Durch iibersichtlichen Abbau der nach Lysin-[2-14C]-Applikation ausFriihere Untersuchungen 2,3) zeigten, daI3 Lupanin (11) und Hydroxylupanin (V) von Lupinus angustifolius spezifisch aus 3 Moll. Cadaverin gebildet werden konnen. Bisher ist jedoch Cadaverin in Lupinen nicht nachgewiesen worden. Daher war zu priifen, ob es tatsachlich dievorstufe der Lupinen-Alkaloide PI undV darstellt, oder ob es nur eine mogliche Vorstufe ist. Durch Applikation von Lysin-[2-14C] an L. angustifolius und chemischen Abbau der erhaltenen, radioaktiv markierten Alkaloide sollte entschieden werden, ob das Cadaverin intermediar auftritt, oder ob die Aminosaure, ohne dieses Diamin zu bilden, auf anderen Wegen in die Alkaloide iibergeht.
Durch Verfütterung von Cadaverin‐[1.5‐14C] und Lysin‐[2‐14C] an Lupinus lureus (gelbe Lupine) und Sarothamnus scoparius (Besenginster) wird radioaktiv markiertes Spartein (V) gewonnen. Chemische Abbauversuche von V zeigen, daß Lysin‐[2‐14C] spezifisch in das Alkaloid eingebaut wird. Es ergibt sich eine Radioaktivitätsverteilung, die derjenigen nach Cadaverin‐[I.5‐14C]‐Fütterung entspricht. Nach Applikation von Lysin‐[2‐14C, α‐15N] an die untersuchten Pflanzen enthält das isolierte Spartein einen Überschuß an 15N. Das Verhältnis der spezif. 14C‐Aktivität zum 15N‐Überschuß zeigt, daß Lysin ¨︁ber die Stufe des Cadaverins in das Alkaloid eingebaut wird.
Chromsaureoxydation von Hydroxylupanin: a) 307 mg V wurden in 0.4 ccm Wasser mit der Losung von 2.02 g CrOj in 1.5 ccm konz. Schwefelsaure + 4.5 ccm Wasser 72 Stdn. im siedenden Wasserbad oxydiert. Die Atherextraktion lieferte keine Bernsteinsaure. Aus der schwefelsauren Losung konnten P-Alanin und wenig Glycin chromatographisch abgetrennt werden. b) 225 mg V wurden mit 1.44 g CrO3 in 1.1 ccm konz. Schwefelsaure -t 3.6 ccm Wasser in 24 Stdn. bei 100" (siedendes Wasserbad) oxydiert. Die unverbrauchte Chromsaure wurde durch Einleiten von SO2 reduziert und die Losung anschlieRend mit Ather extrahiert. Aus dem Atherauszug wurden 14 mg Bernsteinsiiure (VII) durch Hochvakuumsublimation isoliert ; Schmp.
