Several catalytic properttes of the hydrogenase from S~~n~~~~.~nz~~ obhqtrus have been examined to opttmize the purificatton conditions. The &,-value for HZ-evolution m the presence of the most effective electron medtator methylviologen is 0.66 mM The pH-opttmum is 6.3. the temperatureoptimum is 50°C and the energy of acttvation 1s 38.4 i: 2 kJ,molF'. The soluble hydrogenase from the green alga, Scenedesmus ohliquw, was purtfied 1290-fold to homogeneity. The enzyme consists of two subunits with molecular masses of 55 kDa and 36 kDa. The molecular werght of the native enzyme, determined by gel filtration. is 150 f 5 kDa.
