Die Komplexe vom Typ CoX(D2H2)B (mil X z. B. = Halogen, D = Dimethylglyoxim-Dianion und B = Base) setzen sich mit Grignard-Verbindungen iihnlich wie Cyano-oder Hydroxocorrinoide unter Bildung von coenzym BIZ-artigen Kobaltorganylen der Zusammensetzung CoR(D2HdB urn. Bei der Reaktion mit Alkalimetallen bilden sich Salze vom Typ Me[Co(D2H2)B]e, die mit Alkyl-und Arylhalogeniden die Verbindungen des obengenannten Typs ergeben. Bei der Reduktion entstehen blau-bis blaugrune, dem Vitamin BIZ entsprechende Verbindungen (vermutlich komplexe Kobalthydride). die sich mit Diazomethan, Alkylhalogeniden, Alkenen und Alkinen unter Bildung von Kobaltorganyldimethylglyoxim-Komplexen umsetzen. Die Rontgenanalyse des Cobalamin-Coenzyms 3) ergab als wohl uberraschendstes Resultat das Vorhandensein einer Kobalt-Kohlenstoff bindung im Komplexmolekul. Die Coenzyme der B12-Gruppe stellen somit naturlich vorkommende metallorganische Verbindungen dar ; die Aufklarung ihrer Wirkungsweise ist von noch nicht abzusehender biochemischer Bedeutung4). Die Bildung stabiler Kobalt-Kohlenstoffa-Bindungen in vivo wirft sofort die Frage a d , ob iihnliche Reaktionen nicht auch in vitro an einfachen Modellsubstanzen durchgefuhrt werden konnen. Unsere in dieser Richtung unternommenen Versuche verliefen erfolgreich und werden in dieser und in nachfolgenden Veroffentlichungen mi tgetei 1 t . A. DIMETHYLGLYOXIM-KOBAKOMPLEXE ALS MODELLSUBSTANZEN Im Jahre 1907 beschrieb L. TSCHUGAEFF5) eine Anzahl von Kobalt(II1)-dimethylglyoxim-Komplexen der allgemeinen Zusammensetzung CoX(D2H2)B * *). Die Co -X-Bindungen sind reaktionstriige; mit Age-Ion tritt z. B. mit den Halogenkomplexen des obengenannten Typs erst nach langerer Zeit eine Triibung auf. In diesen Verbindungen liegt das Kobaltatom genau wie in den Komplexen der Vitamin-und Coenzym-Bl2-Gruppe in einem Ligandenfeld aus fiinf Stickstoffatomen. Aus Versuchen mit Vitamin B1h und reduzierten Bis-dimethylglyoximkobalt- Komplexen (,,Cobaloxing") wird gezeigt, daO Vitamin B12r und Cobaloxim, anionische Komplexe des + I-wertigen Kobalts sind, die rnit den korrespondierenden freien Sauren im Hydrolysegleichgewicht stehen : Aus pH-Messungen wird der pK,-Wert der freien Saure fur ein spezielles Cobaloxim, zu 10.54 bestimmt; die Siiuren selbst sind i m freien Zustand unbestiindig und zerfallen unter Wasserstoffentwicklung. Die charakteristische Farbe der Cobaloxime, und des Vitamin B12~ wird im wesentlichen durch Ligandenfeldubergiinge im sichtbaren Spektralbereich verursacht, deren Lage von der Natur der noch an das Kobalt gebundenen Basenkomponente abhiingt. Die hohe nucleophile Reaktivitat des Kobalts in diesen Komplexen und der Mechanismus der Reaktionen mit organischen Substraten wird erkllrt. Bei der Reduktion von Vitamin B12 rnit einer Reihe von Agenzien (z.B. NaBH4, Zn in alkalischem Medium, CrII-Salzen oder rnit H2 in Gegenwart von Katalysatoren) entstehen graugrune Losungen einer als ,,Vitamin Bla" bezeichneten Verbindung, die als Ausgangsprodukt zur Synthese Coenzym BIZ-artiger Verbindungen rnit Co -C-Bindungen und als moglicher Zwischenstoff bei enzymatischen Reaktionen von besondereni Interesse ist2). Vitamin B1% reagiert mit Diazomethan unter Bildung von Methylcobalamin3~4), was nahelegte, d i e m Reduktionsprodukt des Vitamins als Kobalt-Wasserstoffverbindung aufzufassen. Es wurde ursprunglich als Kobalt(III> hydrid formuliert; inzwischen besteht jedoch Einigkeit dariiber, da13 die Verbindung einwertiges Kobalt enthalt .
