DNA-content and size of the nuclear areas in different zones of malignant melanomas of different histological types and in dysplastic naevi were measured in order to provide information on the histogenesis and proliferative behaviour of human malignant melanoma. The results were compared with those from normal epidermis, common naevi, and reactive melanocytic hyperplasias. The mitotic index of melanomas--divided into different topographic zones in an analogous way--was also determined. The DNA-histographs of all naevi and reactive melanocytic hyperplasias showed a diploid maximum, but the dysplastic naevi had a larger proportion of nuclei with hyperdiploid and tetraploid DNA-content, indicating an increased proliferative activity. The mean values (X) of nuclear areas in dysplastic naevi (DN) were about the same as in common naevi (CN) and slightly lower than in superficial spreading melanomas (SSM). The coefficient of variability (cv) as an indicator of anisokaryosis was markedly higher in DN (27.8) and SSM (29.3) than in CN (20.2). In DNA-content we found similar results: almost no difference in mean values, but DN taking an intermediate position between CN and SSM with respect to cv (CN: 12.3; DN: 21.0; SSM: 36.6). There was no unequivocal evidence in these data for DN being a precancerous stage. Superficial melanomas with a nodular component ("SSM/NM") differed from SSM and NM by increased DNA-content and greater variability of nuclear areas and showed the clearest features of malignancy in their DNA-histographs. The mitotic indices had rather low values in SSM and intraepidermal marginal zones of "SSM/NM" on one hand and markedly higher values in NM and nodular parts of "SSM/NM" on the other. The highest mitotic counts were found in the three investigated metastases.

Mit 11 Abbildungen in 31 EinzeldarstellungenZu Beginn mSchte ich Ihnen sehr herzlich ffir diese Einladung danken, vor Ihnen zu einem Thema sprechen zu dfirfen, das seit jeher im Mittelpunkt chirurgischen Itandelns steht und zugleich zu einem der interessantesten modernen Forschungsgebiete gehSrt.Denn die Wundheilung ist primer ein Bindegewebsproblem, wie erstreals MARCIA,1) in seiner klassischen Beschreibung und seitdem viele berfihmte Autoren zeigten. Diese morphologische Bindegewebsforschung verwendete in den letzten Jahren zunehmend neue Methoden. Die wichtigsten sind: 1. die Histochemie, 2. die Elektronenmikroskopie und 3. die Autoradiographie. Die damit gewonnenen Erweiterungen unserer bisherigen Kenntnisse fiber die Morphologie der Wundheilung will ich Ihnen zeigen. Dabei werden also 1. die neueren Daten fiber den Ersatz des in der Wunde zerst6rten Gewebes im Vordergrund stehen --und 2. die Beeinflussung dieser Wundheilung durch vor, ws und nach Operationen gebrs Medikamente.Bindegeweb~'bausteine. Da die Wundheilung also prim~ir ein Bindegewebsprozefi ist, stelle ich Ihnen zun~chst die wichtigsten neueren Daten fiber dessen drei }tauptbestandteile : Zelle, Grundsubstanz und Faser vor.Sie sind in einer schematischen Darstellung zusammengefaBt --zur Erinnerung an die morphologischen Details, mit denen wir jetzt zu arbeiten haben (Abb. l a).Wir besprechen :
Wound healing was investigated by morphological and biochemical methods. The results show that wounds cause disturbances in the equilibrium between the physiological anabolic and catabolic processes of cells, ground substance, and fibers (with increased turnover rates). The primary increase of catabolism is responsible for the secondary compensatory enhanced anabolism, especially of proteoglycans and collagen. Kind, size, and localization of wounds determine the course of wound healing and therefore the extent of adaptation of structure, function, and biomechanical capacities on the surrounding uninjured tissue.
1 Allgemeines Schulz und Ruff [1] postulieren aufgrund von numerischen Studien an vierseitig gelenkig gelagerten Platten, daß die derzeit in den maßgebenden Regelwerken DIN 18 800 [2], [3] und Eurocode 3 [4], [5] vorgesehene Beulkurve Ergebnisse auf der unsicheren Seite liefern kann, sofern geometrische Imperfektionen in Anlehnung an die in DIN 18 800-3 [3] festgelegten Höchstwerte der unvermeidbaren Herstellungsungenauigkeiten angesetzt werden. In einer ergänzenden Stellungnahme [6] wird diese Aussage zwar teilweise relativiert, allerdings erscheint es weiterhin sinnvoll, aus Sicherheitsgründen die derzeitigen Regelungen zu überprüfen. Dies gilt insbesondere dann, wenn zusätzlich Eigenspannungen berücksichtigt werden. Deshalb wurde der zweitgenannte Autor vom Deutschen Institut für Bautechnik (DIBt) beauftragt, Untersuchungen zur Klärung dieser Fragestellung durchzuführen [7]. 2 Beulkurven Die Tragfähigkeit eines Bauteils kann durch örtliches Beulen einzelner Querschnittsteile reduziert werden. Die Größenordnung der Reduktion kann durch eine Trag-In neuester Literatur werden Zweifel geäußert, ob die in DIN 18 800 und Eurocode 3 verwendete Winter-Kurve ausreichend den Einfluß von Imperfektionen auf das Tragverhalten normalspannungsbeanspruchter, beulgefährdeter Querschnittselemente berücksichtigt. Basierend auf früheren Forschungsarbeiten und eigenen Untersuchungen werden Einflüsse der Lagerung, des Seitenverhältnisses und von Eigenspannungen auf die Traglast diskutiert. Es ergibt sich ein Widerspruch zwischen numerisch ermittelten Traglasten und den empirischen Erfahrungswerten. Dieser kann nur dadurch aufgelöst werden, daß anstatt von schlimmstmöglichen, nur anregende Imperfektionen in Ansatz gebracht werden. Eine Zuschärfung der in den Normen verwendeten Beulkurven wird als nicht notwendig erachtet. Load carrying capacity of thin-walled cross-section elements in view of imperfections. It was concluded from recent research results that the plate buckling curves, which are established similar in Eurocode 3 as well as in DIN 18 800, do not in all cases safely reflect the ultimate load of locally buckled cross-section elements. Based on earlier research work and own investigations, the influence of boundary conditions, of ratio of dimensions and of residual stresses on the load carrying capacity is discussed in detail within this paper. The studies result in a contradiction between the numerically determined ultimate loads and the empirical experiences. This can only be resolved, if stimulating imperfections are taken into account instead of worst-case imperfections. It is not considered necessary to tighten the current plate buckling curves used in the codes. lastkurve, also eine Beulkurve, ausgedrückt werden.
