Commercially available probiotics are routinely administered as feed supplements in aquaculture important species. Among them, the European sea bass (Dicentrarchus labrax) is the most widely reared fish in the Mediterranean, whose rearing systems are highly variable between countries, affecting at some level the sustainability of production. After random isolation of autochthonous gut bacteria of the sea bass, their identification and pathogenicity testing, we have selected three potentially probiotic isolates; Pseudoalteromonas sp., Alteromonas sp., and Enterovibrio coralii. Selected isolates were tested and their immunostimulative efficiency was compared with a commercially available Lactobacillus casei isolate, inferring inflammatory, apoptotic and anti-pathogen response of sea bass’ peripheral blood leukocytes. Phagocytic activity, respiratory burst, and expression of lysozyme, Mx protein, caspase 3, TNF-α, IL-10 genes was measured 1, 3, 5, and 12 h post-stimulation by four bacterial isolates to evaluate early kinetics of the responses. Best immunostimulative properties were observed in Pseudoalteromonas-stimulated leukocytes, followed by Alteromonas sp. and L. casei, while Enterovibrio coralii failed to induce significant stimulation. Based on such in vitro assay intestinal autochthonous bacterial isolates showed to have better immunostimulative effect in sea bass compared to aquaculture-widely used L. casei, and further steps need to engage tank and field feeding trials to evaluate long-term prophylactic suitability of the chosen isolates. A panel of biomarkers that represent pro-/anti-inflammatory, pro-/anti-apoptotic, and anti-bacteria/viral responses of the fish should be taken into consideration when evaluating the usefulness of the potential probiotic in aquaculture.
Within this paper, reproduction traits of the Adriatic population European hake, Merluccius merluccius (Linnaeus, 1758), were investigated. Specimens of the target species were monthly collected (N=1173) during 2019 along the eastern Adriatic Sea with commercial bottom trawlers. Overall, European hake total body length varied between 15.5 and 49.5 cm (mean±SD=27.5±5.0 cm) and domination of females was noted in larger length classes (TL>31.0 cm). The sexual ratio of all specimens was m/f=0.64; male prevalence was established over the year, except during the spawning peaks (December, June) when the sex ratio was in favour of the females. According to European hake’s maturity stages, gonad weights, gonadosomatic indexes and histological analysis of the gonad tissues, this species in Adriatic spawn twice in one year; the spawning period was from December to February, while second smaller signs of spawning were observed at the beginning of the summer, in June. 50% of the European hake male and female specimens sexually matured at 20.7 cm and 22.4 cm of total length, respectively.
This study tests the suitability of the gilthead seabream scales as a proxy for origin selection in wild and anthropogenically pressured environments. Scale morphology and microchemistry were used to discriminate the habitat selection of two wild, farmed and wild farm-associated populations where landmark and outline-based scale morphometrics, trace-element chemistry and scale microstructure characteristics were analysed. The morphometric techniques successfully differentiated between the farmed and wild origin scale phenotypes. Reduced discrimination sensitivity between the wild and wild farm-associated origin was, however, reported. The discrimination based on microchemistry (B, Ba, Mn, K, Sr and Zn) classified the scales with high accuracy according to their origin (wild vs. farmed vs. wild farm-associated) and sampling locations, thus proving itself as a powerful tool in provenance study of gilthead seabream. Disparity in scale microstructure characteristics accounted for radii, circuli and inter-circulus spacing, hence unveiling the differences in growth and environmental conditions between the wild and farmed fish. In brief, scale shape was found to be a potent exploration tool for farmed fish identification, whereas scale microchemistry yielded a good resolution in identifying gilthead seabream membership among different habitats. Considering the importance of this species in aquaculture and fisheries throughout the Mediterranean, more research is needed to assess the usefulness of scales as nonlethal biogeochemical tags.
The Pacific oyster Crassostrea gigas was introduced for the aquaculture purposes in many different parts of the world. However, the species has never been officially introduced for commercial farming in the Croatian part of the Adriatic Sea. Interestingly, in the 1970s, the Pacific oyster has been reported in the natural habitats of the Lim Bay, in northern Adriatic Sea. Although the species was recorded there, there is a lack of knowledge about its biology and ecology in this region, including a reproductive cycle. Information on the reproductive biology of non-indigenous species in new areas is crucial for prediction of their future survival and possible spread in invaded habitats. In this study, we provide the first data on the reproductive biology of the Pacific oyster in the Adriatic Sea, the northernmost part of the Mediterranean Sea. Quantitative and qualitative methods of gonad tissue analysis were conducted, and effects of environmental conditions on the gametogenesis were evaluated during two reproductive cycles. Our study confirmed that environmental conditions in the Lim Bay were favourable for the reproduction of the species. The Pacific oyster reproduced once per year and had a prolonged spawning period. Water temperature was the main factor affecting gonad development and oocyte size, while chlorophyll a concentration had an impact on the oocyte development in terms of their size.
The blue jack mackerel Trachurus picturatus (Bowdich, 1825) specimens (N = 155) were collected during the MEDITS survey, done along the eastern side, precisely, of the Croatian fishing ground in July 2018. Biometrical analysis of ten morphometric and five meristic characters, as well as genetic analysis proved that the collected specimens were blue jack mackerel. The total length (TL) and weight (W) of all observed specimens ranged from 9.2 to 33.7 cm (12.15 ± 2.95 cm) and from 5.79 to 384.94 g (17.64 ± 39.42 g), respectively. All calculated length–length relationships were linear (r > 0.923). Sex was determined only on two larger specimens (28 cm < TL < 32.8 cm), which were females. In the length–weight relationship, positive allometry was established (b = 3.1789). Based on 37 partial cytochrome b sequences, the overall haplotype diversity (h) of 0.812 ± 0.048 and nucleotide diversity (π) of 0.0064 ± 0.0007 indicated high levels of haplotype and low nucleotide diversity. The obtained sequences were compared to previously published research within the Northeast Atlantic Ocean and the Mediterranean Sea, confirming the absence of genetic structure among these populations.
