BackgroundTrehalase, an enzyme that hydrolyzes trehalose to yield two glucose molecules, plays a pivotal role in various physiological processes. In recent years, trehalase proteins have been purified from several insect species and are divided into soluble (Tre-1) and membrane-bound (Tre-2) trehalases. However, no functions of the two trehalases in chitin biosynthesis in insects have yet been reported.Principal FindingsThe membrane-bound trehalase of Spodoptera exigua (SeTre-2) was characterized in our laboratory previously. In this study, we cloned the soluble trehalase gene (SeTre-1) and investigated the tissue distribution and developmental expression pattern of the two trehalase genes. SeTre-1 was expressed highly in cuticle and Malpighian tubules, while SeTre-2 was expressed in tracheae and fat body. In the midgut, the two trehalase genes were expressed in different locations. Additionally, the expression profiles of both trehalase mRNAs and their enzyme activities suggest that they may play different roles in chitin biosynthesis. The RNA interference (RNAi) of either SeTre-1 or SeTre-2 was gene-specific and effective, with efficiency rates up to 83% at 72 h post injection. After RNAi of SeTre-1 and SeTre-2, significant higher mortality rates were observed during the larva-pupa stage and pupa-adult stage, and the lethal phenotypes were classified and analyzed. Additionally, the change trends of concentration of trehalose and glucose appeared reciprocally in RNAi-mutants. Moreover, knockdown of SeTre-1 gene largely inhibited the expression of chitin synthase gene A (CHSA) and reduced the chitin content in the cuticle to two-thirds relative to the control insects. The chitin synthase gene B (CHSB) expression, however, was inhibited more by the injection of dsRNA for SeTre-2, and the chitin content in the midgut decreased by about 25%.Conclusions
SeTre-1 plays a major role in CHSA expression and chitin synthesis in the cuticle, and SeTre-2 has an important role in CHSB expression and chitin synthesis in the midgut.
The way a dendrite is patterned determines how a neuron will receive information. The Rho GTPases have been reported to play increasingly well defined roles in determining dendritic branch and spine development and morphology. Much is known about how these small GTPases regulate the actin cytoskeleton; however, very little is known about how they alter the microtubule cytoskeleton. Our laboratory previously cloned and characterized cypin, a guanine deaminase that increases dendrite number by binding to tubulin heterodimers and promoting microtubule assembly. In the present study, we show that cypin and RhoA are part of a common pathway that regulates dendrite number. Inhibition of Rho kinase activity does not affect cypin-mediated dendrite branching. Furthermore, cypin does not affect the activity of RhoA, as measured by GTP binding to RhoA. In fact, activated RhoA acts to inhibit cypin protein expression and, by doing so, decreases dendrite number. In addition, this decrease in cypin protein occurs via a translation-dependent mechanism. Together, our data suggest that cypin acts downstream of the small GTPase RhoA to regulate dendrite branching in hippocampal neurons, providing a novel mechanism for RhoA action on microtubule dynamics.
Abstract. Debris flow material properties change during the initiation, transportation and deposition processes, which influences the runout characteristics of the debris flow. A quasi-three-dimensional depth-integrated numerical model, EDDA (Erosion–Deposition Debris flow Analysis), is presented in this paper to simulate debris flow erosion, deposition and induced material property changes. The model considers changes in debris flow density, yield stress and dynamic viscosity during the flow process. The yield stress of the debris flow mixture determined at limit equilibrium using the Mohr–Coulomb equation is applicable to clear water flow, hyper-concentrated flow and fully developed debris flow. To assure numerical stability and computational efficiency at the same time, an adaptive time stepping algorithm is developed to solve the governing differential equations. Four numerical tests are conducted to validate the model. The first two tests involve a one-dimensional debris flow with constant properties and a two-dimensional dam-break water flow. The last two tests involve erosion and deposition, and the movement of multi-directional debris flows. The changes in debris flow mass and properties due to either erosion or deposition are shown to affect the runout characteristics significantly. The model is also applied to simulate a large-scale debris flow in Xiaojiagou Ravine to test the performance of the model in catchment-scale simulations. The results suggest that the model estimates well the volume, inundated area, and runout distance of the debris flow. The model is intended for use as a module in a real-time debris flow warning system.
A multiwavelength fiber ring laser obtained by use of a semiconductor optical amplifier (SOA) with a simple laser cavity configuration is reported. A Fabry-Perot filter was used in the fiber laser ring cavity to achieve more than 50 simultaneous wavelength lasing oscillations with a frequency separation of 50 GHz. The resulting stable broadband multiwavelength lasing operation was attributed to broadband and flat gain of the SOA, which has a gain flatness of 0.8 dB for more than 20 nm. The laser has a total output power of -3 dBm and a signal-to-spontaneous-noise ratio of 30 dB.
Effective English writing has long been a challenge in English language teaching. With the development of cognitive psychology, metacognition has drawn more and more researchers’ attention and provides a new perspective for EFL writing. Metacognitive theory mainly includes metacognitive knowledge and metacognitive strategy. Among all the learning strategies, metacognitive strategy is a higher-order executive skill which includes planning, monitoring and evaluating. Once learners have a good command of metacognitive strategy, they will become more independent and autonomous and will be more capable of planning, monitoring and evaluating their learning process and thus become efficient learners.
However, the study of metacognitive strategies-based writing instruction for vocational college students has been neglected for the past years. The author, therefore, carried out an empirical study to investigate the effect of metacognitive strategy training on students’ writing performance in the hope of finding an optimal teaching approach for English teachers of vocational colleges. This teaching approach really embodies the teaching idea “student-centered” and is targeted to foster students’ metacognitive strategy, monitoring and evaluating abilities in English writing.
