Diaphorina citri Kuwayama (Hemiptera: Psyllidae) carries Candidatus liberibacter spp., the putative causal agents of Huanglongbing. D. citri reproduces and develops only on the flushing shoots of its rutaceous host plants. Here we examined whether D. citri is attracted to host plant odors and a mixture of synthetic terpenes. Tests conducted in a vertically oriented Y-tube olfactometer showed that both males and females preferentially entered the Y-tube arm containing the odor from the young shoots of Murraya paniculata (L.) Jack and Citrus limon L. Burm. f. cultivar Eureka. Only males exhibited a preference for the odor of C. sinensis L., whereas the odor of C. x paradisi MacFadyen cultivar Rio Red was not attractive to both sexes. The volatiles emitted by young shoots of grapefruit cultivar Rio Red, Meyer lemon (Citrus x limon L. Burm.f.), and M. paniculata were analyzed by gas chromatograph-mass spectrometry. The samples were comprised of monoterpenes, monoterpene esters, and sesquiterpenes. The number of compounds present varied from 2 to 17, whereas the total amount of sample collected over 6 h ranged from 5.6 to 119.8 ng. The quantitatively dominant constituents were (E)-beta-ocimene, linalool, linalyl acetate, and beta-caryophyllene. The attractiveness of a mixture of synthetic terpenes, modeled on the volatiles collected from M. paniculata, was evaluated in screened cages in a no-choice test. At three observation intervals, significantly more individuals were trapped on white targets scented with the mixture than on unscented targets. These results indicate the feasibility of developing D. citri attractants patterned on actual host plant volatiles.
Molecular imaging by mass spectrometry (MS) is emerging as a tool to determine the distribution of proteins, lipids, and metabolites in tissues. The existing imaging methods, however, mostly rely on predefined rectangular grids for sampling that ignore the natural cellular organization of the tissue. Here we demonstrate that laser ablation electrospray ionization (LAESI) MS can be utilized for in situ cell-by-cell imaging of plant tissues. The cell-by-cell molecular image of the metabolite cyanidin, the ion responsible for purple pigmentation in onion (Allium cepa) epidermal cells, correlated well with the color of cells in the tissue. Chemical imaging using single-cells as voxels reflects the spatial distribution of biochemical differences within a tissue without the distortion stemming from sampling multiple cells within the laser focal spot. Microsampling by laser ablation also has the benefit of enabling the analysis of very small cell populations for biochemical heterogeneity. For example, with a ∼30 μm ablation spot we were able to analyze 3-4 achlorophyllous cells within an oil gland on a sour orange (Citrus aurantium) leaf. To explore cell-to-cell variations within and between tissues, multivariate statistical analysis on LAESI-MS data from epidermal cells of an A. cepa bulb and a C. aurantium leaf and from human buccal epithelial cell populations was performed using the method of orthogonal projections to latent structures discriminant analysis (OPLS-DA). The OPLS-DA analysis of mass spectra, containing over 300 peaks each, provided guidance in identifying a small number of metabolites most responsible for the variance between the cell populations. These metabolites can be viewed as promising candidates for biomarkers that, however, require further verification.
Intercropping with flowering herbaceous plants increases parasitoid survivorship, fecundity and retention and pest suppression in agroecosystems. Few studies, however, have examined the compatibility of parasitoid morphology and foraging ability with floral architecture. This study shows that floral architecture influences the selection of floral host resources used to provide nutrients to parasitoids in cropping systems. Parasitoid foraging performance was evaluated using real and artificial flowers which varied in degree of nectar accessibility for two eulophid parasitoids, Edovum puttleri Grissell and Pediobius foveolatus Crawford. Comparisons were made of searching performance on artificial flowers with nectars that were either scented (made from 1:1 honey-water solution) or scentless (made from 1 m sucrose solution) and differences in head widths were compared with corolla apertures. Our results showed a disparity in the ability of E. puttleri and P. foveolatus to gain access to nectar from particular types of floral architectures. E. puttleri fed efficiently only from flowers with exposed nectaries while P. foveolatus foraged efficiently from flowers having either exposed nectaries or nectaries partially obstructed by petals and stamens. Neither wasp species could forage on flowers with cup-or tube-shaped corollas because their heads are wider than the floral apertures. E. puttleri's foraging performance decreased as nectar inaccessibility increased in the artificial flowers, while P. foveolatus' foraging performance was uniform among the different artificial flowers. This indicates that E. puttleri has less propensity to search small openings for nectar than does P. foveolatus. The foraging success of both E. puttleri and P. foveolatus on artificial flowers was lower when 1 M sucrose solution was used as an artificial nectar rather than honey-water solution, indicating that the wasps were stimulated and attracted by the nectar odor. Our systematic evaluation of floral architecture with respect to parasitoid foraging ability has enabled us to predict which types of flowers would serve as suitable floral host plants for parasitoids in the field. That is, only flowers with nectaries that are completely exposed would function as suitable floral host plants for E. puttleri, while P. foveolatus could forage on flowers with either exposed or partially exposed nectaries. Examples of potentially suitable floral hosts suggested from our study include dill (Anethum graveolens L.) and fennel (Foeniculum vulgare L.) for both E. puttleri and P. foveolatus and coriander (Coriandrum sativa L.) for P. foveolatus.
The Asian citrus psyllid, Diaphorina citri (Hemiptera: Psyllidae), is an invasive pest of citrus in the United States. The psyllid feeds and reproduces primarily on new flush growth of citrus and other rutaceous plants. Because it vectors the bacterial causal agents of the deadly citrus greening disease, D. citri is potentially a pest of economic importance in all citrus growing areas where it occurs together with the disease. We investigated the diurnal patterns of its flight activity in the field and the effects of light on its host selection and egg laying behaviors. The numbers of adult psyllids caught on yellow sticky traps were 3 to 4-fold higher during daytime than nighttime. Daytime flight activity of D. citri adults also varied with time of the day with peak catches occurring at midday from 1200 to 1500 h. Illumination of the traps at night increased their attractiveness to adult psyllids by 5-fold. Similarly, light significantly increased plant colonization by adults and female egg deposition on potted plants in the laboratory. These results showed that the flight activity and host selection behavior of adult psyllids are regulated by light and circadian rhythms. Thus, adult psyllids utilize light as visual cues in their host-plant selection process.
Abstract. 1. Predaceous insects may benefit from feeding on non‐prey foods, such as pollen, nectar, and honeydew, because they can provide nutrients that help maintain metabolism and enhance overall nutrient intake. Yet, the extent to which predaceous insects can assimilate non‐prey food and the importance of diet mixing during particular life history stages is poorly understood. In this study the relative contribution of an omnivorous diet to the growth and survivorship of a predaceous larva was tested in a hypothetical situation in which nutritionally optimal prey was not available. The study system comprised a predaceous larva (second‐ and third‐instar larvae of the green lacewing Chrysoperla carnea), nutritionally poor prey (larvae of Drosophila melanogaster), and non‐prey food (pollen suspension, a mixture of bee pollen and artificial nectar (1 M sucrose solution)). Chrysoperla carnea larvae in the mixed diet treatment were provided with both Drosophila larvae and pollen suspension, while those reared on the prey and non‐prey diet treatments received only Drosophila larvae or pollen suspension respectively. 2. The inclusion of pollen and sucrose in their diet enhanced the growth of C. carnea larvae. Second instars reared on the mixed diet developed significantly faster than their cohorts reared on the prey diet, however third instars reared on the mixed diet did not develop faster than their cohorts reared on the prey diet. Larvae reared on the mixed diet became larger adults than did those reared on either the prey or non‐prey diets. Third instars reared on the non‐prey diet completed their development while second instars in the non‐prey diet treatment failed to pupate. 3. Stable isotope analysis indicated that the larvae obtained most of their carbon (55–73%) and nitrogen (71–73%) from Drosophila but acquired only a minor amount of carbon (2–5%) and nitrogen (3–11%) from pollen. Larvae reared on the mixed and non‐prey diets acquired a relatively significant amount of carbon (23–51%) from sucrose. 4. A model, which included a novel fractionation factor to account for the isotopic effect of metamorphosis, was developed to explain the proportion of larval growth attributable to each diet item. It explained the adult δ13C values to within 0.2‰ and adult δ15N values to within 0.7‰ in all treatments. 5. Adults fed 15N‐labelled pollen as larvae retained the 15N signal of the pollen as adults. 6. The collective results of this study support the view that, despite their dependence on prey arthropods to obtain most of their dietary nitrogen, omnivorous lacewing larvae can enhance their growth and development by supplementing their diets with alternative non‐prey food resources. This finding is consistent with the notion that omnivory has evolved as a feeding strategy to acquire both additional nitrogen as well as trace nutrients.
Asian citrus psyllid (Diaphorina citri) transmits the causal agent of Huanglongbing, a devastating disease of citrus trees. In this study we measured behavioral responses of D. citri to combinations of visual, olfactory, and gustatory stimuli in test arenas. Stimuli were presented to the psyllids in droplets or lines of an emulsified wax formulation in two different arena types in no-choice tests. First, when placed on a colored ring situated halfway between the center and perimeter of a petri dish, D. citri spent more time on yellow versus gray rings; however, this response disappeared when either gray or yellow wax droplets were applied. When the psyllids were presented with droplets scented with terpenes, the response to both scent and color was increased. The addition of a dilute (≍0.1 M) sucrose solution to the wax droplets increased the magnitude of D. citri responses. Next, groups of D. citri were placed on plastic laboratory film covering a sucrose solution, to mimic a leaf surface. Test stimuli were presented via two 'midribs' made from lines of emulsified wax formulation. Probing levels were measured as a function of color saturation and scent composition, and concentration. The test scents were based on qualitatively major volatiles emitted by Murraya paniculata (L.) Jack, Citrus aurantifolia (Christm.) Swingle, and C. sinensis (L.) Osbeck. The highest probing response was observed on the middle concentration (20-μl scent/10 ml wax formulation) of the C. aurantifolia-scented wax lines. Results indicate that there are interactive effects between the different sensory modalities in directing host-plant assessment behavior.
Developmental rate and survivorship of small hive beetle, Aethina tumida Murray (Coleoptera: Nitidulidae), life stages were measured across different temperatures (21, 25, 28, 32 and 35 degrees C) and diets, which included natural and artificial pollen, honey, and bee pupae. Temperature affected hatch success, time to hatching, and larval growth. Eggs hatched in 61 h at 21 degrees C but in < 22 h at 35 degrees C. Larvae achieved peak weight in < 8 d at 35 degrees C but needed 17 d at 21 degrees C. Diet had comparatively little effect on larval survivorship or maximum weight, although larvae fed only bee pupae had lower survivorship. Access to soil influenced pupation success. Duration of the life stage spent in the soil, during which pupation occurs, was also affected by temperature: adults emerged after 32.7 d at 21 degrees C but after only 14.8 d at 35 degrees C, albeit with high mortality. Minimum temperature for development was estimated at 13.5 degrees C for eggs, and 10.0 degrees C for larvae and pupae. Temperature influenced adult longevity and oviposition: on a honey and pollen diet average adult lifespan was 92.8 d at 24 degrees C but only 11.6 d at 35 degrees C. Beetles lived longer at 28 degrees C or lower but produced the most eggs per female, regardless of diet, at 32 degrees C. Beetle density influenced fecundity: beetles kept at three pairs per vial laid 6.7 times more eggs per female than those kept as single pairs. Overall, beetles fared best at 28-32 degrees C with mortality of all stages highest at 35 degrees C.
