Our results show better post-DC improvements in the CBF of patients receiving CP < 12 weeks after DC, compared to those receiving CP ≥ 12 weeks after DC. Therefore, early CP has potential benefits for cerebral perfusion.
Background and objectiveCognitive impairments have been reported in patients with hyperprolactinemia; however, there is a lack of knowledge of brain structure alterations relevant to hyperprolactinemia in prolactinomas. Thus, we aimed to identify changes in brain structure in prolactinomas and to determine whether these changes are related to cognitive performance and clinical characteristics.MethodsParticipants were 32 female patients with prolactinomas and 26 healthy controls (HC) matched for age, sex, education, and handedness. All participants underwent magnetic resonance imaging brain scans, neuropsychological assessments, and clinical evaluations. Voxel-based morphometry analysis was used to identify changes in gray matter volume (GMV). Partial correlation analysis and multiple linear regression were performed to determine the relationship between GMV, cognition, and clinical characteristics.ResultsCompared to HC, patients with prolactinomas demonstrated a decrease in GMV in the left hippocampus, left orbitofrontal cortex, right middle frontal cortex (MFC), and right inferior frontal cortex (IFC). In addition, patients performed worse than controls on tests for verbal memory and executive function, and this was significantly related to the GMV of the left hippocampus and right MFC, respectively. Moreover, in the patients, we found a negative relationship between serum prolactin levels and the GMV of the left hippocampus and right IFC, whereas a positive relationship was found between the GMV of the left hippocampus and serum levels of estradiol and luteinizing hormone.ConclusionIn patients with prolactinomas, specific brain structure abnormalities have been identified and are associated with cognitive impairments and dysfunctional hormones. This study enhances our understanding of brain structure changes that may occur with prolactinomas and provides novel and fundamental evidence for previous behavioral findings relevant to hyperprolactinemia.
It has been shown that emotionally positive facial expressions are recognized substantially faster than emotionally negative facial expressions, the positive classification advantage (PCA). In this experiment we explored the involvement of configural computations while processing positive and negative faces in an expression categorization task using artificial faces. Analyzing the reaction times (RTs), we found that happy faces were categorized more quickly than sad faces (PCA) and this effect disappeared for inverted faces. Event-related potentials (ERPs) data showed that the face-sensitive N170 component was larger for sad than for happy faces only at upright condition and that face inversion significantly enhanced N170 amplitudes only for happy faces. Moreover, the happy faces elicited shorter N170 latency than did the sad faces, whereas for inverted condition the N170 latency did not differ between happy and sad faces. Finally, the significant positive correlation between the RTs and the latency of the N170 was not found for N170 amplitudes. Because the configural computation was task-irrelevant in the present study, these behavioral and ERP data indicated that one of the sources of PCA is the configural analysis applied by default while categorizing facial emotions.
OBJECTIVES: To explore the feasibility of achieving diagnostic images in low-dose abdominal CT using a Deep Learning Image Reconstruction (DLIR) algorithm. METHODS: Prospectively enrolled 47 patients requiring contrast-enhanced abdominal CT scans. The late-arterial phase scan was added and acquired using lower-dose mode (tube current range, 175–545 mA; 80 kVp for patients with BMI ≤24 kg/m2 and 100 kVp for patients with BMI > 24 kg/m2) and reconstructed with DLIR at medium setting (DLIR-M) and high setting (DLIR-H), ASIR-V at 0% (FBP), 40% and 80% strength. Both the quantitative measurement and qualitative analysis of the five types of reconstruction methods were compared. In addition, radiation dose and image quality between the early-arterial phase ASIR-V images using standard-dose and the late-arterial phase DLIR images using low-dose were compared. RESULTS: For the late-arterial phase, all five reconstructions had similar CT value (P > 0.05). DLIR-H, DLIR-M and ASIR-V80% images significantly reduced the image noise and improved the image contrast noise ratio, compared with the standard ASIR-V40% images (P < 0.05). ASIR-V80% images had undesirable image characteristics with obvious “waxy” artifacts, while DLIR-H images maintained high spatial resolution and had the highest subjective image quality. Compared with the early-arterial scans, the late-arterial phase scans significantly reduced the radiation dose (P < 0.05), while the DLIR-H images exhibited lower image noise and good display of the specific image details of lesions. CONCLUSIONS: DLIR algorithm improves image quality under low-dose scan condition and may be used to reduce the radiation dose without adversely affecting the image quality.
The function of TRPV1 (transient receptor potential vanilloid subfamily, member 1) in the central nervous system is gradually elucidated. It has been recently proved to be expressed in nucleus accumbens (NAc), a region playing an essential role in mediating opioid craving and taking behaviors. Based on the general role of TRPV1 antagonist in blocking neural over-excitability by both pre- and post-synaptic mechanisms, TRPV1 antagonist capsazepine (CPZ) was tested for its ability to prohibit persistent opioid craving in rats. In the present study, we assessed the expression of TRPV1 in nucleus accumbens and investigated the effect of CPZ in bilateral nucleus accumbens on persistent morphine conditioned place preference (mCPP) in rats. We also evaluated the side-effect of CPZ on activity by comparing cross-beam times between groups. We found that morphine conditioned place preference increased the TRPV1 expression and CPZ attenuated morphine conditioned place preference in a dose-dependent and target–specific manner after both short- and long-term spontaneous withdrawal, reflected by the reduction of the increased time in morphine-paired side. CPZ (10 nM) could induce prolonged and stable inhibition of morphine conditioned place preference expression. More importantly, CPZ did not cause dysfunction of activity in the subjects tested, which indicates the inhibitory effect was not obtained at the sacrifice of regular movement. Collectively, these results indicated that injection of TRPV1 antagonist in nucleus accumbens is capable of attenuating persistent morphine conditioned place preference without affecting normal activity. Thus, TRPV1 antagonist is one of the promising therapeutic drugs for the treatment of opioid addiction.
