Prolonged storage of seeds may lead to decreases in seed quality, negatively affecting germination and vigor and preventing the obtaining of a promising stand. Seed quality is a critical factor, and the performance of the lot can be altered by vigor, dormancy and, mainly, by the time and conditions in which the seeds were stored. In the experiment, the physiological quality of seeds 14 genotypes of buffel grass (Cenchrus ciliaris L.) genotypes stored for a period of thirteen years in cold conditions was evaluated. The seeds were subjected to germination induction treatment with potassium nitrate (KNO3) and evaluated to identify seeds with presence and absence of embryos. The seed storage period affected their germination capacity and the use of KNO3 did not increase seed germination. A low number of full seeds was found for genotype 613 (12%). The relative germination percentages did not underestimate the buffel grass seed germination potential, as it is usually calculated, providing an improved distinction between the evaluated genotypes. The use of KNO3 increased the seedling root and shoot weights of some genotypes, as well as the dry matter weight of seedlings, an important characteristic for forage production. The prolonged storage affects the physiological quality of buffel grass seeds. The production of seeds without embryo is a problem found for this species, which affects the final quality of the seed lots produced.

Tetrazolium test (TZT) can quickly evaluate in detail the viability and vigor of seeds. This study aimed to determine the optimal conditions for conducting the TZT on seeds of Handroanthus spongiosus. For this purpose, seeds from three lots were pre-soaked in water for 16 h, followed by extraction of the tegument and immersion in tetrazolium salt solutions at different concentrations (0.01–0.1%), for increasing periods (1–4 h) and at 30 °C in the dark. The experimental design was completely randomized with a 4 × 4 factorial scheme with 25 seeds per repetition. We applied generalized linear models and the Tukey test for pairwise comparisons of the means at 5% probability. The viability/vigor results were compared with data obtained from the germination test at 25 °C using a subsample of seeds from the same lots. The time (1 h to 4 h) of immersion of the seeds in tetrazolium salt solutions did not cause a clear coloration difference. The seeds subjected to all treatment concentrations for 3 h presented average viability greater than 60%, with no difference in germination percentage. The TZT at 0.01% tetrazolium salt solution for 3 h was most efficient in assessing the viability of the Handroanthus spongiosus seeds.

Handroanthus spongiosus (Rizzini) S. Grose is an endangered tree species. However, its seed quality, storage, and conservation strategies are issues still unexplored. This study aimed to evaluate the physiological quality of H. spongiosus seeds subjected to different storage times, packaging, and environments for their conservation. A completely randomized experimental design was used, in a double factorial arrangement with an additional treatment (recently-harvested seeds), consisted of five storage times (up to 24 months) and six storage conditions, combining packaging types (permeable and impermeable) and environments (room, cold chamber, freezer, and liquid nitrogen conditions). Seed germination percentage and normal seedling percentage, shoot length, root length, and root to shoot dry weight ratio were evaluated. The seed germination and normal seedling percentages of H. spongiosus seeds conserved under room conditions decreased over the storage time. Normal seedling percentages decreased from the 12th month of storage onwards. Low and ultralow temperatures are recommended for short and medium-term conservation of H. spongiosus seeds, since they did not affect the growth of seedlings.

Morphoanatomical analysis of seeds contributes to knowledge of the development of seedlings and identification of species, as well as supporting conservation studies. The conservation of the species belonging to the Passiflora genus is crucial due to of the threats to the genetic resources of these species. Thus, the objective of this study was to morphoanatomically characterize Passiflora seeds, verify possible injuries to the tissues after cryopreservation and thus contribute to the conservation strategies of the species of this genus. Initially, seeds of Passiflora coccinea, P. edulis, P. gibertii, P. maliformis, P. morifolia, P. setacea, P. suberosa, and P. tenuifila collected from the Passion Fruit Active Germplasm Bank of the Embrapa Cassava and Fruits research unit (Embrapa Mandioca e Fruticultura) were analyzed. Then, their length, width and thickness, shape of the base and tip, and ornamentations present on the body and edge of the seeds were evaluated. The seeds of the species were placed in cryotubes and immersed in liquid nitrogen to assess possible cryoinjuries. The tegument and tissues of the seeds were examined by scanning electron microscopy. The seeds had varied biometric data, with average values of 4.63 mm for length, 3.28 mm for width, and 1.51 mm for thickness. Six ornamentation types were observed: reticulate for the species P. coccinea; finely reticulate for P. edulis; foveolate reticulate for P. gibertii and P. setacea; alveolate reticulate for P. maliformis and P. tenuifila; coarsely reticulate for P. morifolia; and falsifoveolate reticulate for P. suberosa. Some seeds suffered tegument cracks due to the freezing in liquid nitrogen, but without physiological damages to the embryo and endosperm. The cryopreservation of the seeds in the presence of the tegument significantly reduced the cryoinjuries caused to the embryo. Cryopreservation can be promising for long-term conservation of passion fruit seeds.

Studies of plant phenology in the Atlantic Forest can be enhanced by a greater understanding of the factors regulating vegetative and reproductive cycles. Dalbergia nigra (Vell.) Allemão ex Benth. is endemic and vulnerable in the Atlantic Forest. We analyzed abiotic aspects and plant traits that modulate the phenologies of D. nigra by monitoring 135 individuals in four subpopulations from different remnant forests for 24 months. The growth and shapes of the plants, as well as environmental variables, were determined. Circular analysis evidenced phenological variations among subpopulations and evaluation periods. Multiple factor analysis evidenced that phenological variations are mainly correlated with precipitation, temperature, and tree height. The combination of environmental conditions and plant characteristics affect synchronicity and phenological intensity. Low fruiting intensity (less than 50%) limits seed production and recruitment. We emphasize the importance of forest remnants and the need to increase D. nigra populations in future reforestation projects.
