Summary Composite milk samples were collected monthly during one year from 80 Ayrshire cows in order to analyze the influence of lactation stage, parity and daily milk yield on milk antitrypsin, N‐acetyl‐β‐D‐glucosaminidase (NAGase) and plasmin activities. Growth of S. aureus and E. coli was analyzed parallelly in whey. The levels of milk antitrypsin, NAGase and plasmin as well as replication rates of E. coli and S. aureus, were high in early‐lactation and decreased during the first months of lactation. The levels of these mastitis parameters started to increase again at 3–4 months and continued until the end of lactation. The latter increasing tendency became largely eliminated when daily output values were calculated and used rather than concentration values. Milk NAGase and plasmin increased constantly with increasing parity. One can therefore, improve the function of the mastitis parameters by eliminating sources of “non‐mastitic” variation. NAGase and plasmin showed strong association with decreased milk yield and so these parameters seem to be useful when predicting production loss due to subclinical mastitis. Whey was supportive for the growth of E. coli and S. aureus during early and endlactation when the daily milk yield was lowest. Zusammenfassung Milch‐Antitrypsin, NAGase, Plasmin und bakterielle Replikationsrate in Molke Einfluß des Laktationsstadiums, der Anzahl der Geburten (Parität) und des täglichen Milchertrags Von 80 Kühen der Rasse Ayrshire wurden während eines Jahres monatlich Sammelmilchproben gewonnen, um den Einfluß des Laktationsstadiums, der Anzahl der Geburten und des täglichen Milchertrags auf die Aktivitäten des Milch‐Antitrypsins, der N‐acetyl‐β‐D‐Glucosaminase (NAGase) und des Plasmins zu analysieren. Parallel dazu wurde in der Molke das Wachstum von S. aureus und E. coli untersucht. Die Spiegel von Milch‐Antitrypsin, NAGase und Plasmin waren ebenso, wie die Replikationsrate von E. coli und S. aureus in der Frühlaktation hoch und fielen während der ersten Monate der Laktation ab. Nach 3–4 Monaten bis zum Ende der Laktation begannen die Spiegel dieser Mastitisparameter erneut zu steigen. Letztere Tendenz wurde weitgehend beseitigt, sofern tägliche Ausstoßwerte berechnet und Konzentrationswerten vorgezogen wurden. Milch‐NAGase und Plasmin stiegen konstant mit zunehmender Parität an. Deshalb kann man die Funktion der Mastitisparameter durch die Beseitigung der Quellen nicht mastitisbedingter Veränderungen verbessern. NAGase und Plasmin zeigten eine starke Beziehung zu sinkendem Milchertrag und so scheinen diese Parameter zur Voraussage von Produktionseinbußen infolge subklinischer Mastitis nützlich zu sein. Molke war während der Frühlaktation und dem Laktationsende, als der tägliche Milchertrag am niedrigsten war, zum Wachstum von E. coli und S. aureus förderlich.
Summary Milk antitrypsin and NAGase content, CMT scores and bacterial growth in lactoserum were investigated in 423 clinically infected quarters in 268 cows. 26.7% of the affected quarters were infected by S. aureus. 19.4% by streptococci, 17% by coliforms and 3.8% by A. pyogenes. The values of inflammatory parameters in all clinically infected quarters differed significantly (p < 0.001) from those in healthy control quarters. The lactation stage of the cow and type of infecting agent had a significant effect (p < 0.001, p < 0.05) on the values in mastitic quarters at the acute phase of the disease. Antitrypsin, NAGase and CMT levels were highest in the quarters infected by A. pyogenes but the order of other major pathogens varied with the parameter. Cows infected by A. pyogenes or coliforms showed the most severe clinical signs. Of the clinical signs, milk appearance correlated best with milk levels of NAGase and antitrypsin and CMT score in the diseased quarter. There was no clear correlation between systemic signs and values of the parameters studied, milk NAGase values fell with increasing severity of systemic signs. Klinische Aspekte und Entzündungsparameter bei Mastitiden, die durch unterschiedliche Erreger verursacht wurden Milch‐Antitrypsin und NAGase‐Gehalt, CMT‐Werte und bakterielles Wachstum wurden aus Milchseren von 268 Kühen mit insgesamt 423 klinisch infizierten Eutervierteln untersucht. 26,7% der betroffenen Viertel waren mit S. aureus, 19,4% mit Streptokokken, 17% mit koliformen Keimen und 3,8% mit A. pyogenes infiziert. Die Werte der Entzündungsparameter unterschieden sich in allen klinisch erkrankten Fällen deutlich (p<0,001) von denen in gesunden Vierteln. In der akuten Phase der Erkrankung wirkten sich die Laktationsphase der Kuh und die Art des infektiösen Keimes (p < 0,001, p < 0,05) auf die Werte der an Mastitis erkrankten Viertel aus. Der Antitrypsin‐, NAGase‐ und CMT‐Gehalt war in den Vierteln am höchsten, die mit A. pyogenes infiziert waren. Die Zuordnung der Parameter zu den anderen wichtigen Pathogenen variierte jedoch. Kühe die mit A. pyogenes oder koliformen Keimen infiziert waren, zeigten die schwersten klinischen Symptome, wobei Veränderungen der Milch am stärksten mit den Werten von NAGase, Antitrypsin und DMT in den erkrankten Vierteln korrelierte. Keine klare Zuordnung konnte von systemischen Krankheitszeichen und den überprüften Parametern aufgezeigt werden; der NAGase‐Spiegel der Milch fiel mit der Schwere der klinischen Symptome.
Summary The outcome of 423 quarters with acute bovine mastitis treated with eight different therapeutic regimes was evaluated by means of milk antitrypsin and NAGase 3 weeks after treatment. Analysis of variance was used to predict the 3‐week status from information relating to the acute stage of the disease. Neither the parity of the cow nor the location of the quarter had any effect on post‐treatment values. Pre‐treatment and post‐treatment values of the indicators of inflammation usually correlated positively; cases with high levels at the acute stage recovered poorly. In contrast, the correlation in coliform mastitis was found to be negative, and it was suggested that in these cases an intensive inflammatory response is beneficial to the course of the disease. Of the clinical signs, only local signs predicted recovery from mastitis. The details of antibiotic treatment only affected recovery from S. aureus mastitis; longer duration of treatment and sensitivity of the strain to the drug used showed a positive effect. Zusammenfassung Der Heilerfolg von 8 verschíedenen Therapiearten wurde bei, an akuter Mastitis erkrankten, Kühen 3 Wochen nach der Behandlung anhand der Milchparameter Antitrypsin und NAGase überprüft. Eine Varianzanalyse wurde herangezogen, um den 3‐Wochenstatus anhand von Informationen, die von der akuten Phase der Erkrankung gewonnen wurden, vorherzusagen. Weder die Anzahl der Geburten des Tieres noch die Lage des Euterviertels hatten irgendeinen Einfluß auf diese Werte nach der Behandlung. Die Entzündungsparameter vor und nach der Therapie korrelierten gewöhnlich positiv. Fälle mit hohen Werten während der akuten Phase erholten sich schlecht. Dagegen zeigte sich bei der Mastitis durch Escherichia coli eine negative Korrelation, was darauf hindeutet, daß eine starke Entzündungsreaktion sich günstig auf den Krankheitsverlauf auswirkt. In bezug auf die klinischen Anzeichen geben nur lokale Symptome einen Aufschluß über einen Heilungserfolg. Die Details der antibiotischen Therapien bewirkten lediglich bei S. aureus‐Mastitiden eine Heilung, wobei die Dauer der Behandlung und die Empfindlichkeit des Stammes gegenüber den Medikamenten eine positive Auswirkung hatten.
