The number of solute-binding protein-dependent transporters in rhizobia is dramatically increased compared with the majority of other bacteria so far sequenced. This increase may be due to the high affinity of solute-binding proteins for solutes, permitting the acquisition of a broad range of growth-limiting nutrients from soil and the rhizosphere. The transcriptional induction of these transporters was studied by creating a suite of plasmid and integrated fusions to nearly all ATP-binding cassette (ABC) and tripartite ATP-independent periplasmic (TRAP) transporters of Sinorhizobium meliloti. In total, specific inducers were identified for 76 transport systems, amounting to Ϸ47% of the ABC uptake systems and 53% of the TRAP transporters in S. meliloti. Of these transport systems, 64 are previously uncharacterized in Rhizobia and 24 were induced by solutes not known to be transported by ABC-or TRAP-uptake systems in any organism. Hydroxyproline-rich proteins in plants offer a source of carbon and nitrogen to soil-dwelling microorganisms in the form of root exudates and decaying organic matter. This report describes an ABC-type transport system dedicated to the uptake of hydroxyproline in the legume endosymbiont Sinorhizobium meliloti. We have designated genes involved in hydroxyproline metabolism as hyp genes and show that an S. meliloti strain lacking putative transport genes (DeltahypMNPQ) is unable to grow with or transport trans-4-hydroxy-l-proline when this compound is available as a sole source of carbon. Expression of hypM is upregulated in the presence of trans-4-hydroxy-l-proline and cis-4-hydroxy-d-proline, as modulated by a repressor (HypR) of the GntR/FadR subfamily. Although alfalfa root nodules contain hydroxyproline-rich proteins, we demonstrate that the transport system is not highly expressed in nodules, suggesting that bacteroids are not exposed to high levels of free hydroxyproline in planta. In addition to hypMNPQ, we report that S. meliloti encodes a second independent mechanism that enables transport of trans-4-hydroxy-l-proline. This secondary transport mechanism is induced in proline-grown cells and likely entails a system involved in l-proline uptake. This study represents the first genetic description of a prokaryotic hydroxyproline transport system, and the ability to metabolize hydroxyproline may contribute significantly toward the ecological success of plant-associated bacteria such as the rhizobia.

It was hypothesized that EMG biofeedback relaxation training, applied to a diabetic patient, would result in a decreased level of insulin with fewer episodes of ketoacidosis. A 20-year-old female, diabetic since age nine, kept daily records of insulin doses and rated herself on an emotionality and a diabetic scale. A full-semester baseline was taken. This was followed by a semester-long training program during which the patient practiced relaxing her frontalis muscle with a portable EMG feedback unit which produced a geiger-counter-like click feedback. A cassette-tape series was used along with the portable EMG. The patient was encouraged to practice twice each day and to attempt to maintain a relaxed state even when not in the practice situation. The daily use of the portable unit was terminated at the end of the semester. In addition, the patient ceased practicing twice daily with the cassette tape. Daily insulin averaged 85 units for the six-week baseline and 59 for the final six weeks of the training period. Moreover, at the end of the training period the average dose had reached 43 units. During the training period the patient rated herself as decreasing in emotionality and in diabetic fluctuations.
