La papa (Solanum tuberosum L.) es el tercer cultivo alimenticio más importante del mundo en términos de consumo humano, después del arroz y del trigo, es un alimento básico para países desarrollados como Europa y Estados Unidos. Este cultivo es afectado por problemas fitosanitarios, entre los que sobresale Ralstonia solanacearum. El objetivo de este estudio fue identificar mediante prueba fenotípica los mecanismos de resistencia de la bacteria Ralstonia solanacearum, aislada del cultivo de papa (S. tuberosum L.). La colecta de muestras se realizó en fincas de la reserva natural Miraflor y Tisey del departamento de Estelí. La bacteria fue aislada a partir de tejido vegetal mediante la prueba de flujo bacteriano y por técnica de dilución a partir de las muestras de suelo. La identificación de biovares se realizó con pruebas individuales de carbohidratos y la caracterización de razas fue mediante la prueba de hipersensibilidad Ralstonia solanacearum en hojas de tabaco. Se identificaron 19 aislados con características de Ralstonia solanacearum que manifestaron reacciones positivas a las pruebas de Oxidasa, Catalasa y KOH 3%, además se caracterizaron por su crecimiento acuoso, colonias grandes, blancas y centro de color rosado en el medio agar cloruro de tetrazolium (TZC 1%), con la prueba individual de azúcares se identificaron 14 aislados de R. solanacearum, como biovar 1, y los cinco restantes como biovar 3. Las reacciones en hojas de tabaco después de la infiltración de la bacteria mostraron que los 19 aislados de R. solanacearum pertenecen a la raza 1. Se identificaron 12 aislados de R. solanacearum con mecanismo de resistencia AmpC hiperproducido y siete aislados considerados como salvajes.
En Nicaragua sistemas de producción pecuaria se ven afectados por la baja disponibilidad de alimento y agua en la época seca, ocasionando el aumento de enfermedades y desnutrición, afectando directamente los parámetros productivos y reproductivos del hato. De igual forma para contrarrestar este tipo de situaciones entra el uso de antibióticos de uso profi láctico, terapéuticos y promotores de crecimientos capases de eliminar patógenos y actuar de manera drástica sobre la fl ora microbiana necesaria para el funcionamiento del aparato digestivo además de infl uir en la acumulación de residuos en los productos fi nales de importancia para los consumidores. El género lactobacillus representa un alto potencial biotecnológico, estas bacterias contribuyen al desarrollo de características organolépticas de los alimentos y generan un ambiente poco favorable para el desarrollo de microorganismos patógenos. El objetivo de este trabajo fue identifi car mediantes pruebas in vitro las especies de Lactobacillus con potencial probiotico procedentes de sustrato fermentado de yuca. Mediante las pruebas bioquímicas se identifi caron dos lactobacillus sp, L. casei y L. Brevis. Las especies de Lactobacillus previamente identifi cadas fueron sometidas la prueba de resistencia a diferentes niveles de pH, mostrando resistencia a las condiciones acidas. La prueba de antagonismo microbiano frente E. Coli ATCC 25922 y Salmonella sp, demostraron presencia de halos de inhibición, indicando acción antimicrobiana sobre el crecimiento de los patógenos. Las especies de L. casei y L. Brevis fueron sensibles a los antibióticos Trimetropim Sulfametaxona y oxitetraciclina al realizar la prueba de sensibilidad. Estos resultados demuestran que las cepas identifi cadas pueden ser utilizadas como probiótico en la alimentación de los animales o como inoculante en el proceso de ensilaje.
La producción de café está limitada por plagas que reducen las cosechas y disminuyen su calidad. Los objetivos de esta investigación fueron la identificación de bacterias fitopatógenas, descripción de síntomas en campo y sensibilidad in vitro a bactericidas de uso agrícola. Se seleccionaron cuatro fincas cafetaleras en el departamento de Jinotega y una en el departamento Matagalpa, con antecedentes de enfermedades bacterianas; las muestras de tejido colectadas consistieron en hojas, bandolas, frutos, y flores. La identificación de géneros de bacterias fitopatógenas se realizó a partir de características morfológicas de crecimiento bacteriano y presencia de pigmentos fluorescentes en King B. La identificación de especies de bacteria se realizó con prueba de oxidasa y pruebas de producción de ácidos a partir de carbohidratos. La patogenicidad de las bacterias se evaluó mediante la prueba de hipersensibilidad en plantas de tabaco (Nicotiana tabacum L.) como planta no huésped. La sensibilidad e inhibición de las bacterias a bactericidas se determinó mediante el método de Bauer-Kirby (método de difusión en agar) y fue evaluado en diámetros (mm). Los bactericidas evaluados fueron, clorhidrato de oxitetraciclina 5 WP, estreptomicina más oxitetraciclina 16. 5 WP, ácido oxolínico 20 WP y sulfato de cobre 24 SC. Las especies de bacterias identificadas fueron Pseudomonas syringae, Pseudomonas corrugata y Pseudomonas cichorii. En esta investigación Pseudomonas cichorii fue la más frecuente en los diferentes aislados, Pseudomonas syringae, fue aislada en todas las variedades de café muestreadas. Las especies de bacterias aisladas resultaron positivas a la prueba de hipersensibilidad, indicando ser patógenas para el cultivo de café. Los síntomas en hojas, bandolas y flores causados por Pseudomonas spp son manchas de coloración pardo-oscura, de forma y tamaño irregular, rodeada, generalmente por halos amarillentos y traslucidos. El bactericida que mostró mejor efecto al mostrar mayores diámetros en los halos de inhibición bacteriano fue el ácido oxolínico.
El presente trabajo se realizó en el ganado criollo de la raza Reyna de la facultad ciencia animal. Esta raza ha sido importante en el desarrollo de la ganadería centroamericana, en principio constituyó el hato fundador de la misma. El ganado Reyna, es una raza adaptada a las condiciones tropicales de Nicaragua, seleccionada por Joaquín Reyna en el año de 1920. La proteína κ-caseína está relacionada con características de producción y calidad de la leche, tales como, rendimiento en queso, tiempo de coagulación, firmeza de micela y niveles altos de lactoferrina. La leche derivada de animales con genotipo CASκ AA tiene menor porcentaje de κ-caseína, por el contrario, la leche de animales CASκ BB presenta mayor proporción de κ-caseína y micelas más pequeñas. El presente estudio tuvo como objetivo determinar las frecuencias genotípicas y alélicas del gen kappa-caseína (κ-CN) en ganado de la raza Reyna. Se recolectaron 22 muestras de sangre en hembras reproductoras de la finca Santa Rosa de la Facultad Ciencia Animal de la Universidad Nacional Agraria (UNA). El gen de la kappa caseína fue amplificado mediante la técnica PCR, con los cebadores BLKC-For 5`-ATTAGCCCATTTCGCCTTCT-3` y BLKC-Rev 5`-ATT TATGGCCATTCCACCAA-3`, obteniendo un fragmento de 351 pb. La identificación de los alelos fue realizada mediante la técnica PCR-FRLP, obteniendo frecuencias genotípicas de 0.090, 0.045 y 0.863 para los genotipos AA, BB y AB, respectivamente. La frecuencia alélica fue de 0.52 y 0.48 para los alelos A y B respectivamente, indicando mayor número de genotipos AB y AA.
En el cultivo de papa se han registrado diversos problemas fitosanitarios importantes por la presencia de virus, bacterias y hongos. En los últimos años, productores de los departamentos de Estelí y Jinotega han reportado plantas con síntomas de la enfermedad Zebra chip ocasionada por la bacteria fastidiosa Candidatus liberibacter solanacearum. La enfermedad Zebra chip ha causado grandes pérdidas económicas a los productores de papa del país donde la producción se desarrolla todo el año. Ante esta problemática el Instituto Nicaragüense de tecnología Agropecuaria (INTA) junto con la Universidad Nacional Agraria (UNA) realizó un estudio comprendido entre los meses de agosto 2015 a Julio 2016 con el objetivo de generar información relacionada a la presencia y distribución de Ca. L. solanacearum en los departamentos mencionados. Se colectaron 162 muestras de tejidos con síntomas de la enfermedad en cuatro localidades de Jinotega y tres localidades de Estelí durante los tres ciclos de siembra del cultivo. Las muestras de tejido fueron procesadas y analizadas en el Centro Nacional de Investigación Agropecuaria y Biotecnología (CNIA-INTA) para detectar el agente causal Ca.l. solanacearum utilizando los cebadores moleculares ligados a la bacteria Lso Tx 16/23 y al gen de limpieza Lso adk. 50 muestras resultaron positivas con el cebador Lso Tx 16/23 indicando alta especifi cidad para la detección de este patógeno, mientras que con los cebadores Lso ADK 42 muestras fueron positivas presentando polimorfi smos. Se detectó la bacteria en las zonas de El Tayacán y El Zacatón en el departamento de Estelí y en el departamento de Jinotega El Mojón, La Palmera y San Gregorio. El mayor porcentaje de afectación por Ca. L. solanacearum se presentó en Jinotega con un 52.17% en comparación con Estelí que presento un 22.41% de las muestras colectadas.
