Foram caracterizados 41 isolados de Acidovorax avenae subsp. citrulli com base em aspectos fisiológicos e bioquímicos. Todos os isolados induziram sintomas típicos da mancha-aquosa em plântulas, plantas e frutos de meloeiro (Cucumis melo) e melancieira (Citrullus lanatus). Pelo teste de agrupamento de Scott-Knott (P = 0,05) os isolados foram separados quanto ao índice de doença em 5 e 7 grupos, respectivamente para plântulas de meloeiro e melancieira, e em 2 grupos para plantas das duas hospedeiras. Em frutos, os isolados foram separados em 3 e 10 grupos para a variável diâmetro da lesão externa e 2 e 9 grupos para profundidade da lesão, respectivamente para melão e melancia. Todos os isolados induziram reação de hipersensibilidade em fumo (Nicotiana tabacum); utilizaram os compostos asparagina, L-leucina e DL-ácido lático; produziram enzimas lipolíticas e o fitohormônio ácido indol acético; foram sensíveis a oxicloreto de cobre (120 µg mL-1), óxido cuproso (120 µg mL-1), hidróxido de cobre (138,2 µg mL-1), sulfato de estreptomicina (25 µg mL-1) e Agrimaicin 500 (428 µg mL-1); e resistentes a kasugamicina (87 µg mL-1), agrimicina (200 µg mL-1), eritromicina (15 µg), gentamicina (10 µg), amoxicilina (10 µg), neomicina (30 µg), estreptomicina (10 µg), norfloxacina (10 µg) e rifampicina (5 µg). Nenhum isolado apresentou atividade pectinolítica, amilolítica, celulolítica e proteolítica ou produção do polissacarídeo levana e da toxina siringomicina. Foi constatada variabilidade entre os 41 isolados de A. avenae subsp. citrulli quanto à sensibilidade à tetraciclina (30 µg), sendo 41,5% resistentes, 46,3% moderadamente sensíveis e 12,2% altamente sensíveis.

RESUMOXanthomonas campestris pv. viticola (Xcv), que causa o cancro bacteriano da videira, sobrevive em plantas infectadas, epifiticamente em órgãos da parte aérea e pode ser veiculada em mudas e/ou bacelos infectados. O trabalho teve como objetivo investigar possíveis hospedeiros alternativos do patógeno, visando fornecer subsídios para o manejo da doença. A partir das plantas invasoras Alternanthera tenella, Amaranthus sp., Glycine sp. e Senna obtusifolia com sintomas similares aos do cancro bacteriano da videira, coletadas em parreirais de Juazeiro e Petrolina, no Submédio São Francisco, foram isoladas bactérias semelhantes a Xcv. No entanto, nenhuma bactéria foi isolada de plantas de Commelina benghalensis e Azadirachta indica com sintomas semelhantes. A patogenicidade dos isolados bacterianos obtidos foi confirmada em plantas de A. tenella, Amaranthus sp., Glycine sp., S. obtusifolia e em mudas de videira cv. Red Globe, em condições de casa de vegetação. As plantas invasoras Chamaesyce hirta, Dactyloctenium aegyptium, Eragrostis pilosa e Pilea sp., inoculadas artificialmente com os isolados Xcv1 e UnB1216, também desenvolveram sintomas típicos do cancro bacteriano.Palavras-chave adicionais: Vitis sp., cancro bacteriano da videira, plantas invasoras. ABSTRACT Alternative hosts of Xanthomonas campestris pv. viticolaXanthomonas campestris pv. viticola (Xcv), which causes the bacterial canker of grapevine, survives in infected plants as an epiphyte on aerial plant parts and may be carried in infected transplants and/or cuttings. This study was performed to investigate putative alternative hosts of the pathogen, aiming to find support for disease management. Bacteria similar to Xcv were isolated from the weeds Alternanthera tenella, Amaranthus sp., Glycine sp. and Senna obtusifolia showing canker-like symptoms and collected in vineyards of Juazeiro and Petrolina at the Submédio São Francisco. No bacteria were isolated from Commelina benghalensis and Azadirachta indica showing similar symptoms. The pathogenicity of the isolates was confirmed in plants of A. tenella, Amaranthus sp., Glycine sp., S. obtusifolia and grapevine cv. Red Globe, under greenhouse conditions. The aim of this work was to investigate the ability of Acidovorax citrulli, the causal agent of bacterial fruit blotch (BFB) of cucurbits, to colonize melon tissues. Under greenhouse conditions leaves and seeds were inoculated with a spontaneous mutant of A. citrulli (group I) resistant to rifampicin (Ac1 Rif), and samples of hypocotyls, cotyledonary leaves, roots, leaves and stems were processed at three-day intervals. When the 1 st pair of melon leaves had been inoculated, A. citrulli was only detected until the 10 th pair of leaves (1.3 x 10 3 CFU g-1 of leaf) and the consecutive stem (3.3 x 10 3 CFU g-1 of stem) on the 30 th day. However, after seed inoculation A. citrulli colonized the hypocotyl, roots, cotyledonary leaves, leaves and stems and was only detected until the 4 th pair of leaves (1.62 x 10 2 CFU g-1 of leaf) and the consecutive stem (9.1 x 10 1 CFU g-1 of stem) on the 24 th day post-inoculation. In conclusion, A. citrulli colonized different parts of the melon plant over time, depending on its initial location, leaves or seed. This confirms what has been observed in the field, that expanded leaves and stems are the main inoculum sources for melon blossoms and fruit, therefore providing scientific bases for developing more effective strategies for BFB management.
O objetivo deste estudo foi determinar o efeito da temperatura (0 a 45°C), pH (4 a 10) e concentração de NaCl (1 a 10%) sobre o crescimento de Acidovorax avenae subsp. citrulli (Aac). Quatro estirpes de Aac foram cultivadas em meio de cultura líquido específico e o crescimento avaliado pela absorbância (580nm). Os dados obtidos foram submetidos à análise de regressão não linear. O crescimento das estirpes também foi avaliado em caldo base para fermentação contendo 1% dos carboidratos glicose, galactose, ramnose, sacarose, lactose, maltose, amido, inulina, manitol, dulcitol, sorbitol e salicina, indicada pela mudança da cor do meio. Com base nas análises de regressão, as temperaturas mínima, ótima e máxima para crescimento de Aac foram, respectivamente, 1, 32 e 41°C; o pH ótimo para crescimento dessa bactéria foi 7,4 com os extremos mínimo de 4,0 e máximo de 10,8 e; o crescimento de Aac decresceu com o aumento da concentração de NaCl, sendo o nível de 6,2% letal. Todos os carboidratos testados foram utilizados pelas estirpes de Aac como fonte de carbono, com pequena variação de crescimento observada pela velocidade e intensidade da utilização do substrato com produção de ácido.
