Drug repositioning has been an important ally in the search for new antifungal drugs. Statins are drugs that act to prevent sterol synthesis in both humans and fungi and for this reason they are promissory candidates to be repositioned to treat mycoses. In this study we evaluated the antifungal activity of atorvastatin by in vitro tests to determine the minimum inhibitory concentration against azole resistant Candida albicans and its mechanisms of action. Moreover, the efficacy of both atorvastatin-loaded oral and vaginal emulgels (0.75%, 1.5% and 3% w/w) was evaluated by means of in vivo experimental models of oral and vulvovaginal candidiasis, respectively. The results showed that atorvastatin minimal inhibitory concentration against C. albicans was 31.25 μg/ml. In oral candidiasis experiments, the group treated with oral emulgel containing 3.0% atorvastatin showcased total reduction in fungal load after nine days of treatment. Intravaginal delivery atorvastatin emulgel showed considerable effectiveness at the concentration of 3% (65% of fungal burden reduction) after nine days of treatment. From these findings, it is possible to assert that atorvastatin may be promising for drug repositioning towards the treatment of these opportunistic mycoses. Lay Summary Atorvastatin is a statin drug that presents antifungal activity. This study showed that atorvastatin-containing oral and vaginal emulgels were able to treat vulvovaginal and oral candidiasis of infected animal model. Therefore, we showcased that atorvastatin may be a possible therapeutic agent in order to be a used to control opportunistic mucosal fungal infections caused by Candida albicans.

The gastrointestinal tract houses the largest and most complex community of microorganisms, and this bacterial colonization of the human intestine by environmental microbes begins immediately after the birth. The intestinal microbiota has several important and unique functions, including metabolic functions such as the biotransformation of drugs and the digestion of dietary compounds; a mucosal barrier function by inhibiting the invasion of pathogens and an immunomodulatory function. On the other hand, some commensal bacteria can be pathogenic, causing infections if the natural host is compromised and, in predisposed hosts, the intestinal microbiota can be involved in nosocomial infection. The translocation of bacteria through the intestinal wall is considered one of the main causes of nosocomial infections. The aim of this review is to provide a comprehensive view of the human gut microbiota, its main functions, its role in health and disease, addressing the correlation between intestinal microbial composition and nosocomial infections.

O agente etiológico mais comumente isolado das infecções do trato urinário (ITUs) é a bactéria Escherichia coli (E. coli), a qual é responsável por cerca de 40% dos casos. Sendo microrganismos Gram-negativos, as E. coli possuem uma parede celular complexa, motivo pelo qual alguns antibióticos são menos eficazes quando comparados com sua com sua ação contra bactérias Gram-positivas. Diante disso, o objetivo deste trabalho é avaliar o perfil de sensibilidade de amostras clínicas de bactérias E. coli aos antimicrobianos usados no tratamento da ITU através do teste de difusão em ágar. Após a coleta e análise dos dados foi possível observar que a maioria dos isolados clínicos apresentou resistência a amoxicilina e foram mais sensíveis a nitrofurantoína e a cloranfenicol. A partir das informações obtidas após a análise dos dados foi possível observar que apesar do rígido controle da administração de medicamentos antimicrobianos, a incidência de bactérias resistentes a tais medicamentos é alarmante.

A 4-hidroxicumarina (4H) é um composto fenólico natural e sintético que apresenta atributos importantes, incluindo propriedades anti-HIV, antitumoral e anticoagulante. Considerando suas importantes atividades biológicas e a utilização da 4H, o objetivo do presente estudo foi investigar as atividades tóxica, mutagênica, antimutagênica e antioxidante da 4H, obtido comercialmente, em experimentos in vivo e in vitro. Os efeitos da administração oral aguda na dose de 200 mg / kg de 4H foram investigados pela triagem hipocrática em animais. O ensaio de micronúcleo em sangue periférico e o teste de Ames foram utilizados para análise de mutagenicidade, enquanto a antimutagenicidade foi testada contra a ciclofosfamida também pelo ensaio de micronúcleo in vivo. Para todos os testes in vivo foram utilizados camundongos Swiss. A atividade antioxidante foi determinada pela capacidade da amostra 4H de sequestrar o radical estável 2,2-difenil-1-picrilhidrazil (DPPH). Os resultados mostraram que o 4H não induziu alterações no comportamento ou na histologia dos animais tratados nem na atividade mutagênica (gênica ou cromossômica). Por outro lado, exibiu efeito protetor contra ciclofosfamida e potencial antioxidante. A 4H é amplamente utilizada clinicamente como uma molécula precursora na síntese de fármacos anticoagulantes sintéticos. Portanto, os resultados obtidos são de extrema importância, pois demonstram ausência de potencial mutagênico e proteção contra danos cromossômicos.
