Healthy mice were treared intravenously with the antileukemic L-asparaginase A by a single injection of 5,000 U/kg. At various times after injection the L-asparaginase (Asnase) content was determined in the blood-plasma and in the whole body; furthermore the simultaneous content of L-asparagine (Asn) was determined in the whole animal.Methods are described for the determination of Asnase and Asn in whole mice. The Asnase activity in the blood-plasma decreased rapidly during the first 4 hours after application. Afterwards an equilibrium of distribution was apparently reached, and the further decrease followed first order kinetics with a half-life of 3 h approximately.In the whole animal, the Asnase decrease followed from the beginning first order kinetics with a half-time of 3 h. After reaching the equilibrium, the relative volume of distribution was approximately 0.1 I/kg.In untreated mice the mean total Asn content was 26 nmoles per g. After Asnase application Asn fell, in just over one hour, below the detection limit. After two days it began to reincrease slowly; only after several days the initial value was found.The experiments have shown that Asnase also exhibits its effect apart from the circulation. It is concluded that the disturbance of the blood-mediated Asn supply is not the essential effect on sensitive cells. The problem of Asnase distribution and its influence on sensitive and resistant cells is discussed with regard to the reported results.
Die üblichen Bestimmungen der Katalase-Aktivität 1 " 3 sind mit dem Nachteil behaftet, daß die angewendeten H 2 0 2 -Konzentrationen so groß sind, daß sie das Ferment schädigen 4 . Dies kommt in einer Abnahme der Geschwindigkeitskonstanten mit der Zeit und somit in einer Krümmung der theoretisch als Gerade verlaufenden In Substrat/Zeit-Kurve zum Ausdruck. Will man trotzdem einigermaßen genaue Werte erhalten, so sind die Messungen in derartig kurzer Zeit durchzuführen, daß schon kleine Zeitfehler stark ins Gewicht fallen; eine weitere Behelfsmöglichkeit, das Arbeiten unter Eiskühlung 2 , stellt ebenfalls keine günstige Lösung dar. Feinstein 5 hat versucht, die wirksame Konzentration des H 2 0 2 klein zu halten, indem er an dessen Stelle Perborat einsetzte; jedoch findet nach seinen eigenen Angaben auch dann noch in kurzer Zeit eine erhebliche Inaktivierung des Enzyms statt.Die Tatsache, daß trotz bestehender Nachweise für kleine H 2 0 2 -Konzentrationen diese Nachweise, soweit wir feststellen konnten, nicht für die Katalase-Bestimmung angewendet werden, hat wohl ihre Ursache in scheinbaren methodischen Schwierigkeiten. Kleine Mengen Wasserstoffperoxyd lassen sich bequem mit Peroxydase und Di-o-anisidin nachweisen 6 ' 7 ; allerdings konnten wir feststellen, daß nach Anwendung'der üblichen Eiweißfällungsmittel von den eingesetzten kleinen H 2 0 2 -Mengen ein nicht unbeträchtlicher Teil verloren gegangen war. Dementsprechend führten auch unsere zunächst durchgeführten Versuche (über die hier im einzelnen nicht berichtet werden soll), die Katalase-Reaktion durch Eiweißfällung mittels Perchlorsäure oder dergl. zu unterbrechen und dann 1 G.
