Os anfíbios são suscetíveis à toxicidade, devido à absorção cutânea dos produtos químicos presentes no ambiente. O uso de herbicidas à base de glifosato é comum e pode ser transportado pelas águas superficiais para os locais de reprodução. O glifosato possui propriedades oxidantes e inibe a ação de enzimas-chave na regulação do ciclo celular. O ácido fólico e o selênio possuem propriedades antioxidantes, que podem reduzir o estresse oxidativo gerado por pesticidas. Ovos de rã-touro (n = 47) foram mantidos em aquários com aeração constante, temperatura controlada (25 ± 2 ° C) e com ciclo claro e escuro de 12/12 horas. Os ovos foram divididos em: Grupo Controle (GC, n = 12), Grupo Exposto ao Roundup Original ® (GR, n = 11); grupo tratado com ácido fólico exposto ao Roundup Original ® (GAFR, n = 12) e Grupo tratado com selênio exposto ao Roundup Original ® (GSR n = 12). As larvas foram mantidas em condições experimentais durante os três primeiros dias de desenvolvimento e depois foram realocadas para aquários com água desclorinada. Após 12 dias, os girinos tiveram sua massa e comprimento aferidos, anestesiados por hipotermia e fixados para o preparo histológico. Os animais expostos ao herbicida exibiram diminuição da massa corporal, comprimento e redução no diâmetro dos cardiomiócitos, enquanto os animais tratados com ácido fólico e selênio não apresentaram alterações nas medidas corporais nem nas trabéculas do tecido cardíaco. Os resultados mostram que o glifosato tem efeitos prejudiciais ao desenvolvimento da rãtouro e a suplementação periconcepcional com antioxidantes pode mitigar danos.
Os herbicidas à base de glifosato têm amplo espectro de ação e estão associados a uma série de impactos ambientais. O glifosato é amplamente utilizado em áreas agrícolas e urbanas e é frequentemente encontrado em águas superficiais e no solo. Apresenta efeitos nocivos para o funcionamento ecossistêmico, devido à sua toxicidade para organismos de todos os níveis tróficos, além de casos de bioacumulação relatados. Além disso, a contaminação de recursos hídricos com glifosato põe em perigo a saúde humana. Medidas de mitigação são necessárias para reduzir os impactos do glifosato na saúde ecossistêmica e humana, com controles mais rígidos necessários para prevenir a sua crescente contaminação em águas superficiais e subterrâneas no território brasileiro.
The high incidence of melanoma has received significant attention. Despite advances in early detection and standard treatment options, new strategies that improve therapy with reduced side effects are highly desirable. Several studies have demonstrated the efficiency of doxorubicin (Dox) to treat melanoma, however, side effects limit its clinical use. Drug delivery systems, especially nanostructured ones, are a useful approach to enhance antitumor activity and reduce the toxicity of drugs. Here, we report the use of calcium phosphate nanoparticles functionalized with Dox and hyaluronic acid (N-Dox) to enhance Dox antiproliferative activity. The effects were accessed in A-375 melanoma cells, in which N-Dox significantly decreased IC50 over 48 hours (0.142 ± 0.07) compared to the free drug (0.44 ± 0.25). Treatment triggered DNA damage, increased nuclear area, and senescent phenotype. Furthermore, it did not form colonies after 14 days of incubation preceded by short exposure treatment. These preliminary results indicate that N-Dox hold promise for melanoma treatment, reducing the minimum effective dose and perhaps a reduction in the cost of treatment.
