The combination of dimethyl disulfide (DMDS) and dazomet (DZ) is a potential alternative to methyl bromide (MB) for soil disinfestation. The efficacy of DMDS plus DZ in controlling key soilborne pests was evaluated in a laboratory study and in two commercial cucumber greenhouses. Laboratory studies found that all of the combinations had positive synergistic effects on root-knot nematodes, two key soilborne fungi, and two major weed seeds. Greenhouse trials revealed that the combination of DMDS and DZ (30 + 25 g m(-2)) successfully suppressed Meloidogyne spp. root galling, sharply reduced the colony-forming units of Fusarium spp. and Phytophthora spp. on media, maintained high cucumber yields, and was not significantly different from MB or DMDS alone, but better than DZ alone. All of the chemical treatments provided significantly better results than the nontreated control. The results indicate that the combination of DMDS and DZ is an efficient MB alternative for cucumber production.

This study confirms that the newly developed real-time PCR quantitative assay for F. oxysporum was suitable for the control efficacy evaluation of soil fumigants and that the novel fumigant combination of CP and DMDS offers a promising effective alternative to MB for the control of F. oxysporum in strawberry greenhouses.

Cattle-yak milk is an important raw material and an indispensable source of high-quality food for local farmers and herdsmen to produce ghee, milk residue, yogurt, and other dairy products. In this study, Lactobacillus strains were isolated from cattle-yak milk for potential probiotic candidates using a series of in vitro tests, including probiotic characterization and safety evaluation (antibiotic susceptibility and hemolytic ability). The results found that the Lactobacillus rhamnosus CY12 strain showed a high survival rate in bile salts, under acid conditions, and in the gastrointestinal juice environment, as well as showing high antimicrobial activity and adhesive potential. The safety evaluation showed that all strains were considered non-hemolytic. In addition, the whole-genome sequencing indicated that the strain CY12 spanned 2,506,167 bp, with an average length of 881 bp; the GC content in the gene region (%) was 47.35, contained 1347 protein-coding sequences, and accounted for 85.72% of the genome. The genome annotation showed that genes mainly focused on the immune system process, metabolic process, carbohydrate utilization, carbon metabolism, galactose metabolism, and biological adhesion, etc. This study revealed that the Lactobacillus rhamnosus CY12 strain might be an excellent potential probiotic in the development of feed additives for animals and has the ability to promote health.

The intestinal barrier is vital for preventing inflammatory bowel disease (IBD). The objectives of this study were to assess whether the Lactobacillus rhamnosus CY12 could alleviate oxidative stress, inflammation, and the disruption of tight junction (TJ) barrier functions induced by lipopolysaccharide (LPS), and therefore to explore the potential underlying molecular mechanisms. Our results showed that LPS-induced Cancer coli-2 (Caco-2) cells significantly increased the levels of reactive oxygen species (ROS), lactate dehydrogenase, inflammatory cytokines interleukin-1β, interleukin-6, interleukin-8, and tumor necrosis factor-α (IL-1β, IL-6, IL-8, and TNF-α), and the cell apoptosis rate while decreasing the levels of TJ proteins occludin, zonula occludens-1 (ZO-1), and claudin and antioxidant enzymes, such as catalase, superoxide dismutase, and glutathione peroxidase(CAT, SOD, and GSH-Px) (p < 0.05). However, Lactobacillus rhamnosus CY12 could relieve cytotoxicity, apoptosis, oxidative stress, and pro-inflammatory cytokine expressions, and also inhibit the Toll-like receptor 4/ nuclear factor kappa-B(TLR4/NF-κB) signaling pathway. Furthermore, the gene expression of antioxidant enzymes, as well as the mRNA and protein expressions of TJ proteins, was improved. Particularly, the concentration of 108 cfu/mL significantly prevented the inflammatory injury induced by LPS in Caco-2 cells (p < 0.05). These findings support a potential application of Lactobacillus rhamnosus CY12 as a probiotic to prevent LPS-induced intestinal injury and treat intestinal barrier dysfunction.

BackgroundChloropicrin (Pic) offers a potential alternative to methyl bromide (MB) against Ralstonia solanacarum in ginger (Zingiber officinale Rosc.) production. MB is scheduled to be withdrawn from routine use by 2015 in developing countries.MethodsPic treatments were evaluated in a laboratory study and in three commercial ginger fields.ResultsLaboratory studies showed that the EC50 value and EC80 value of Pic were 2.7 and 3.7 mg a.i. kg−1 soil, respectively. Field trials in highly infested soil revealed that treatments of Pic at the dose of 50 g m−2 covered with totally impermeable film (TIF) or polyethylene film (PE) sharply reduced Ralstonia solanacarum and maintained high ginger yields. Both of the Pic treatments provided results similar to, or in some cases slightly lower than, MB with respect to Ralstonia solanacarum control, plant survival, plant growth and yield. All of the fumigant treatments were significantly better than the non-treated control.ConclusionsThe present study confirms that the Pic is a promising alternative with good efficacy against Ralstonia solanacarum for ginger production and could be used in integrated pest management programs in China.

The combination of 1,3-dichloropropene + dimethyl disulfide (1,3-D + DMDS), which forms a pre-plant soil fumigant, can provide a substitute for the environmentally unfriendly methyl bromide (MB). Three greenhouse trials were performed to evaluate the root-knot nematode and soilborne fungi control efficacy in the suburbs of Beijing of China in 2010-2014. Randomized blocks with three replicates were designed in each trial. The combination 1,3-D + DMDS (10 + 30 g m −2) significantly controlled Meloidogyne incognita, effectively suppressed the infestation of Fusarium oxysporum and Phytophthora spp., and successfully provided high commercial fruit yields (equal to MB but higher than 1,3-D or DMDS). The fumigant soil treatments were significantly better than the untreated controls. These results indicate that 1,3-D + DMDS soil treatments can be applied by soil injection or chemigation as a promising MB alternative against soilborne pests in cucumber in China.

Animals have adapted behavioral and physiological strategies to conserve energy during periods of adverse conditions. Hepatic glucose is one such adaptation used by grazing animals. While large vertebrates have been shown to have feed utilization and deposition of nutrients—fluctuations in metabolic rate—little is known about the regulating mechanism that controls hepatic metabolism in yaks under grazing conditions in the cold season. Hence, the objective of this research was to integrate transcriptomic and metabolomic data to better understand how the hepatic responds to chronic nutrient stress. Our analyses indicated that the blood parameters related to energy metabolism (glucose, total cholesterol, low-density lipoprotein cholesterol, high-density lipoprotein cholesterol, lipoprotein lipase, insulin, and insulin-like growth factor 1) were significantly (p &lt; 0.05) lower in the cold season. The RNA-Seq results showed that malnutrition inhibited lipid synthesis (particularly fatty acid, cholesterol, and steroid synthesis), fatty acid oxidation, and lipid catabolism and promoted gluconeogenesis by inhibiting the peroxisome proliferator-activated receptor (PPAR) and PI3K-Akt signaling pathways. For metabolite profiles, 359 metabolites were significantly altered in two groups. Interestingly, the cold season group remarkably decreased glutathione and phosphatidylcholine (18:2 (2E, 4E)/0:0). Moreover, integrative analysis of the transcriptome and metabolome demonstrated that glycolysis or gluconeogenesis, PPAR signaling pathway, fatty acid biosynthesis, steroid biosynthesis, and glutathione metabolism play an important role in the potential relationship between differential expression genes and metabolites. The reduced lipid synthesis, fatty acid oxidation, and fat catabolism facilitated gluconeogenesis by inhibiting the PPAR and PI3K-Akt signaling pathways to maintain the energy homeostasis of the whole body in the yak, thereby coping with the shortage of forages and adapting to the extreme environment of the Qinghai-Tibetan Plateau (QTP).
