The triangular correlation heatmap aiming to visualize the linkage disequilibrium (LD) pattern and haplotype block structure of SNPs is ubiquitous component of population-based genetic studies. However, current tools suffered from the problem of time and memory consuming. Here, we developed LDBlockShow, an open source software, for visualizing LD and haplotype blocks from variant call format files. It is time and memory saving. In a test dataset with 100 SNPs from 60 000 subjects, it was at least 10.60 times faster and used only 0.03–13.33% of physical memory as compared with other tools. In addition, it could generate figures that simultaneously display additional statistical context (e.g. association P-values) and genomic region annotations. It can also compress the SVG files with a large number of SNPs and support subgroup analysis. This fast and convenient tool will facilitate the visualization of LD and haplotype blocks for geneticists.
Malus baccata
is one of four wild apple species that can hybridize with the cultivated apple species (
Malus domestica
). It is widely used in high-latitude apple-producing areas as a rootstock and breeding resource because of its disease resistance, and cold tolerance. A lack of a reference genome has limited the application of
M. baccata
for apple breeding. We present a draft reference genome for
M. baccata
. The assembled sequence consisting of 665 Mb, with a scaffold N50 value of 452 kb, included transposable elements (413 Mb) and 46,114 high-quality protein-coding genes. According to a genetic map derived from 390 sibling lines, 72% of the assembly and 85% of the putative genes were anchored to 17 linkage groups. Many of the
M. baccata
genes under positive selection pressure were associated with plant–pathogen interaction pathways. We identified 2,345 Transcription factor-encoding genes in 58 families in the
M. baccata
genome. Genes related to disease defense and cold tolerance were also identified. A total of 462 putative nucleotide-binding site (NBS)-leucine-rich-repeat (LRR) genes, 177 Receptor-like kinase (RLK) and 51 receptor-like proteins (RLP) genes were identified in this genome assembly. The
M. baccata
genome contained 3978 cold-regulated genes, and 50% of these gene promoter containing DREB motif which can be induced by
CBF
gene. We herein present the first
M. baccata
genome assembly, which may be useful for exploring genetic variations in diverse apple germplasm, and for facilitating marker-assisted breeding of new apple cultivars exhibiting resistance to disease and cold stress.
MicroRNAs (miRNAs) are noncoding single‐stranded RNAs, approximately 20‐24 nucleotides in length, known as powerful posttranscriptional regulators. miRNAs play important regulatory roles in cellular processes by changing messenger RNA expression and are widely involved in human diseases, including tumors. It has been reported in the literature that miRNAs have a precise role in cell proliferation, programmed cell death, differentiation, and expression of coding genes. MicroRNA‐124 (miR‐124) has reduced exparession in various human neoplasms and is believed to be related to the occurrence, development, and prognosis of malignant tumors. In our review, we focus on the specific molecular functions of miR‐124 and the downstream gene targets in major cancers, which provide preclinical evidence for the treatment of human cancer. Although some obstacles exist, miR‐124 is still attracting intensive research focus as a promising and effective anticancer weapon.

Monte Carlo simulations are performed to study the adsorption of aromatic molecules (toluene, styrene, o-xylene, m-xylene, p-xylene, 1,3,5-trimethylbenzene, and naphthalene) in all-silica faujasite (FAU) zeolite. For monoaromatics, a two-stage "ideal adsorption" and "insertion adsorption" mechanism is found by careful inspection of locations and distributions of the adsorbed toluene molecules. The validity of this mechanism is confirmed for all monoaromatics considered in the current study. Remarkably, the number of C atoms per unit cell corresponding to the inflection point of adsorbate loading (CI-P) is defined as a valid and convenient characterizing factor in the packing efficiency of monoaromatics in the FAU zeolite. For the case of naphthalene, a type of diaromatic, the three-stage mechanism is proposed, which consists of the first two stages and a third stage of "overideal adsorption". The so-called overideal adsorption is labeled because the naphthalene molecules start to occupy the S site nonideally at loadings that approach saturation, leading to a more localized feature of the adsorbates. The explicit adsorption mechanism can be used to understand the loading dependence of isosteric adsorption heat for the aromatics concerned.
The macrocyclic urea 2, a byproduct in the synthesis of benzoxaborole 1, was identified to be a novel and potent HCV protease inhibitor. We further explored this motif by synthesizing additional urea-based inhibitors and by characterizing them in replicase HCV protease-resistant mutants assay. Several compounds, exemplified by 12, were found to be more potent in HCV replicon assays than leading second generation inhibitors such as danoprevir and TMC-435350. Additionally, following oral administration, inhibitor 12 was found in rat liver in significantly higher concentrations than those reported for both danoprevir and TMC-435350, suggesting that inhibitor 12 has the combination of anti-HCV and pharmacokinetic properties that warrants further development of this series.
