The aim of investigation was to compare the action of novel 4-thiazolidinone derivaties (compounds les-5054 and les-5055)
The use of nonsteroidal anti-inflammatory drugs (NSAIDs) in combination with being physiologically stressed often occurs in in the course of different pathologies. This situation may result in the alteration of digestive system functioning. The effect of stress brings about changes in the activity of nitric oxide synthase (NOS), arginase, cyclooxygenase (COX) and lipid peroxidation, whereas the use of NSAIDs interrupts the multiple functions of the cell via the inhibition of prostaglandins (PGs) synthesis. Taking into account that NOS and COX-systems are connected in their regulation, the aim of the study was to determine the role played by NOS and lipid peroxidation under conditions of the combined action of NSAIDs and stress. In our study, male rats were used. The NSAIDs (naproxen - a non-selective COX inhibitor, celecoxib - a selective COX-2 blocker, and the compound 2A5DHT (which is the active substance of dual COX, and the lipoxygenase (LOX) inhibitor, darbufelone) were all administered at a dose 10 mg/kg, prior to water restraint stress (WRS). WRS brought about an increase of inducible NOS (iNOS) activity in the intestinal mucosal and muscular membranes, as well as in the pancreas. Because of this, constitutive NOS izoform (cNOS) and arginase activities decreased. Moreover, the MDA concentration increased, indicating the development of oxidative stress. In our work, pretreatment with naproxen, as in the WRS model, engendered a decrease in iNOS activity. What is more, administration of Celecoxib did not change iNOS activity, as compared to WRS alone, and it showed a tendency to reduce lipid peroxidation. In addition, 2A5DHT prior WRS brought about a decrease of iNOS activity, with the subsequent rise of cNOS activity. Of note, MDA concentration decreased in all studied organs, indicating the reduction of lipid peroxidation under the action of the darbufelone active substance.
Вивчали зміни NO-синтазної системи та стан макрофлори щурів за умов поєднаної дії водно-іммобілізаційного стресу та блокування циклооксигенази. Показано, що на тлі стресу різко зростала активність індуцибельної NO-синтази, підвищувалась інтенсивність ліпопероксидації у тонкій та товстій кишці та змінювалась мікрофлора: кількість ешерихій зростала, а ентерококів знижувалась у тонкій кишці та підвищувалась у товстій кишці. Блокування циклооксигенази напроксеном на тлі стресу супроводжувалося зниженням активності індуцибельної NO-синтази у тонкій та товстій кишці, порівняно з одночасним активуванням конститутивної NO-синтази в товстій кишці. При цьому було виявлено неістотне зростання числа ентерококів у дванадцятипалій кишці, різке зменшення кількості ешерихій у клубовій кишці, помірне зниження їх у проксимальній частині товстої кишки та зростання у дистальній. Дисбіоз, активація процесів ліпопероксидації та зміни показників NO-синтазної системи за умов поєднаної дії стресу та блокування циклооксигенази можуть створювати передумови для розвитку деструктивних змін, що лежать у основі ентеропатій.
Offered all-around different from known scientific by novelty, set, sequence, and modification of tests. It is completely safe for health, because it is gentle and therefore can be used: a) with the purpose of medical supervision in training of physical education to assess the adequacy and prevention the excessive loads of individuals' different level of physical health, which in medical examination related to primary, preparatory or special medical groups; b) in the course of pedagogical supervision as a physical education teacher or coach-teacher for correction training loads; c) for self-monitoring during the training process, including to assess the effectiveness of physical training; d) in the form of competition "Physiological all-round" to determine the winners in the academic departments, formed for physical education. With the aim of improving the motor abilities the students, in addition to the main training in physical education, were offered rehabilitation complex, which includes special exercises for eyes, posture and feet, breathing exercises with volitional control of breathing, self massage of the eye, as well as aerobic exercises for self-study task in the home.Exercises that are offered, is carried out according to individual motor mode: No.1 (limited action), No.2 (moderate action) or No.3 (extended action). It is chosen taking into account the physical condition. Those under study in the main groups were received a memo, developed by Klapchuk and Maryukhnich, which had recommendations for self-control. Pupils chose exercises, starting with simple, gradually upgrading and replacing them periodically with a view to the execution of the complex was at least 20-30 minutes 4-5 times a week. Along with this, in the second main group conducted individual training acomodating muscles, including reserves of accommodation.Under the influence of rehabilitation course the statistically significant positive dynamic was held by all the studied indicators of motor abilities and in a comprehensive assessment of the effectiveness of physical training. The results of the research make reason to recommend a designed program for more physical training and exercises for self-study lessons at home until the introduction in practice of physical rehabilitation and physical education. Further in this group of pupils with impaired vision, it is advisable to analyze the changes of reserves accommodation under the influence of rehabilitation course, using one eye. Doing this, there should be conditions for full correction for distance, and tests need to be shown to further correspond to the maximum visual acuity.Key words: pupils, impaired vision, physical capabilities, rehabilitation complex.
