The presence, in the cell membrane, of high-conductance K+ channels and voltage-gated Ca2+ channels (CaV) forming complexes has been reported. These complexes have important functions in excitable cells. The [Ca2+]i at the mouth of the CaV channel decreases with distance and with the concentration of chelators. For the BK channel to be activated with internal Ca2, a concentration of the order of M is necessary and this implies a closeness between the BK-CaV channels. A simulator of the decay of Ca2+ in the presence of BAPTA to estimate the distance between the channels was developed. The mathematical models were implemented in Visual Basic® 6.0 and were solved numerically. The results indicate the coexistence of L-type CaV channel and BK grouped in nanodomains with a distance between channels of ~30 nm.
Las prácticas de laboratorio virtuales adquieren relevancia en tiempos de pandemia. En este estudio se presenta un simulador de los canales de K+ de alta conductancia (BK) con fines de investigación y de enseñanza-aprendizaje. Estos canales están relacionados con epilepsia e hipertensión arterial. El simulador está basado en el modelo matemático de Gupta y Manchanda, y se implementó en Visual Basic 6.0 para ambiente Windows®. El simulador reproduce los datos experimentales publicados de la cinética de la corriente macroscópica BK de canales transfectados en diferentes células. Los datos simulados y los experimentales no presentaron diferencias (p > 0.05). Con el simulador se pueden realizar experimentos virtuales; determinar parámetros como la sensibilidad al Ca2+ (ΔV1/2), la dependencia de Ca2+ interno y de voltaje; y reproducir la relación conductancia-voltaje (G-V). El simulador es ejecutable en cualquier computadora con recursos mínimos y puede ser utilizado como herramienta didáctica en cursos presenciales y a distancia.
RESUMENEl análisis y comprensión de los procesos fisiológicos de los seres vivos, requiere del desarrollo e interacción académica multidisciplinaria en investigación científica básica, debido a la complejidad y la diversidad biológica de los individuos. Como ejemplo, en este caso se abordan los mecanismos cronobiológicos, cíclicos y circadianos, a nivel molecular. Estos mecanismos, generalmente están basados en un sistema de oscilación periódica diaria (~24 horas), adaptados a las fases día-noche, regulados exógena, endógena y diferencialmente por los estímulos de luz-oscuridad. Algunas de las funciones biológicas, como el reposo, sueño, actividad, locomoción, metabolismo, apareamiento, desarrollo y envejecimiento, son reguladas por estos sistemas de relojes internos, que operan mediante cambios conformacionales protéicos postraduccionales. El primer mecanismo descrito de autorregulación rítmica gen-proteína fue el modelo PER (period), caracterizado a detalle en el insecto Drosophila melanogaster (mosca de la fruta). En este trabajo se presenta el diseño y desarrollo de un simulador interactivo computacional, que reproduce el ritmo circadiano PER. El simulador llamado "OSCILAR-PER," está basado en el modelo propuesto por Goldbeter (1995), fue desarrollado en lenguaje Visual Basic®, versión 5.0, para ambiente Windows®, de XP a Windows 10. El colesterol es una molécula lipídica que participa en la permeabilidad de la membrana celular, en procesos de señalización transmembranal y en la producción de hormonas esteroideas. El hígado y el intestino regulan la concentración sanguínea de colesterol (colesterolemia) a través de la síntesis, consumo de alimentos, almacenamiento y excreción. Niveles altos de colesterol sanguíneo (hipercolesterolemia) causan complicaciones fisiopatológicas que implican costos biológicos y económicos. Se realizó un programa de cómputo formado por tres módulos: (1) leccionario, (2) modelos matemáticos y (3) módulo de simuladores: simulador hepato-intestinal y simulador de regulación génica de la síntesis de colesterol en el hepatocito. El programa fue compilado en lenguaje Visual Basic® 6.0 para ambiente Windows®. Los simuladores pueden reproducir el control de la concentración de colesterol en condiciones normales y patológicas.
Las células del nodo sinusal generan la principal actividad eléctrica marcapaso del corazón. La actividad marcapaso se debe a la presencia de la corriente funny (If). Esta corriente iónica es entrante y se activa con la hiperpolarización en rangos de voltaje presentes durante la fase de despolarización diastólica; contrario a la mayoría de las corrientes iónicas que se activan con la despolarización. El canal funny es permeable a iones Na+ y K+. Generalmente, en el curso de biofísica a nivel de licenciatura los alumnos conocen algunos canales dependientes de voltaje del tipo de Hodgkin y Huxley que se activan con la despolarización. Sin embargo, no se estudia ningún canal que se active con una hiperpolarización. En este trabajo se diseñó y desarrolló un simulador para el estudio y comprensión de la corriente funny presente en el nodo sinusal del conejo. El simulador fue programado en lenguaje Visual Basic ver. 6.0 para ambiente Windows® de XP a Windows® 10. El usuario puede realizar los experimentos con la técnica de fijación de voltaje, modificar las variables y concentraciones de Na+ y K+ externos y observar el efecto en la amplitud de If y la cinética de la curva I-V. Se recomienda su uso como material didáctico de apoyo durante los cursos de biofísica y fisiología en una sala de cómputo o a distancia en cualquier computadora personal con recursos mínimos. The sinus node cells generate the main electric pacemaker activity of the heart. The pacemaker activity is due to the presence of the current funny (If). This ionic current is incoming and is activated with hypolarization in voltage ranges present during the diastolic depolarization phase, contrary to most ionic currents that are activated by depolarization. The funny channel is permeable to Na+ and K+ ions. Generally, students enrolled in the biophysics course at the undergraduate level are familiar with some voltage-gated channels of the Hodgkin and Huxley type. However, no channel that is activated by hiperpolarization is studied. In this work, a simulator was designed and developed for the study and understanding of the funny current present in the sinus node of rabbits. The simulator was programmed in Visual Basic Language ver. 6.0 for Windows® environment from XP to Windows® 10. The user can perform the experiments with the voltage clamp technique, modify the variables and concentrations of external Na+ and K+ and observe the effect on the amplitude of If and the kinetics of the curve I-V. It is recommended to be used as a support didactic material during biophysics and physiology courses in a computer room or remotely on any personal computer with minimal resources.
