ﻨﻔﺫﺕ ﻫﺫﻩ الدراسة بهدف عزل و تشخيص ثلاث عزلات تابعة للفطر Pythium aphanidermatum المسبب لمرض تعفن بذور و موت بادرات الخيار (Cucumis sativus L.) جمعت من مناطق العباسية و أم عباسيات و القزوينية في محافظة النجف. أظهرت نتائج اختبار القدرة الامراضية لهذه العزلات الفطر P. aphanidermatum على بذور نبات الخيار ان جميع العزلات ادت الى زيادة تعفن البذور و كان أكثرها تأثيرا هي عزلة P. aphanidermatum المعزولة من منطقة العباسية, تلتها العزلة P. aphanidermatum المعزولة من منطقة أم عباسيات و من ثم العزلة المعزولة من منطقة القزوينية و بنسب تعفن بذور بلغت 90 و 74 و 72 %, على التوالي قياسا مع معاملة المقارنة التي بلغت فيها نسبة التعفن 0 %. كما اثبتت نتائج اختبار تفاعل البلمرة المتسلسل (Polymerase chain reaction, PCR) لعزلتي الفطر P. aphanidermatum الاولى و الثانية و التي كانت الاكثر امراضية لبذور نبات الخيار, امكانية مضاعفة نواتج من الحامض النووي (PCR-amplified products) بأحجام تراوحت بين 700-800 زوج قاعدة نايتروجينية (Base pairs, bp). اظهرت نتائج تحليل التتابع النيوكلوتيدي (Nucleotide sequence analysis) لنواتج الحوامض النووية المضاعفة من العزلات الفطرية و باستخدام برنامج BLAST بأن عزلتي الفطر المعزول تعود الى الفطر P. aphanidermatum. كما اظهرت احدى العزلات نسبة تشابه بلغت 100% مع العزلات المعزولة عالميا و المثبتة في المركز الوطني لمعلومات التقنية الحيوية (National Center for Biotechnology Information, NCBI). وجد من خلال استخدام برنامج BLAST ان عزلة الفطر P. aphanidermatum المعزولة من منطقة العباسية في محافظة النجف ذات نسبة تشابه وراثي تراوحت بين 99-98 %, مما يثبت بأن هذه العزلة هي عزلة جديدة غير مذكورة سابقا في قاعدة البيانات العائدة للمركز الوطني لمعلومات التقنية الحيوية (NCBI), لذا تم تسجيلها و ايداعها في المركز المذكور تحت رقم الادخال (GenBank Accession No.) MF347709.
