Die an den Hydroxygruppen acetyl-, pivaloyl-und methylgeschutzten 2-Hydroxy-3-ketoglycale 5a, 5b bzw. 11 werden durch Oxidation der entsprechenden 1-0-Acylglucopyranosen 3a, 3b bzw. 9 an der freien 3-Hydroxygruppe mit Dimethylsulfoxid/Acetanhydrid oder Chlorsulfonylisocyanat hergestellt und charakterisiert. Nur beim Acetylderivat 3a verlaufen die Oxidation und die Eliminierung des 1-Acetyloxysubstituenten in einem Schritt. Beim Pivaloylderivat 3 b und beim Methylderivat 9 mu0 die Eliminierung aus der 3-Ulose 4 bzw. 10 mit Diazabicycloundecen (DBU) vervollstandigt werden. -Die geschutzten Hex-1-enopyran-3-ulosen 5a, 5 b und 11 addieren Brom stereoselektiv unter Bildung der D-ribo-konfigurierten 1,2-Dibromide 13/14. Im Falle der Pivaloyl-und Methylderivate entsteht dariiber hinaus nur das a-Anomere 13. Die Konfiguration der Addukte wird durch NMR-und CD-Spektren ermittelt. Synthesis of 2-Acyloxy-and 2-Alkoxy-3-oxoglycals and of Their Dibromo AdductsThe 2-hydroxy-3-oxoglyca1 derivatives 5a, 5 b, and 11 -acetyl-, pivaloyl-, and methyl-protected, respectively, at their hydroxylic functions -are synthesized by oxidation with dimethyl sulfoxide/ acetic anhydride or chlorosulfonyl isocyanate of the corresponding 1-0-acylglucopyranoses 3a, 3b, and 9, respectively, each containing a free 3-hydroxy group. Only in the case of the acetyl derivative 3a the oxidation is immediately followed by the elimination of the 1-acetyloxy substituent. In contrast, the oxidation of both the pivaloyl derivative 3 b and the methyl derivative 9 yields mainly the corresponding 3-ulose 4 and 10, respectively. Here, the elimination of the 1-acyloxy group must be completed by diazabicycloundecene (DBU). -Bromine is added stereoselectively to the hex-1-enopyran-3-uloses Sa, 5 b, and 11 forming the D-ribo-configurated 1,2-dibromides 13/14. Moreover, in the cases of the pivaloyl and the methyl derivatives only the a anomers 13 are formed. The configurations of the adducts have been estimated by means of NMR and C D spectra.Vor mehr als 20 Jahren beschrieb Theanderz) die Bildung der unsubstituierten 1-Desoxyerythro-hex-1 -enopyran-3-ulose beim basenkatalysierten Abbau von Polysacchariden. Zur Prufung des von ihm postulierten Reaktionsverlaufes unterwarfen Kenne, Larm und S2!ensson3) permethylierte 3-Ulosen dern basenkatalysierten Abbau. Dabei formulierten sie das permethylierte 2-Hydroxy-3-ketoglycal 11 als instabile Zwischenstufe auf dem Weg zu einem komplexen Produktgemisch. Aspinall und Mitarb. 4,5a) konnten, wiederum bei Modellreaktionen zum basischen Abbau von Kohlenhydraten, erstmals zwei 2-acetylgeschutzte Vertreter dieser Substanzklasse isolieren. Allerdings war die von ihnen beschriebene olige Probe der 2,4,6-Tri-O-acetyl-1-desoxy-oerythro-hex-1-enopyran-3-dose (5a) nicht rein. Uber Darstellung und Chemie von Enolen wurde in einem Ubersichtsaufsatz zusammenfassend b e r i~h t e t~~) .
Im Rahmen unserer Untersuchungen uber Komplexe aus paramagnetischen Azomethinliganden und diamagnetischen Schwermetall-Ionen I' haben wir versucht, als 4-zahnige Liganden Bis-benzyliden-2,3-diamino-l,4-naphthohydrochinone aus 2,3-Diamino-1,4-naphthohydrochinon und verschiedenen o-funktionalisierten Benzaldehyden, analog bekannter Bis-benzyIiden-o-phenylendiamine*~", zu synthetisieren.Die Massenspektren der so gewonnenen tiefblauen Produkte sind mit den Zielverbindungen vereinbar. Im Gegensatz dazu stehen die Ergebnisse der anderen spektroskopischen Verfahren. In den IR-Spektren sind intensive Banden zwischen 1620 und 1640 cm-' zu beobachten (vgl. Tab. 5), die C = 0-Schwingungen in p-Chinonen zugeordnet werden konnten. Die 'H-NMR-Spektren zeigen im Erwartungsbereich der Azomethine 6 = 8--9 keine Signale, dafur lassen sich Absorptionen bei 6 = 4 -5 beobachten (vgl. Tab. 1).Die 13C-NMR-Spektren zeigen in Ubereinstimmung rnit I R-Untersuchungen Signale im Carbonylbereich. 'H-und "C-NMR-Spektren sprechen in guter Ubereinstimrnung fur das Vorliegen von sp'-Kohlenstoffatomen.Diese spektroskopischen Befunde legen es nahe, dal3 unter C --C-Knupfung 1,2,3,4-Tetrahydrobenzo[g]chinoxalin-5,lO-chinone 1-7 entstanden sind. Somit ist ein neuer, praparativ einfacher Zugang zu diesen Heterocyclen gefunden4).Bei den von uns mit verschiedenen Benzaldehyden durchgefuhrten Reaktionen wurden stets sterisch einheitliche Produkte in guten Ausbeuten erhalten. Wir nehmen deshalb an, 'H-NMR-Untersuchungen an bekannten 1,2,3,4-Tetrahydro~hinoxalinen~.~) haben gezeigt, daB die Konformeren sich schnell beziiglich der NMR-Zeitskala ineinander umwandeln. Deshalb kann auch bei den vorliegenden Verbindungen erwartet werden, daB fur R' = R (vgl. Schema 2) Mittelwerte gemessen werden und die zwei Protonen des Heterorings magnetisch aquivalent sind. Aus diesem Grunde haben wir versucht, Tetrahydrochinoxaline aufzubauen, bei denen die Protonen an C-2 und C-3 magnetisch nicht aquivalent sind. Derartige Verbindungen entstehen entweder, wenn 2,3-Diamino-1,4-naphthochinon stufenweise mit zwei unterschiedlichen Benzaldehyden umgesetzt wird oder wenn symmetrische Verbindungen (R = R ) nachtraglich an einem Stickstoff substituiert werden. 'H-und '3C-NMR-UntersuchungenDie 'H-NMR-Spektren der Verbindungen 1-7 zeigen charakteristische Multipletts im Aromatenbereich und die envarteten Signale der Substituenten R und R' (vgl. Tab. 1). AuBerdem werden Signale bei 6 = 5 -6 beobachtet, die jeweils 2 Protonen entsprechen. Diese werden bei Zusatz von CD30D nicht mehr registriert, so daB wir diese Signale den NH-Protonen zuordnen. Die aciden Protonen der funktionellen Gruppen in R bzw. R konnen ebenfalls durch Deuterium ausgetauscht werden. Im Bereich 6 = 4 -5 werden in allen Fallen Singuletts rnit der relativen Intensitat 2 beobachtet, die den Protonen in 2-und 3-Stellung des Tetrahydrochinoxalin-Systems zugeordnet werden konnen, so daB die 'H-NMR-Spektren mit der angenommenen Struktur in Einklang sind.Urn sicherzustellen, daO bei unserer Reaktion tatsachlich ein Ri...
202 ChemInform Abstract The reaction between 2,3-diaminonaphthohydroquinone (I) and the aromatic aldehydes (II) does not afford the expected bis-arylidene-2,3-diamino-1,4-naphthohydroquinones, but the symmetrically substituted tetrahydrobenzo(g)quinoxaline-5,10-quinones (III), which result from the intermediary bis-azomethines by a disrotatoric cyclization/intramolecular redox reaction sequence. The structures of the products (III) are in particular deduced from their 1H and 13C NMR spectra. The asymmetrically 2,3-disubstituted quinoxalinequinone (V) is obtained from 2,3-diamino-1,4-naphthoquinone (IV) by a two-step procedure. The N-acetyl derivative (VII) is prepared from (IIIa). The compounds (V) and (VII) with magnetically non-equivalent protons in positions 2 and 3 are prepared in order to compare their spectral data with those of the symmetrically substituted compounds (III). The compounds (III), (V) and (VII) have trans-configuration. This is deduced from their 1H and 13C NMR data and by comparison with their cis-analogues, which are prepared from the trans compounds by oxidation and subsequent catalytic hydrogenation. The compounds (IIIa) and (VII) are additionally characterized by X-ray analyses: (IIIa) has the space group P212121 with Z=4 and (VII) has the space group I4 with Z = 8.
