SUMMARYThe virus growth in the pharyngeal area and the virus excretion in milk of susceptible and vaccinated dairy cows after intranasal instillation of foot-andmouth disease (FMD) virus type 01 were examined. Ten vaccinated cows were purchased through a market. Of these, nine had delivered their first calf. The cows were inoculated 2-9 months after having received the last dose of vaccine. All vaccinated cows resisted the intranasal challenge.
In skim milk obtained from susceptible cows after intramammary and intravenous inoculation (primary infected milk), foot-and-mouth disease (FMD) virus type O1 was slower inactivated by heat treatment than virus that had been added to pre-exposure skim milk. Residual virus infectivity in heated primary infected milk was more efficiently detected in bovine thyroid cell cultures than in secondary pig kidney (PK2) cell cultures. Untreated primary infected milk was found to inhibit both FMD-virus and vesicular stomatitis virus plaque formation in PK2 cells, suggesting the presence of interferon. The results of further tests confirmed that the interfering activity in unheated primary infected milk was indeed caused by an interferon. Interferon excretion in primary infected milk was investigated using a series of milk samples from three cows. Maximum interferon titres were found after 24 h, coinciding with or shortly after the first virus excretion peak. The results are discussed with particular reference to the use of primary infected milk in studies of the thermal inactivation of FMD-virus.
Zusammenfassung Die Testinfektion geimpfter Schweine mit Maul‐ und Klauenseuchevirus Empfängliche Schweine schieden nach oraler Infektion (Einreibung mittels Tuch) und nach parenteraler Infektion bis zu 107 plaquebildende Einheiten (PBE) Maul‐ und Klauenseuchevirus pro ml Maulhöhlenflüssigkeit aus. Nach oraler Testinfektion 6 Wochen zuvor vakzinierter Schweine war die Virusausscheidung vergleichsweise viel geringer. Bei oraler Testinfektion 20 Wochen nach der Impfung betrug die Virusausscheidung bis zu 105,5 PBE pro ml Maulhöhlenflüssigkeit, die Tiere blieben jedoch gesund. Die Art der Auf‐stallung (Einzel‐ oder Gruppenhaltung) hatte auf die Virusausscheidung keinen Einfluß. Nach parenteraler Testinfektion einzeln aufgestallter vakzinierter Schweine war die Virusausscheidung über die Maulhöhle abhängig von der Immunitätslage des jeweiligen Tieres. Wurden solche Schweine zusammen aufgestallt, so schieden selbst voll immune Tiere Virus aus. Bei parenteral testinfizierten Schweinen mit nachfolgender generalisierter Maul‐ und Klauenseuche fielen die Titer neutralisierender Serumantikörper zunächst geringgradig, stiegen dann aber am 4. Tag wieder deutlich an. Die Abhängigkeit der Ergebnisse von Impfstoff‐Wirksamkeitsprüfungen vom Modus der Testinfektion wird diskutiert. Résumé L'infection d'épreuve de porcs vaccinés avec un virus de la fièvre aphteuse Des porcs réceptifs ont excrété jusqu'à 107 unités formant des plaques (UFP) de virus de la fièvre aphteuse par ml de salive après une infection orale (frottement avec un linge) et une infection parenterale. Une infection d'épreuve orale chez des porcs vaccinés 6 semaines auparavant n'a provoqué comparativement qu'une excrétion viral beaucoup plus faible. Une infection d'épreuve 20 semaines après la vaccination a donné une élimination virale jusqu'à 105,5 UFP par ml de salive, les animaux restant sains. Le genre d'installation (animal isolé ou porcs en groupes) n'a eu aucune influence sur l'excrétion de virus. L'excrétion virale salivaire chez le porc isolé vacciné après une infection parentérale fut dépendante de la situation immunitaire de chaque animal. Si les porcs étaient tenus en groupe, même les animaux totalement immunisés, excrétèrent du virus. Chez les porcs infectés par voie parentérale présentant une fièvre aphteuse généralisée, les titres d'anticorps sériques neutralisants deminuèrent faiblement au début pour nettement augmenter le quatrième jour. Summary Two different aluminium hydroxide‐formaldehyde FMD vaccines were compared in pigs, with and without addition of 0.5 g. DEAE‐D per dose. Both showed improved potency after addition of DEAE‐D, manifested by higher serum neutralizing antibody titers and better protection against challenge. A modification of the second vaccine, containing only DEAE‐D as adjuvant, induced a slightly better serological response, but afforded slightly less protection against contact challenge than the one containing both DEAE‐D and aluminium hydroxide. In subsequent experiments in pigs with aluminium hydroxide FMD vaccines to which DEAE‐D had been added, it was shown that the method of challenge used greatly affects the results obtained. Contact challenge proved to be more severe than needle challenge, but both these methods were much more severe than oral challenge of vaccinated pigs either in a group or individually housed. Addition of DEAE‐D to concentrated aluminium hydroxide stock vaccines offers a possibility for a “last‐minute” potentiation of such vaccines for use in pigs. Wurde nur DEAE‐D als Adjuvans zugesetzt, so hatte dies eine geringgradig bessere serologische Reaktion und eine etwas schlechtere Immunität zur Folge. In weiteren Versuchen wurde nachgewiesen, daß die Ergebnisse von Immunitätsprüfungen am Schwein abhängig sind von der Methode der Testinfektion. Summary The efficacy of oral vaccination of calves with a modified live virus vaccine (Scour‐vax®‐2, Smith Kline) against rotavirus and/or coronavirus induced diarrhoea was examined under controlled conditions. Calves were vaccinated one hour after the first feed. Challenge was done orally with a mixture of virulent bovine rotavirus and bovine coronavirus. In the first experiments calves received colostrum free of antibodies against these viruses. Three control calves, challenged on the sixth day of life, developed severe diarrhoea after two to three days. Diarrhoea lasted for five to twelve days and on average the calves lost 12.5 per cent body weight. Rotavirus and coronavirus shedding was found in all three. In contrast, no diarrhoea was observed in two vaccinated calves challenged on day 6. After vaccination one shed rotavirus and one rotavirus plus coronavirus, but no virus excretion was found after the challenge. Three other vaccinated calves, that were not held completely isolated, all developed diarrhoea within three days, i.e. before the experimental challenge was performed. In each calf the beginning of diarrhoea coincided with rotavirus and coronavirus excretion. These calves lost six to seven per cent body weight. In further experiments calves were fed “positive” colostrum (from a pool collected on a Dutch dairy farm) for two days. Antibody titres of this pool against rotavirus and bovine coronavirus were as expected for mixed first‐day colostrum of dairy cows under the Dutch conditions. One out of three control calves fed colostrum from this pool developed severe diarrhoea after challenge on day 6; the others only passed somewhat loose or yellow faeces for a few days. The first calf shed both viruses, the last two either rotavirus or coronavirus. Weight losses were limited or did not occur. More severe diarrhoea was observed in six vaccinated calves, three of which were challenged on day 6 and three on day 8. None of these calves excreted rotavirus or coronavirus before the challenge. Two to four days after the challenge all six developed diarrhoea, the beginning of which coincided with the start of rotavirus excretion. In three calves coronavirus excretion was also found, but this started a few days later. The calves challenged on day 6 were less severely affected than those challenged on day 8; body weight losses averaged four and eight per cent, respectively. These results show that effective oral vaccination of calves against diarrhoea induced by rotavirus or bovine coronavirus may be blocked by specific antibodies present in colostrum and thereby confirm an earlier hypothesis based on observations in the field.
