Targeted oncolytic viruses and immunostimulatory therapeutics are being developed as novel cancer treatment platforms. These approaches can be combined through the expression of immunostimulatory cytokines from targeted viruses, including adenoviruses and herpesviruses. Although intratumoral injection of such viruses has been associated with tumor growth inhibition, eradication of distant metastases was not reported. The major limitations for this approach to date have been (1) inefficient intravenous virus delivery to tumors and (2) the lack of predictive, immunocompetent preclinical models. To overcome these hurdles, we developed JX-594, a targeted, thymidine kinase(-) vaccinia virus expressing human GM-CSF (hGM-CSF), for intravenous (i.v.) delivery. We evaluated two immunocompetent liver tumor models: a rabbit model with reproducible, time-dependent metastases to the lungs and a carcinogen-induced rat liver cancer model. Intravenous JX-594 was well tolerated and had highly significant efficacy, including complete responses, against intrahepatic primary tumors in both models. In addition, whereas lung metastases developed in all control rabbits, none of the i.v. JX-594-treated rabbits developed detectable metastases. Tumor-specific virus replication and gene expression, systemically detectable levels of hGM-CSF, and tumor-infiltrating CTLs were also demonstrated. JX-594 holds promise as an i.v.-delivered, targeted virotherapeutic. These two tumor models hold promise for the optimization of this approach.
Current structures across the Tsushima Straits are studied using results from long-term acoustic Doppler current profiler (ADCP) observations by a ferryboat between Hakata and Pusan conducted since February 1997. Two maxima of the northeastward current are observed in the central parts of the eastern and western channels, and the maximum velocity in the western channel is stronger than that of the eastern channel. Downstream of the Tsushima Islands, a southwestward countercurrent is observed associated with a pair of cyclonic and anticyclonic eddies. In the western channel, the deep countercurrent is observed pronouncedly on the bottom slope of the Korean side from summer to winter. The volume transport of the Tsushima Warm Current through the straits has strong seasonal variation with a minimum in January and two maxima from spring to autumn (double peaks). The spring peak of the volume transport through the eastern channel is more pronounced than the autumn peak, and the autumn peak of the western channel is more pronounced than the spring peak. The inflow volume transport into the Japan Sea through the western channel significantly increases in autumn because of an incrementation of the freshwater transport. The total volume transport averaged over the observation period (5.5 yr) is 2.64 Sv (Sv ϵ 10 6 m 3 s Ϫ1 ). The average volume transports through the eastern and western channels are 1.10 and 1.54 Sv, respectively.
The aim of the present study was to assess the effects of skipping breakfast on diet quality and metabolic disease risk factors in healthy Korean adults. Subjects included 415 employees (118 men, 297 women; 30-50 years old) of Jaesang Hospital in Korea and their acquaintances. Data collected from each subject included anthropometric measurements, 3-day dietary intake, blood pressure, and blood analyses. The subjects were classified into three groups based on the number of days they skipped breakfast: 'Regular breakfast eater', 'Often breakfast eater', or 'Rare breakfast eater'. Participants in the 'Rare breakfast eater' group consumed less rice, potatoes, kimchi, vegetables, fish and shellfish, milk and dairy products, and sweets than did participants in the other two groups (P for trend < 0.05) and ate more cookies, cakes, and meat for dinner (P for trend < 0.05). Participants in the 'Rare breakfast eater' group consumed less daily energy, fat, dietary fiber, calcium, and potassium than did participants in the other groups (P for trend < 0.05). The percent energy from carbohydrates was lower and fat intake was higher in the 'Rare breakfast eater' group than in the other groups (P for trend < 0.01). When diets were compared using the Acceptable Macronutrient Distribution Range for Koreans, 59.1% of subjects in the 'Rare breakfast eater' group consumed more energy from fat compared with the other two groups (P < 0.005). According to the Estimated Average Requirements for Koreans, intake of selected nutrients was lower in the 'Rare breakfast eater' group than in the other two groups (P < 0.05). The risk of elevated serum triglycerides was decreased in the 'Rare breakfast eater' group (OR, 0.3 [0.1-1.0], P for trend = 0.0232). We conclude that eating breakfast regularly enhances diet quality, but may increase the risk of elevated serum triglycerides.
There is an established link between food promotions and children's food purchase and consumption. Children in developing countries may be more vulnerable to food promotions given the relative novelty of advertising in these markets. This study aimed to determine the scope of television food advertising to children across the Asia-Pacific to inform policies to restrict this marketing. Six sites were sampled, including from China, Indonesia, Malaysia and South Korea. At each site, 192 h of television were recorded (4 days, 16 h/day, three channels) from May to October 2012. Advertised foods were categorized as core/healthy, non-core/unhealthy or miscellaneous, and by product type. Twenty-seven percent of advertisements were for food/beverages, and the most frequently advertised product was sugar-sweetened drinks. Rates of non-core food advertising were highest during viewing times most popular with children, when between 3 (South Korea) and 15 (Indonesia) non-core food advertisements were broadcast each hour. Children in the Asia-Pacific are exposed to high volumes of unhealthy food/beverage television advertising. Different policy arrangements for food advertising are likely to contribute to regional variations in advertising patterns. Cities with the lowest advertising rates can be identified as exemplars of good policy practice.
On the basis of a new East Asian winter monsoon (EAWM) index and by analyzing the relationship between sea surface temperature (SST) anomalies and different atmospheric and oceanic factors in winter, this study investigates the causes of the recent unusual warming in the western North Pacific Ocean. Analyses presented here emphasize the dual contribution from the atmosphere and ocean to the local SST variability, with the relative importance of each contributor varying with the period and place. During the period 1970–89, the EAWM, controlled mostly by the Siberian high, is predominantly responsible for the SST variability in most of the western North Pacific, whereas in the period 1990–2005 ocean dynamics become increasingly important in most places or even dominant in the Kuroshio–Oyasio Extension (KOE) region. The delayed response of the KOE SST to basinwide wind stress curl forcing via Rossby waves is epoch dependent and is significant at lags of 1, 3, and 4 yr before 1990 but only at 1 yr afterward. This epoch dependency of the impact of Rossby waves is related to the different locations of the centers of action of wind stress curl in the midlatitude North Pacific between the two epochs. In addition, mean advection of the EAWM-driven anomalous SST from the southern East China Sea, which can be transported into the KOE region in about a year by the Kuroshio, likely affects the KOE SST lagged by 1 yr. The strongest positive SST trend observed in the western North Pacific results from the combined effects of the abrupt weakening of the EAWM due to the unprecedented decline of the Siberian high and the increasing role of the ocean. The latter is best evidenced by the 1-yr delayed response of the western North Pacific via the gyre circulation adjustment to the basinwide decadal-scale wind stress curl change associated with the northward shift of the strengthened Aleutian low.
