Ethnopharmacological relevanceIn Austria, like in most Western countries, knowledge about traditional medicinal plants is becoming scarce. Searching the literature concerning Austria's ethnomedicine reveals its scant scientific exploration.Aiming to substantiate the potential of medicinal plants traditionally used in Austria, 63 plant species or genera with claimed anti-inflammatory properties listed in the VOLKSMED database were assessed for their in vitro anti-inflammatory activity.Material and methods71 herbal drugs from 63 plant species or genera were extracted using solvents of varying polarities and subsequently depleted from the bulk constituents, chlorophylls and tannins to avoid possible interferences with the assays. The obtained 257 extracts were assessed for their in vitro anti-inflammatory activity. The expression of the inflammatory mediators E-selectin and interleukin-8 (IL-8), induced by the inflammatory stimuli tumor necrosis factor alpha (TNF-α) and the bacterial product lipopolysaccharide (LPS) was measured in endothelial cells. The potential of the extracts to activate the nuclear factors PPARα and PPARγ and to inhibit TNF-α-induced activation of the nuclear factor-kappa B (NF-κB) in HEK293 cells was determined by luciferase reporter gene assays.ResultsIn total, extracts from 67 of the 71 assessed herbal drugs revealed anti-inflammatory activity in the applied in vitro test systems. Thereby, 30 could downregulate E-selectin or IL-8 gene expression, 28 were strong activators of PPARα or PPARγ (inducing activation of more than 2-fold at a concentration of 10 µg/mL) and 21 evoked a strong inhibition of NF-κB (inhibition of more than 80% at 10 µg/mL).ConclusionOur research supports the efficacy of herbal drugs reported in Austrian folk medicine used for ailments associated with inflammatory processes. Hence, an ethnopharmacological screening approach is a useful tool for the discovery of new drug leads.
Summary• Achillea , a temperate genus of herbaceous allogamous perennials, is a model for evolutionary radiation through hybridization and polyploidization.• AFLP analyses were performed on 300 individuals of 66 populations and 27 taxa/ cytotypes, mainly from the polyploid A. millefolium aggregate and its suspected hybrid links with other clades of the genus.• The mosaic genetic structure of hybrids and polyploids is revealed by specific AFLP bands shared with their assumed parents. In E Asia, A. alpina -4x and A. wilsoniana4x are allotetraploids between A. acuminata -2x (sect. Ptarmica ) and A. asiatica -2x (sect. Achillea-A. millefolium agg.). A. virescens -4x is a hybrid species linking A. nobilis agg. and A. millefolium agg. in S Europe. The hybrid swarm A. clypeolata2x × A. collina -4x recently formed in Bulgaria shows no AFLP bands additive to its parents; by contrast, other more ancient allopolyploids exhibit genetic innovations. Relationships within A. millefolium agg. are complex. Five 2x-taxa, mostly well separated and regressive, are limited to Eurasia; seven 4x-and 6x-taxa are intimately linked by hybridization, are expansive, and through A. asiatica -2x/4x have formed the N American polyploids.• All these results from AFLPs correspond well to other evidence, and indicate a long history of reticulate evolution in Achillea .
The rhizomes of Peucedanum ostruthium (L.) Koch (masterwort) are traditionally used in the alpine region as ingredient of liqueurs and bitters, and as a herbal drug. A sensitive and specific high-performance liquid chromatography-diode-array detection-mass spectrometry (HPLC-DAD-MS) method has been developed for the simultaneous identification and quantification of its main coumarins, oxypeucedanin hydrate, oxypeucedanin, ostruthol, imperatorin, osthole, isoimperatorin, and ostruthin. Fast HPLC separation could be achieved on an Acclaim C18 column (150 mm × 2.1 mm i.d., 3 μm) using a mobile phase gradient of acetonitrile-water modified with 0.01% acetic acid. The quantification by HPLC-DAD was performed with imperatorin as external standard and validated to demonstrate selectivity, linearity, precision, and accuracy. The content of the main coumarins was quantitated in various batches of commercial and field-collected rhizomes of Peucedanum ostruthium, as well as in beverages prepared thereof.
Achillea millefolium L. s.l. is a cytogenetically, morphologically, and chemically polymorphic aggregate. Besides the sesquiterpenes that possess chemotaxonomic relevance and mediate the antiphlogistic activity, the plant contains phenolic compounds such as dicaffeoylquinic acids and flavonoids causing choleretic and spasmolytic effects. To evaluate their contribution to the chemotaxonomy of European taxa of the A. millefolium group, we developed a SPE-HPLC/UV method that allows quantification of the phenolic constituents in the different taxa. The investigated species displayed differences in the quantitative and qualitative composition of phenolic acids and flavonoids. Hence, they seem to be of chemotaxonomic significance, especially for the distinction of the diploid taxa. Combining the obtained results with the data of the sesquiterpene analyses gives a comprehensive insight into the distribution of those pharmacologically relevant plant constituents in the A. millefolium group.
A taxonomic study into the anatomical, morphological and phytochemical differentiation of the genus Mentha L (Lamiaceae) in Bosnia & Hercegovina and Slovakia is presented. Following a population-based approach and using hierarchical cluster analyses the following basic species and hybrids corresponding to exclusive branches, i.e. groups, in the constructed hierarchies were recognized: Mentha aquatica, M. spicata, M. arvensis, M. longifolia, M. rotundifolia, M. × piperita, M. × villosa, M. × verticillata, M. × gentillis, M. × gracilis and M. pulegium. These groups were independently found by separate analyses of the sampled anatomical and morphological variation. In contrast, these anatomically and morphologically defined species exhibited a high level of phytochemical polymorphism which was largely inconsistent with the hierarchical classification. Among the analysed characters, the inflorescence type, dentation of the leaf margin, hair density, the type of capitate glandular hairs, and the main containing compound in essential oil proved most useful for the discrimination of taxa. However, most of the observed traits were not exclusive to a particular species and only the combined consideration of traits revealed coherent taxonomic groups. Basic species and hybrids are described in detail based on the anatomical, morphological and phytochemical characters used for their definition. The performed cluster analysis finally supported hypotheses on the parentage of some of the studied hybrids.
