Purpose: Oncolytic adenoviruses are attractive therapeutics for cancer because they selectively replicate in tumors. However, targeting tumor metastasis remains a major challenge for current virotherapy for cancer. Oct-3/4 is specifically expressed in embryonic stem cells and tumor cells. Oct-3/4 highly expressed in cancer cells may be a potential target for cancer therapy. We developed an E1B-55 kDa–deleted adenovirus, designated Ad.9OC, driven by nine copies of Oct-3/4 response element for treating Oct-3/4–expressing metastatic bladder cancer.
Experimental Design: We examined the expression of Oct-3/4 in human bladder tumor tissues and bladder cancer cell lines. We also evaluated the cytolytic and antitumor effects of Ad.9OC on bladder cancer cells in vitro and in vivo.
Results: Oct-3/4 expression was detected in bladder cancer cell lines, as well as in human bladder tumor tissues. Notably, Oct-3/4 expression was higher in metastatic compared with nonmetastatic bladder cancer cells. Ad.9OC induced higher cytolytic activity in metastatic bladder cancer cells than in their nonmetastatic counterparts, whereas it did not cause cytotoxicity in normal cells. Pharmacologic and short hairpin RNA–mediated Oct-3/4 inhibition rendered bladder cancer cells more resistant to Ad.9OC-induced cytolysis. Replication of Ad.9OC was detected in murine bladder cancer cells and bladder tumor tissues. We also showed the effectiveness of Ad.9OC for treating bladder cancer in subcutaneous, as well as metastatic, bladder tumor models.
Conclusions: Ad.9OC may have therapeutic potential for treating Oct-3/4–expressing tumors. Especially, metastatic bladder tumors are good target for Ad.9OC treatment. Because Oct-3/4 is expressed in a broad spectrum of cancers, Ad.9OC may be broadly applicable.
We used the tropical oceanic planktonic diatom species Nitzschia marina, Rhizosolenia bergonii and Azpeitia africana/Azpeitia neocrenulata, most commonly found in the surface sediments of the northeasternmost South China Sea (SCS) where the Kuroshio Current enters the SCS through the Bashi Strait north of Luzon, to analyse the influence of the the Kuroshio Current on the SCS. The impact of the Kuroshio Current seems to be relatively strong during major warm periods and, to a lesser degree, in minor warm periods during the last 115 000 years. The strongest influence took place during the main part of the Holocene and during the very late phase of Marine Isotope Stage (MIS) 5e. It is possible to distinguish two magnitudes of change in the impact of the Kuroshio Current on the SCS: large changes occurred at shifts between glacial and interglacial conditions, while smaller changes seem to have recurred in both glacial and interglacial episodes as well as during the Holocene. Climatic/oceanographic changes and sea-level variations were possibly the two most important mechanisms for the varying influences of the Kuroshio Current on the SCS. The interaction between the Kuroshio Current and monsoon-related processes may also have played a role. However, because of restricted knowledge of the present-day Kuroshio Current and the absence of a modern analogue to the ancient SCS due to the marked changes in palaeogeography, this relationship is difficult to establish.
Tillage systems can influence C sequestration by changing aggregate formation and C distribution within the aggregate. This study was undertaken to explore the impact of no-tillage without straw (NT-S) and with straw (NT+S), and moldboard plow without straw (MP-S) and with straw (MP+S), on soil aggregation and aggregate-associated C after six years of double rice planting in a Hydragric Anthrosol in Guangxi, southwest of China. Soil samples of 0.00-0.05, 0.05-0.20 and 0.20-0.30 m layers were wet-sieved and divided into four aggregate-size classes, >2 mm, 2.00-0.25 mm, 0.25-0.053 and <0.053 mm, respectively, for measuring aggregate associated C and humic and fulvic acids. Results showed that the soil organic carbon (SOC) stock in bulk soil was 40.2-51.1 % higher in the 0.00-0.05 m layer and 11.3-17.0 % lower in the 0.05-0.20 m layer in NT system (NT+S and NT-S) compared to the MP system (MP+S and MP-S), respectively. However, no statistical difference was found across the whole 0.00-0.30 m layer. The NT system increased the proportion of >2 mm aggregate fraction and reduced the proportion of <0.053 mm aggregates in both 0.00-0.05 and 0.05-0.20 m layers. The SOC concentration, SOC stock and humic and fulvic acids within the >0.25 mm macroaggregate fraction also significantly increased in the 0.00-0.5 m layer in NT system. However, those within the 2.00-0.25 mm aggregate fraction were significantly reduced in the 0.05-0.200 m layer under NT system. Straw incorporation increased not only the SOC stock in bulk soil, but also the proportion of macroaggregate, aggregate associated with SOC and humic and fulvic acids concentration within the aggregate. The effect of straw on C sequestration might be dependent on the location of straw incorporation. In conclusion, the NT system increased the total SOC accumulation and humic and fulvic acids within macroaggregates, thus contributing to C sequestration in the 0.00-0.05 m layer.
Titanium dioxide (TiO2) nanoparticles were modified onto fluorine-doped tin oxide (FTO) via dip-coating technique with different nanoparticle sizes, lifting speeds, precursor concentrations, and dipping numbers. Electrodeposition-based electrochromic device with reversible three-state optical transformation (transparent, mirror, and black) was fabricated subsequently by sandwiching a suitable amount of gel electrolyte between modified FTO electrode and flat FTO electrode. Correlation between dip-coating process engineering, morphological features of TiO2 thin films, i.e., thickness and roughness, as well as performance of electrochromic devices, i.e., optical contrast, switching time, and cycling stability, were investigated. The modified device exhibits high optical contrast of 57%, the short coloration/bleaching switching time of 6 and 20 s, and excellent cycling stability after 1500 cycles of only 27% decrement rate by adjusting dip-coating processes engineering.
