ZusammenfassungBlutplasma wird beim Pferd unter anderem zur Therapie von aktiven Blutungen und Koagulopathien empfohlen. Für das Pferd existieren bezüglich der Stabilität von Gerinnungsfaktoren in Plasmapräparaten keine Untersuchungen. Ziel der vorliegenden Arbeit war es deshalb, den Einfluss von Lagerungsdauer und Lagerungstemperatur auf die Haltbarkeit von Gerinnungsfaktoren in verschieden hergestellten Plasmapräparaten zu untersuchen. In die Studie wurden 6 gesunde Freiberger-Pferde als Spendertiere einbezogen. Sie wurden zunächst einer Plasmapherese und 4 Wochen später einer Vollblutspende mit anschliessender Plasmagewinnung durch Blutbeutel-Zentrifugation oder Schwerkraft-Sedimentation unterzogen. Die Plasmapräparate wurden unmittelbar nach deren Gewinnung tiefgefroren und anschliessend bei -20º C oder bei -70º C gelagert. Die aktivierte partielle Thromboplastinzeit (aPTT), die Prothrombinzeit (PT), die Thrombinzeit (TZ), Faktor (F) V und F VIII sowie Antithrombin (AT) III wurden unmittelbar vor dem Einfrieren und anschliessend nach 90, 180, 270 und 360 Tagen untersucht. Das Herstellungsverfahren wirkte sich nur unbedeutend auf die Eigenschaften des Plasmas während der Lagerung aus. Alle gemessenen Parameter waren über den Untersuchungszeitraum von 360 Tagen bei einer Lagerungstemperatur von -70º C im Wesentlichen stabil im Referenzbereich, wohingegen bei einer Lagerungstemperatur von -20º C eine signifikante Verlängerung der Prothrombinzeit (PT), der aktivierten partiellen Thrombinzeit (aPTT) und der Thrombinzeit (TZ) sowie ein Aktivitätsverlust von Faktor (F) V und F VIII festgestellt werden konnte. Der Plasmaproteingehalt und die Antithrombin III-Aktivität waren auch bei -20º C grösstenteils stabil.Bei einer Lagerungstemperatur von -20º C waren die Globaltests gegenüber einer Lagerungstemperatur von -70º C während der Lagerung signifikant länger (P < 0.05). Die Aktivität der Faktoren V und VIII war bei -20º C signifikant geringer als bei -70ºC (P < 0.05), wohingegen sich die AT IIIAktivität bei Lagerungstemperaturen von -20º C und -70º C nicht unterschied. Nach den Ergebnissen der vorliegenden Untersuchung ist die Lagerung von Pferdeplasma bei einer Temperatur von -70º C gegenüber einer Lagerung von -20º C zu bevorzugen, da über die Dauer von mindestens 1 Jahr ein unverändertes Gerinnungspotenzial aufrecht erhalten werden kann. Auf Grund dieser Eigenschaften bietet bei -70º C gelagertes Pferdeplasma optimale Voraussetzungen zur Therapie von Erkrankungen mit einem Mangel an Gerinnungsfaktoren.Schlüsselwörter: Plasmapherese, Plasma, Pferd, Gerinnungsfaktoren, Lagerung
