The isolation of lectins by affinity chromatography with crosslinked hemicelluloses has been a common practice because of the variety of glycosides that they present, improving the isolation of different kinds of lectins, such as the galactose ligands.Lectins affinity for carbohydrates is so specific that a simple configuration of the chiral carbon can affect affinity, and there are lectins that are more related to alfa-galactosidic than beta-galactosidic residues, setting up that way, an anomeric recognition.The anomeric configuration of galactose residuesseems to have biological importance related to the behavior of some diseases and physiological processes. This work aimed to assess the anomeric recognition of two lectins reported as β-galactose ligands (PNA and ricin) and two lectins reported as α-galactose ligands (frutalin and jacalin) in two types of hemicellulose (xyloglucan of Tamarindus indica and galactomannan of Caesalpinia pulcherrima), subsequently crosslinked and used as chromatographic matrices. As a result,chromatographic profiles and retained fractions suggested preferential anomeric recognition by lectins for the hemicelluloses crosslinked. The galactomannan matrix retained 0,5mg of PNA lectin and 2,3mg of ricin lectin; meanwhile, the xyloglucan matrix retained 3,4mg of PNA and 3,2mg of ricin; results obtained by applying 5 mg of lectin. Ricin expresses a visible flexibility in anomeric recognition, while PNA shows a restricted recognition of β-galactose residues.Frutalin and jacalin did not show recognition of the xyloglucan matrix. This work proposes using hemicellulose reticles with epichlorohydrin as affinity chromatographic matrices for anomeric studies on recognizing galactose binding lectins.
Contextualización: En el artículo se aborda la aplicación de técnicas de análisis de datos multivariados en el sector agro, con el propósito de caracterizar a dos grupos de agricultores de aguacate hass, a partir de los datos que recogen las entidades que tradicionalmente prestan asistencia técnica rural en el país, las unidades municipales de asistencia técnica (UMATAS) y facilitar la toma de datos en campo y arrojando luces sobre la creación de nuevos indicadores de desempeño. Vacío de conocimiento: Deficiente aplicación de métodos e instrumentos de análisis de datos en el proceso de asistencia técnica rural, que permitan definir recomendaciones de actividades, insumos y manos de obra más específicas a la realidad de las distintas asociaciones o individuos de productores agrícolas Objetivos: 1) Analizar los datos recogidos a través del instrumento de diagnóstico a las fincas RUAT que realizan las Umatas en cumplimiento de su papel de acompañamiento técnico rural; 2) Aplicar las técnicas estadísticas de clasificación y organización de datos más pertinente a dicho instrumento de caracterización; y 3) Determinar la importancia que cumplen para la construcción de indicadores las variables que se recogen en los instrumentos de diagnóstico a las fincas. Metodología: A partir de la revisión de varios referentes teóricos sobre los pasos necesarios para la creación y selección de indicadores, se siguieron las siguientes etapas metodológicas: Desarrollo de un marco conceptual, selección de variables, normalización de los datos, análisis multivariado y ponderación de la información; otros pasos posteriores como la agregación de la información, análisis de robustez y sensibilidad, podrán ser parte de subsiguientes trabajos. Resultados y conclusiones: Se encontró que las variables que tuvieron mayor peso en la caracterización y agrupación de los productores fueron las pertenecientes al grupo de las buenas prácticas agrícolas y al de la producción, demostrando que con solo 8 variables se puede tener una buena aproximación a la caracterización de los productores, en lugar de las más de 36 variables de las que se compone el instrumento de diagnóstico denominado Registro de Usuarios de Asistencia Técnica (RUAT)
Los sistemas de producción agroecológicos colombianos pertenecen a los microfundios y minifundios que, por su tamaño, no obtienen adecuados ingresos para los productores. No obstante, el componente pecuario puede contribuir a la sostenibilidad económica de los hogares rurales, debido a los precios que estos productos alcanzan en el mercado y la continuidad en sus ciclos productivos. Esta investigación busca identificar el grado de asociación entre la ganancia neta de sistemas agroecológicas expresada en Salarios Mínimos Legales Vigentes (SMLV) y las variables tamaño del predio y proporción dedicada a la actividad pecuaria. A partir de 27 fincas encuestadas en la zona rural del centro del Valle del Cauca, fueron seleccionadas 14 por la presencia del componente pecuario y el cumplimiento de al menos tres principios agroecológicos y de sostenibilidad económica. A estos sistemas se les determinó la ganancia neta anual, tamaño del predio y proporción del área en producción pecuaria ajustado a un modelo de regresión lineal múltiple. Se estimó que el 77,15 % de la variación de los ingresos obtenidos en el sistema agroecológico son atribuibles al tamaño del predio y a la proporción dedicada a la actividad pecuaria (p = 0,000298). El tamaño del predio y la proporción de la finca dedicada a la actividad pecuaria afectan el nivel de ingresos de los productores y contribuyen a su sostenibilidad económica.
Studies related to DNA extraction are becoming more ambitious in the sense that large studies are intended to be carried out with minimum DNA sources. The DNA extracted must be of quality for genetic, forensic, population and genomic studies, these samples must be easy to obtain and product of efficient manipulation.Samples obtained from horsehair are an important technical challenge since they constitute the preferred non-invasive sample for genetic studies in horses, which has been shown to obtain reliable results in a short time. In this sense, working into effective techniques to optimizeDNA extraction of scarse samples is a pertinent task. In this study, different DNA extraction methods were evaluated from mane samples obtained from a population of wild horses from the Region of Arauca in eastern Colombia.Three DNA extraction methods were evaluated (phenol chloroform, alkaline lysis and twocommercial DNA extraction kit), DNA concentration, purity and qualityweredeterminate and PCR amplification product were obtain using primers for a hypervariable region of DNA mitochondrial. DNA preparation from hair roots using alkaline lysis was the most economical and efficient method with which it was possible to obtain high quality and quantity DNA.
El jaguar americano es una de las especies de felinos que habita en todo el continente americano. En Colombia, su distribución geográfica lo sitúa desde la Sierra Nevada de Santa Marta hasta el Amazonas. Diferentes factores antrópicos amenazan la persistencia de la especie, la cual se encuentra dentro de la lista roja CITIES, como especie amenazada. Análisis moleculares de especies, utilizando diversos marcadores de ADN, permiten discernir subespecies, filogeografía, diversidad genética y patrones de parentesco. El objetivo de este trabajo fue obtener DNA de buena calidad de muestras antiguas. Se analizaron fragmentos de ADN de tejido antiguo de jaguar (14 años) obtenido de fragmentos de piel sin tratamiento de preservación y se probaron tres protocolos de extracción de ADN y amplificó un segmento del gen mitocondrial ATP6-DF3. Los protocolos utilizados fueron: extracción por resina quelante-Quelex, Fenol–cloroformo y Kit de extracción comercial. Los resultados indican que en todos las muestras de fragmentos de piel de jaguar, se obtuvo ADN, a pesar de su antigüedad. Los métodos de extracción que presentaron mejores resultados fueron el que utilizó el kit comercial y fenol cloroformo, además el método de quelex mostró ser util, rápido y barato para obtener ADN de suficiente calidad, en muetras difícles, por su antigüedad y poco estado de preservación.
