Schistosomiasis affects million people and its control is widely dependent on a single drug, praziquantel. Computational chemistry has led to the development of new tools that predict molecular properties related to pharmacological potential. We conducted a theoretical study of the imizadole alkaloids of Pilocarpus microphyllus (Rutaceae) with schistosomicidal properties. The molecules of epiisopiloturine, epiisopilosine, isopilosine, pilosine, and macaubine were evaluated using theory models (B3lyp/SDD, B3lyp/6-31+G(d,p), B3lyp/6-311++G(d,p)). Absorption, distribution, metabolization, excretion, and toxicity (ADMET) predictions were used to determine the pharmacokinetic and pharmacodynamic properties of the alkaloids. After optimization, the molecules were submitted to molecular docking calculations with the purine nucleoside phosphorylase, thioredoxin glutathione reductase, methylthioadenosine phosphorylase, arginase, uridine phosphorylase, Cathepsin B1 and histone deacetylase 8 enzymes, which are possible targets of Schistosoma mansoni. The results showed that B3lyp/6-311++G(d,p) was the optimal model to describe the properties studied. Thermodynamic analysis showed that epiisopiloturine and epiisopilosine were the most stable isomers; however, the epiisopilosine ligand achieved a superior interaction with the enzymes studied in the molecular docking experiments, which corroborated the results of previous experimental studies on schistosomiasis.

The gums Sterculia urens, (caraia), Sterculia striata (chicha) or Anadenanthera macrocarpa Benth were assembled in conjunction with poly(o-methoxyaniline) (POMA) in the form of layered nanostructured films using the layer-bylayer (LbL) technique. All the LbL films displayed a well-defined electroactivity, as confirmed via cyclic voltammetry. In comparison to POMA LbL films fabricated with conventional polyelectrolytes (viz. poly(vinyl sulfonic acid)-PVS), the presence of the gums in the LbL films increased remarkably the electrochemical stability of the films.Keywords: natural gums; poly-o-methoxyaniline; layer-by-layer films. INTRODUÇÃOPolissacarídeos naturais têm sido amplamente investigados nos últimos anos em relação às suas características físico-químicas e aplicações. Algumas de suas propriedades são biodegrabilidade, abundância na natureza e versatilidade de aplicações em engenharia, biotecnologia e medicina, além de serem geralmente atóxicos. A formação de filmes finos de polissacarídeos sobre superfícies sólidas pode gerar nova aplicação em sensores ou biossensores. Filmes de quitosana 1 , galactomananas 2 e de carboximetilcelulose 3 vêm sendo usados para fins variados. As gomas naturais são polissacarídeos obtidos de exsudatos de tronco de árvore, sementes, algas ou por fermentação microbiológica. Esses materiais podem ser hidratados em água fria ou quente formando dispersões coloidais, soluções altamente viscosas ou até mesmo géis. Os exsudatos são obtidos do tronco de árvores como resposta do mecanismo de proteção da planta 4,5 . A liberação do exsudato ocorre de forma espontânea ou exploratória, como é o caso do látex da seringueira. Condições climáticas desfavoráveis e solos pobres também estimulam esta produção 4,5 . No entanto, a formação de exsudato pode também ser causada pela indução de agentes químicos, como o óxido de etileno e derivados de ácido benzóico.A árvore angico (Anadenanthera macrocarpa Benth) pertence à família das leguminosas e exsuda uma goma com as mesmas características das gomas arábica (Acacia senegal) e do cajueiro (Anacardium occidentale). A composição da goma do angico, um heteropolissacarídeo, contém arabinose (67,8%), galactose (24,1%), ácido urônico (5,9%) e traços de ramnose, além de apresentar baixa viscosidade, semelhante às gomas do cajueiro e arábica 6,7 . A goma extraída da árvore do chichá (Strerculia striata) é encontrada no norte e no nordeste do Brasil. A endometriose é uma patologia que afeta milhares de mulheres no mundo, isso se deve à presença de tecido endometrial extrauterino, que responde à estimulação hormonal e pode causar uma reação inflamatória resultando em sintomas como dor pélvica crônica severa e infertilidade. No Brasil, cerca de 7 milhões de mulheres sofrem com a doença, cujo diagnóstico, prontuário e pesquisas são altamente deficientes, o que dificulta a recuperação das pacientes. Desse modo, este estudo teve como objetivo investigar as dificuldades no diagnóstico precoce da endometriose e sua relação com a infertilidade feminina por meio de uma revisão narrativa. Foram realizadas buscas bibliográficas nas seguintes bases de dados: SciELO, PubMed/MEDLINE e BVS, por se tratarem de bases de dados que concentram publicações de excelência voltadas para a área da saúde. A seleção dos estudos publicados nas bases de dados foi compreendida entre os anos de 2001 a 2021, utilizando os descritores reconhecidos pelos DECs: “Endometriosis”, “Early Diagnosis”, “Infertility Female” e “Treatmet Alternatives”. Assim, obtivemos resultados de estudos que abordam aspectos envolvidos na dificuldade de se obter o diagnóstico precoce da endometriose, além dos tratamentos disponíveis. Evidenciamos que a endometriose ainda é uma doença de etiopatogenia incerta, e que essa incerteza contribui para a dificuldade de diagnóstico da doença, na qual a falta de informações se destaca como um dos principais fatores que dificultam o diagnóstico precoce da endometriose.
Leishmaniasis is an infectious disease that affects both animals and humans, caused by flagellated parasites belonging to the genus Leishmania. The disease is estimated to reach about 700,000 to 1 million people, causing the deaths of 20 to 30,000 individuals annually. Thus, the present study aims to perform molecular docking tests and evaluation of antileishmania activity in vitro of a ruthenium complex with epiisopiloturine and nitric oxide. AutoDockTools-1.5.6 software was used to perform molecular docking tests. Molecular targets were considered rigid, and Epiruno 2 considered flexible. The genetic algorithm Lamarckian (AGL) with global search and pseudo-Solis and Wets with local search were the methods adopted in the docking. The most promising results of molecular interaction were achieved in the targets Pteridine reductase and UDP-glucose Pyrophosphorylase with rates of −10.68 Kcal•mol −1 and −10.51 Kcal•mol −1 , respectively. A series of drugs was investigated to determine structural, electronic and pharmacological properties, as well as the molecular affinity for the main protease of severe acute respiratory syndrome coronavirus 2 (SARS‑CoV‑2). The drugs were submitted to density functional theory calculations to optimize structures and predict binding preferences. The optimized geometries were used in molecular docking simulations. In the docking study, the receiver was considered rigid and the drugs flexible. The Lamarckian genetic algorithm with global search and Pseudo-Solis and Wets with local search were adopted for docking. Absorption, distribution, metabolism, excretion and toxicological properties were obtained from the Pre-ADMET online server. In this series, the antiviral atazanavir showed the potential to inhibit the main protease of SARS‑CoV‑2, based on the free binding energy, inhibition constant, binding interactions and its favorable pharmacological properties. Therefore, we recommend carrying out further studies with in vitro tests and subsequent clinical tests to analyze its effectiveness in the treatment of SARS‑CoV‑2.
