This paper presents the Computer Model of the System for Automatic Processing and Analysis of MRI/fMRI tomography data, obtained at the Kurchatov Institute Resource Center “Cognimed”. The System is based on the “Digital Lab” IT-Platform, involving the Kurchatov Institute Supercomputer Cluster HPC4, which allows speeding up the processing of data for groups (2–350 subjects) by parallelization of computations on the supercomputer nodes (1 subject – 1 node). The proposed System allows scientists to remotely use the installed on the supercomputer specialized software to process and analyze MRI/fMRI data; organizes a unified data storage; permits the work with data by web a interface. The System also enables the use of program modules developed by KI researchers which implement mathematical methods to improve data analysis results. As an example of the realization of this Computer Model, the Module “MRI FS” is presented that provides automatic processing and analysis of MRI data using the open specialized software FreeSurfer v.6.0.
Пациенты с эпилепсией вынуждены хронически применять антиэпилептические препараты (АЭП). Ранее нами было показано увеличение эффективности препарата карбамазепина (КБЗ), включенного в наночастицы PLGA (КБЗ-НЧ) в 30 раз, по сравнению со свободной формой (КБЗ). Хроническое применение КБЗ может оказать влияние на поведение. Для КБЗ показаны нормотимические эффекты, а также снижение двигательной активности и тревожности. Изменение поведения при хроническом приеме КБЗ-НЧ в данном исследовании изучили впервые. Эксперимент проводили на крысах Wistar. Животным из 5 групп (n = 60) в течение 28 дней в хвостовую вену вводили следующие препараты: 1) КБЗ: карбамазепин (раствор в 10% Poloxamer 188) 30 мг/кг; 2) КБЗ-НЧ: КБЗ, включенный наночастицы PLGA (суспензия в 1% Poloxamer 188) 1 мг/кг; 3) ненагруженные наночастицы PLGA (плацебо) в 1% Poloxamer 188. Животные в контрольных группах получали физраствор или 10% Poloxamer 188. Поведение исследовали в тестах «открытое поле» (ОП), «приподнятый крестообразный лабиринт» (ПКЛ) и «Лабиринт Барнс».Животные в группах, получавших плацебо, раствор Poloxamer 188 и физраствор, не отличались от контроля ни по одному из исследованных параметров. В тестах ОП и ПКЛ в группе КБЗ отличий от контроля выявлено не было. Крысы из группы КБЗ-НЧ в тесте ОП показали увеличение двигательной и исследовательской активности, а также снижение тревожности животных. В тесте ПКЛ эта же группа показала увеличение времени нахождения в светлых рукавах лабиринта, что также свидетельствует о снижении тревожности. В тесте «Лабиринт Барнс» животные из группы КБЗ-НЧ в процессе обучения дольше оставались на поверхности лабиринта, что связано с их пониженной тревожностью. Однако, они совершали меньше ошибок в финальном тестировании. Животные из группы КБЗ в процессе обучения не отличались от контроля, однако, в финальном тестировании больше времени тратили на поиск убежища.Таким образом, мы показали, что КБЗ снижает обучаемость животных, в то время как КБЗ-НЧ не только повышает обучаемость, но и снижает тревожность, а также увеличивает двигательную и исследовательскую активность. Вероятно, такие эффекты связаны со способностью наночастиц изменять биораспределение препаратов в головном мозге.
Nuclear functional magnetic resonance imaging (fMRI) is one of the most popular methods for studying the functional activity of the human brain. In particular, this method is used in medicine to obtain information about the state of the functional networks of the patient’s brain. However, the process of processing and analysis of experimental fMRI data is complex and requires the selection of the correct technique, depending on the specific task. Practice has shown that different processing methods can give slightly different results for the same set of fMRI data. There are a number of alternative specialized software packages for processing and analysis, but the methodology still needs improvement and development. We are working in this direction: we analyze the effectiveness of existing methods; we develop our own methods; we create software services for processing and analysis of fMRI data on the basis of the distributed modular platform “Digital Laboratory”, with the involvement of the supercomputer NRC “Kurchatov Institute”. For research we use experimental fMRI data obtained on the scanner Siemens Verio Magnetom 3T at the Kurchatov Institute. One of our tasks within the framework of this project is to improve the technology for studying large-scale functional areas of the cerebral cortex at rest. To build a hierarchical model of interaction of large-scale neural networks, a verified binding of functional areas to anatomy is required. Today, there are a number of generally accepted atlases of the functional areas of the human cerebral cortex, which, nevertheless, are constantly being finalized and refined. This article presents the results of our study of the Glasser atlas for the consistency of voxels within one region and the connectivity metrics of voxel dynamics.
