Multicomponent reactions (MCRs) are fundamentally different from two‐component reactions in several aspects. Among the MCRs, those with isocyanides have developed into popular organic‐chemical reactions in the pharmaceutical industry for the preparation of compound libraries of low‐molecular druglike compounds. With a small set of starting materials, very large libraries can be built up within a short time, which can then be used for research on medicinal substances. Due to the intensive research of the last few years, many new backbone types have become accessible. MCRs are also increasingly being employed in the total synthesis of natural products. MCRs and especially MCRs with isocyanides offer many opportunities to attain new reactions and basic structures. However, this requires that the chemist learns the “language” of MCRs, something that this review wishes to stimulate.

Methyl-Derivates durch Umsetzung von Nitrobenzolsulfonamid rnit Methyl-isocyanat und anschliedende Reduktion in allen Einzelheiten beschrieben und ferner angegeben war, da8 man analog durch Verwenden von Athyl-, Propyl-und Butylisocyanat die hoheren Homologen erhalten konne. Die Beschwerdeabteilung stellte sich auf den Standpunkt, es seien vier Butyl-Isomere moglich, fur kein einziges seien in der Entgegenhaltung Eigenschaftsangaben gemacht, also sei die in der Anmeldung allein beanspruchte n-Butyl-Verbindung als neuer Stoff anzusehen. Da diese Verbindung nicht bestrittene, hervorragende therapeutische Eigenschaften besitze, sei auch die Patentfahigkeit des Analogieherstellungsverfahrens anzuerkennen:Dieser Standpunkt der Beschwerdeabteilung des Osterreichischen Patentamtes ist einleuchtend. Zweifellos lehrt die angegebene Literaturstelle, dal3 die homologen Stoffe wahrscheinlich nach der angegebenen Methode erhaltlich sind. Es bedurfte also fur einen Sachverstandigen nur des Aufwandes eines Analogieverfahrens, um gegebenenfalls einen der genannten homologen Korper oder auch die nicht genannten, noch hoheren Homologen, wie die verschiedenen isomeren Amyl-, Hexyl-, Heptyl-Derivate usw. darzustellen. Mit der Tatsache, dad diese Stoffe voraussichtlich e r h a l t l i c h sind, ist aber die Offenbarung der Druckschrift abgeschlossen, die Stoffe selbst sind nicht beschrieben; es fehlen auch die Konstanten und die Angaben iiber die Eigenschaften der iibrigen Homologen. Die Kausalkette: ,,Amgangsstoff -Arbeitsweiseneuer Stoff" ist also nicht vollendet, der neue Stoff, das Endglied, fehlt. 1 -Ferrocenylethyldimethylamine ( 7) is readily obtained from 1 -ferrocenylethanol by treatment with phosgene and then dimethylamine. The resolution of 7 with (i?)-(+)-tartaric acid is particularly easy and both antipodes are obtained in high yield. Qualitatively the steric course of the lithiation 7 -> 8 is predictable and proceeds with high stereoselectivity (96:4). Subsequent reactions of 8 with trimethylchlorosilane, formaldehyde, or benzophenone produce 9a-c. The quaternization of 9 in the presence of nucleophiles leads to substitution of the dimethylamino group in 9 with complete configurational retention, which is confirmed by nmr and the cycle {9a, 10, 14,13 j. The retention is due to the ease of formation of 11 and its configurational stability under suitable conditions. The a-addition of immonium ions and anions (OH-, SeH-, S2O32-, N3-NCO-, NCS-, R-COz-, RO-COz-) to isonitriles, accompanied by secondary reactions, provides a means for the one-stage synthesis of organic nitrogen compounds starting with two to five diferent components. Thus, by the condensations of amines (ammonia, primary, and secondary aliphatic and aromatic amines, hydrazines) and aldehydes or ketones with isonitriles and acids, a number of a-aminocarboxylic acid amides, thioamides, selenoamides, 1,j-disubstituted tetrazoles, hydantoin imides, thiohydantoin imides, a-acylamino carboxylic acid amides, oiigopeptide derivatives, /3--lactams, derivatives of penicillanic acid, urethanes, diacylimides, and various hydrazine derivatives, can be prepared. The reactions are easily carried out and take place under mild conditions. Yields of more than 90 % are frequently encountered.
