Visando a dar suporte a trabalhos de melhoramento genético para porta-enxertos de pera e ajuste de protocolo de germinação de pólen para fins de polinização intra e interespécies, objetivou-se ajustar os componentes básicos do meio de cultura e as condições ambientais para a realização de testes de germinação in vitro e viabilidade de grãos de pólen dos porta-enxertos para pereiras 'Taiwan Nashi-C' (Pyrus calleryana) e 'Taiwan Mamenashi' (P. betulaefolia). O pólen utilizado foi obtido de anteras provenientes de flores em estádio de balão. Em seguida, com auxílio de um pincel n°2, os grãos de pólen foram espalhados sobre a superfície de placas de Petri, contendo 20ml de meio de cultura de acordo com os seguintes experimentos: 1) concentrações de sacarose (0, 30, 60 e 90g L-1) e agar (4, 6, 8 e 10g L-1); 2) concentrações de nitrato de cálcio (0, 200, 400 e 800mg L-1) e ácido bórico (0, 400, 800 e 1200mg L-1); 3) valores de pH (4,0; 4,5; 5,0; 5,5; 6,0 e 6,5); 4) temperaturas (20; 25; 30 e 35); e 5) tempo de emissão do tubo polínico (1, 2, 3, 4, 5, 6 e 12 horas após a inoculação), os quais foram montados de forma sequencial. A utilização de 10g L-1 de agar e 90g L-1 de sacarose para o porta-enxerto 'Taiwan Mamenashi' e 47,78g L-1 para 'Taiwan Nashi-C', com 795 a 838mg L-1 de ácido bórico, na ausência de nitrato de cálcio, pH entre 5,2 e 5,8 e temperatura de 28°C, proporcionou as melhores condições de germinação de grãos de grãos. A porcentagem máxima de polens germinados é obtida com oito horas após a inoculação para 'Taiwan Nashi-C' e com 12 horas para 'Taiwan Mamenashi'.
RESUMOO presente trabalho foi realizado com o objetivo de avaliar a influência do número de pares de folhas e testar o efeito de diferentes concentrações de AIB (ácido indolbutírico) no enraizamento de estacas semilenhosas de oliveira. O delineamento experimental utilizado foi o inteiramente casualizado, em fatorial 4 x 3, sendo quatro concentrações de AIB (0, 1000, 2000, e 3000 mg L -1 ) e três tipos de estacas (sem folhas, com um par de folhas e com dois pares de folhas). As estacas foram padronizadas com 12 cm de comprimento. Após o preparo inicial, as estacas foram imersas nas soluções de AIB por cinco segundos e, em seguida, colocadas em bandejas de polipropileno contendo o substrato Plantmax ® e transportadas para casa-de-vegetação, com umidade e temperatura controladas, onde permaneceram por 90 dias. As variáveis analisadas foram: porcentagem de estacas enraizadas e brotadas, número de folhas, brotos e raízes emitidas por estaca e comprimento médio das brotações e das raízes. O AIB apenas promoveu influência ao sistema radicular; a concentração de 2000 mg L -1 de AIB promoveu maior enraizamento e a concentração de 3000 mg L -1 de AIB em estacas com dois pares de folhas promoveu melhores resultados para número de raízes por estacas e comprimento médio das raízes; estacas ausentes de folhas favoreceram maior brotação nas estacas.Termos para indexação: propagação, estaquia e AIB. ABSTRACTThe present work was developed with the objective to evaluate the influence of the number of leaves and test the effect of different concentrations of IBA (indolbutyric acid) in the cutting propagation of olive tree. The experimental design was complete randomized, in a two factors arrangement (4 x 3), with four different concentrations of IBA (0, 1000, 2000 and 3000 mg L -1 ) and three different types of cuttings (without leaves, one pair of leaves and two pairs of leaves). The cuttings were padronized with 12 cm of length and treated with IBA for five seconds. After that, they were taken to polysty rene trays filled with Plantmax ® and stayed under house green conditions for 90 days. Rooting and sprouting percentage, number of leaves, sprouts and roots, sprout and root length were evaluated. IBA only influenced the root system; the concentration of 2000 mg L -1 promoted best results for rooting percentage and the concentration of 3000 mg L -1 IBA in cuttings with two pairs of leaves promoted best results for number of roots and root length; cuttings of absent leaves favored larger sprouting in the cuttings.
Volume do frasco e consistência do meio de cultura na multiplicação in vitro da bananeira 'Maçã'.Volume do frasco e consistência do meio de cultura na multiplicação in vitro da bananeira 'Maçã' Flask size and growth medium consistence in the in vitro multiplication of 'Maçã' banana tree RESUMO A propagação in vitro da bananeira possibilita produzir, em larga escala, mudas livres de doenças em curto espaço de tempo. Para otimizar o processo de micropropagação, pode-se modificar a metodologia de produção das mudas, levando-se em conta também a manutenção da qualidade. O objetivo deste trabalho foi avaliar a influência do volume do frasco e da consistência do meio de cultura na taxa de multiplicação e no desenvolvimento de mudas de bananeira 'Maçã', propagadas in vitro. A partir de ápices caulinares, foram realizados quatro subcultivos (S2, S3, S4 e S5) em meio MS modificado com metade da concentração dos macronutrientes, acrescido de 30g L -1 de sacarose e 2,5mg L -1 de BAP (6 benzilaminopurina), em tratamento fatorial 2x2: dois tipos de meio de cultivo (semissólido por adição de 1,6g L -1 de Phytagel™ e líquido estacionário) e frascos de dois volumes (250 e 500mL). As plântulas foram induzidas ao enraizamento in vitro e aclimatizadas em estufas, em bandejas de 72 células, e posteriormente cultivadas em sacos de polipropileno de 17x18cm. O cultivo meio líquido em frasco de 500mL pode ser adotado com sucesso para multiplicação de mudas de banana 'Maçã', tendo sido observada maior taxa de multiplicação (25,27 brotos/frasco), melhor adaptação no início da aclimatização e desempenho similar aos demais tratamentos após 60 dias de cultivo em sacos de polipropileno.Palavras-chave: micropropagação, Musa spp., cultura de tecidos, meio líquido. ABSTRACTThe in vitro propagation of the banana is accomplished to produce large quantities of seedlings free from diseases. Micropropagation process can be optimized changing the method, but also considering plantlet quality. The objective of this work was to evaluate the influence of the flask size and of the growth medium consistence on the multiplication rate and on the initial plantlet development of in vitro propagated 'Maçã' banana tree. Starting from stem apexes, four subcultures were accomplished (S2, S3, S4 and S5) in MS medium modified with the half of the concentration of the macronutrients, added of 30g L -1 of sucrose, 2.5mg L -1 of BAP (6-benzylaminopurine) in factorial design 2x2: two types of growth media (semi solid by addition of 1.6g L -1 Phytagel™ and stationary liquid), and two flask sizes (250 and 500mL). Plantlets were induced in vitro rooting and acclimatization in greenhouse using trays with 72 cells and later grown in 17x18cm polypropylene bags. Cultivation in liquid medium bottle of 500mL can be successfully adopted for propagation of 'Maçã' banana plantlets because it had larger multiplication rate (25.27), better adaptation in the initial stage of acclimatization and similar performance to the other treatments after 60 days of cultivation in the bags.
A utilização do umezeiro ou damasqueiro-japonês (Prunus mume Sieb & Zucc.) como porta-enxerto de Prunus sp. vem despertando grande interesse em função de sua rusticidade, resistência a pragas e doenças, adaptação e, principalmente, por reduzir o porte de pessegueiros e nectarineiras. Visto que trabalhos prévios constataram baixo enraizamento de alguns clones de umezeiro e um estímulo a este processo em estacas herbáceas com uso de 2000 mg.L-1 de AIB, objetivou-se no presente trabalho estudar o enraizamento de estacas de pessegueiro e clones selecionados de umezeiros tratados com diferentes concentrações de ácido indolbutírico (AIB) na forma líquida. Estacas lenhosas do pessegueiro 'Okinawa' e dos clones de umezeiro Clone IAC-2, Clone IAC-X, Clone IAC-10 e Clone IAC-XIX, foram padronizadas com 25 cm de comprimento, ausentes de folhas e 5 cm de suas bases tratadas com AIB, nas concentrações 0, 1000, 2000, 3000 e 4000 mg.L-1, por cinco segundos. As estacas foram colocadas em leito de areia umedecido, coberto com sombrite 50% de luminosidade. As avaliações ocorreram após 90 dias após o estaqueamento, avaliou-se a porcentagem de estacas vivas, com calos, enraizadas, brotadas e o número médio de raízes por estacas. Concluiu-se que a concentração de 2000 mg.L-1 de AIB promoveu os melhores resultados para a porcentagem de estacas enraizadas, com calos e número médio de raízes por estacas. De uma forma geral, o Clone IAC-X demonstrou-se superior entre os demais no enraizamento de suas estacas.
LUIZ ANTONIO JUNQUEIRA TEIXEIRA 2 , FERNANDO CÉSAR BACHIEGA ZAMBROSI 3 , JOSÉ EMÍLIO BETTIOL NETO 4 RESUMO -São apresentadas normas DRIS preliminares e derivados níveis críticos de nutrientes no tecido foliar (NC) para bananeiras do subgrupo Cavendish, nas condições de cultivo do Estado de São Paulo. As normas DRIS e os NCs foram obtidos a partir de um banco de dados com 188 registros, relacionando produtividade e teores foliares de N, P, K, Ca, Mg, B, Cu, Fe, Mn e Zn das cultivares Grande Naine e Nanicão. Esses registros foram extraídos de experimentos de adubação realizados no Planalto Paulista e no Vale do Ribeira, em áreas irrigadas e de sequeiro, durante cinco ciclos de cultivo, variando fontes e doses de fertilizantes. Mesmo com a grande diversidade nas condições de cultivo, a relação entre o índice de balanço nutricional e a produtividade das plantas foi altamente significativa (R 2 =0,60; p<0,0001). O método para derivar os NCs a partir das normas DRIS foi eficiente, obtendo-se valores próximos daqueles citados na literatura, com exceção do NC para K, que, para as condições locais, foi inferior. NUTRITIONAL DIAGNOSIS IN BANANA IN THE STATE OF SÃO PAULO (BRAZIL): DRIS NORMS AND CRITICAL LEVELSABSTRACT -Preliminary DRIS norms and leaf nutrient critical levels (NCL) for Cavendish bananas growing in plantations of São Paulo State (Brazil) are presented. DRIS norms and NCL were established from a data bank of leaf nutrient concentration (N, P, K, Ca, Mg, B, Cu, Fe, Mn and Zn) and yield of banana with 188 samples. Data bank comprises information from plants of Grand Naine and Giant Cavendish growing in irrigated and non-irrigated areas, with different sources and rates of fertilizers during five crop cycles. Despite of the great diversity of cropping conditions, the regression between nutrient balance index and fruit yield was significant (R 2 =0.60; p<0.0001). The NCLs were derived using multiple linear regressions relating to the foliar nutrient concentration with DRIS indices of all nutrients. These values were similar to those presented in the literature, except the NCL for K which the value for local conditions was lower.
