A anemia falciforme (AF) é uma patologia crônica que possui fatores hereditários, descrita como autossômica recessiva, no qual as hemácias adquirem a forma de foice, uma condição mutagênica da proteína hemoglobina (Hb) encontrada na hemácia circulante no sangue e responsável pelo transporte de O2 e CO2. Analisar as evidências científicas acerca da indução de genotoxicidade provocada pela Hidroxiuréia (HU) em pacientes com doença falciforme. Trata-se de uma pesquisa de revisão integrativa, dos artigos disponíveis na íntegra, nos últimos 10 anos, dos idiomas português e inglês, com a estratégia de busca PICO. Os resultados obtidos, em populações com faixa etárias distintas, descrevem sendo possível uma instabilidade genômica que pode vir a possibilitar a mutação de células que posteriormente são denominadas carcinogênicas. O uso da HU, pode está causando alterações a nível hematológico e nas células de defesa, apresentando mielotoxicidade, induzindo a um processo de leucopenia e plaquetopenia de acordo com o tempo do uso do fármaco. De acordo com a análise comparativa dos estudos encontrados, pacientes com AF que fazem uso prolongado da HU, apresentam evidências de danos ao DNA.
