinosa was dried by the HAD, But the catalpol content R. glutinosa had a tendency to drop significantly at drying temperature above 80℃. The benzo[α]pyrene content was little detected when R. glutinosa was dried by both the SLD and the CAMAD, and the sampling by the HAD indicated within the scope of 5 ㎍/㎏ which was the scope to regulate by Korean food and drug administration. In conclusion, it seemed that an appropriate drying temperature of R. glutinosa by the CAMAD and the HAD was about 60℃ and about 70℃, respectively, when we consider the catalpol content and benzo [α]pyrene detection in the manufacturing process of drying R. glutinosa. 7. 통계분석 분석결과는 3회 반복 측정치의 평균 ± 표준편차로 표시하였 고, 평균값의 P < 0.05 수준에서 Duncan's multiple range test (DMRT)에 의하여 유의성 검정을 하였다.
Objectives : This study was performed to investigate the antioxidant activity of water and ethanol extracts from Sasa borealis leaves, stems and roots. Methods : Sasa borealis leaves, stems and roots extract were prepared using water and ethanol. The total polyphenol and total flavonoid contents were analyzed. 2,2-diphenyl-1-picrylhydrazyl(DPPH) radical scavenging activity, SOD like activity, 2,2'-azino-bis (3-ethylbenzothiazoline-6-sulfonic acid)(ABTS), hydroxyl radical scavenging activity, and Nitrite scavenging activity assays were carried out to determine the antioxidant activities.Results : The antioxidant activities of the Sasa borealis appeared higher in ethanol extract than water extracts.Total polyphenol and total flavonoids contents in ethanol extracts of leaves were 24.6 ㎎/㎖ and 14.3 ㎎/㎖, respectively, which were much higher than those of any other parts. SOD like activity was 70% ethanol extract of the leaves was highest with 15.68%. Electron donating ability was 70% ethanol extract of the leaves had the highest 59.07%. It exhibited high electron donating ability than BHT(45.68%). Nitrite scavenging activity of 70% ethanol extract was higher than the water extract at pH 2.5 and pH 4.2. Nitrite scavenging activity was 70% ethanol extract of the leaves was the highest 75.2%. Hydroxyl radical scavenging activity was 70% ethanol extract of the leaves was highest with 16.16%, showed very low activity than BHT(61.56%).Conclusions : These results suggest that 70% ethanol extracts from Sasa borealis leaves, exhibited higher antioxidant activities than those of root and stem, and can be potentially used as proper natural antioxidants.
