Microsatellite markers have been developed from a complementary DNA (cDNA) library of red sea bream, Chrysophrys major. Twenty‐eight microsatellites were selected for designing microsatellite primers, of which 11 gave working primer pairs. Observed and expected heterozygosities varied from 0.33 to 1.00 and from 0.38 to 0.83, respectively. Five additional fish species assessed for cross‐species amplification revealed between one and six positive amplifications and between 0 and 6 polymorphic loci per species.
Stocking and introduction of fish have been widely applied in Yunnan Province, China, for many years to mitigate for declines in natural stocks and commercial catches, and to restore or create new fisheries. The present paper assesses the results of activities concerned with stock enhancement programmes to evaluate the factors which have contributed towards the success of these projects. Factors such as natural lake conditions, stocking density, the age and size of fish stocked, and the mechanisms of stocking are discussed. Stocking density was evaluated using the equations Yp = [B(P/B)Uf]/Kf or PYp = (P × Uf)/Kf, which were based on the ecological carrying capacity of the recipient lake and other criteria. The stocking density in most lakes in Yunnan was high, but the recapture rates and total yield did not reach the predicted levels. This may have been an effect of the size of the fingerlings released (finger‐lings >10 cm have a greater chance of survival). Key management measures for stocking and the ecological impacts of overstocking grass carp, Ctenopharyngodon idella (Valenciennes), are also described.
Two experiments were conducted to investigate the immune-enhancing effect of dietary supplement with Astragalus polysaccharides (APS) on the Pacific white shrimp, Litopenaeus vannamei. In experiment 1, the optimal APS dose was determined based on the immune responses of shrimps fed APS diet for 30 days. In experiment 2, the effect of APS supplementation on immune response of shrimp suffering white spot syndrome virus (WSSV) challenge was determined. Results showed that the total haemocyte count and phagocytic activity in shrimps fed APS diets significantly (p < .05) increased in comparison with those fed the basal diet. Dietary supplement with APS markedly (p < .05) increased the activity of phenoloxidase (PO), total superoxide dismutase (SOD), lysozyme (LZM), acid phosphatase and alkaline phosphatase in shrimp hemolymph, but decreased the maleic dialdehyde (MDA) content. Significantly higher (p < .05) activity on PO, SOD and LZM and lower (p < .05) MDA content have also been found in shrimps suffering WSSV challenge. Therefore, APS could be used as a safe and effective feed additive in shrimp aquaculture, and the optimal dose of APS for the Pacific white shrimp was suggested to be 0.2 g/kg based on our results.

To identify quantitative trait loci (QTL) that affect body shape in common carp Cyprinus carpio, a linkage map, 2159·23 cM long, was constructed with a total of 307 markers covering 51 linkage groups (LG). The map included 167 new single nucleotide polymorphism (SNP) markers derived from expressed sequence tags (EST) together with 140 microsatellite markers reported earlier. A primary genome scan was conducted for QTL for standard length (L(S)), head length (L(H)), body height (H(B)), body width (W(B)) and tail length (L(TAIL)) in an F1 line containing 92 offspring. A total of 15 suggestive QTL on six LGs were found to associate with L(S), L(H), H(B), W(B) and L(TAIL) which explained 10·7-17·4% of the variance. Five significant QTL were detected for body-shape related traits and located for LGs (lg1, 12 and 20). These QTL included: one associated with L(S) (21·1% variance explained), three for H(B) (almost 20% variance explained) and one for W(B) (20·7% variance explained).
