To study the structural basis of the GM2 gangliosidosis B variant, we constructed the threedimensional structures of the human b-hexosaminidase a-subunit and the heterodimer of the a-and b-subunits, Hex A, by homology modeling. The a-subunit is composed of two domains, domains I and II. Nine mutant models due to specific missense mutations were constructed as well and compared with the wild type to determine structural defects. These nine mutations were divided into five groups according to structural defects. R178H is deduced to affect the active site directly, because R178 is important for binding to the substrate. C458Y and W420C are predicted to cause drastic structural changes in the barrel structure carrying the active site pocket. R504C/H is deduced to introduce a disruption of an essential binding with D494 in the bsubunit for dimerization. R499C/H, located in an extrahelix, is deduced to disrupt hydrogen bonds with domain I and the barrel. R170W and L484P are deduced to affect the interface between domains I and II, causing destabilization. The structural defects reflect the biochemical abnormalities of the disease.
Aldosterone is an important hormone in the adrenal gland as well as the human body. It is responsible for regulating salt and water in the body, therefore controlling blood pressure. Primary aldosteronism (also known as aldosterone producing while suppressing renin) plays a significant role in the pathophysiology of hypertension. Mutation in the KCNJ5 gene which leads to aldosterone-production adenomas is the main cause of primary aldosteronism. In this study, the entire coding sequence of the KCNJ5 gene including 2 exons was amplified and directly sequenced to detect mutations. The result revealed that two missense mutations were identified in two patients with hypertension and an apparent mass on the adrenal gland. A heterozygous mutation, D223V, is a novel mutation while the homozygous mutation, Q282E, was known as a polymorphism that had a significant effect on viability of the H295R cell line and K+ conduction channel. Analysis of the 3D protein structural model of the mutants clarified the relationship between genotype and phenotype of the two patients. These results partially pointed out causes of the adrenal gland adenomas and endocrine disorders in the the patient.
Ngày nay, tỷ lệ sản phụ mắc rau cài răng lược ngày càng tăng, tuy nhiên, biến chứng do phẫu thuật mổ lấy thai của những sản phụ mắc rau cài răng lược còn ít được quan tâm, một trong số đó có biến chứng hay gặp nhất đó là rách bàng quang. Nghiên cứu biến chứng rách bàng quang ở người bệnh phẫu thuật mổ lấy thai có rau cài răng lược với mục tiêu xác định tỷ lệ rách bàng quang và tìm một số yếu tố liên quan đến rách bàng quang ở người bệnh phẫu thuật mổ lấy thai có rau cài răng lược tại Bệnh viện Phụ sản Hà Nội. Bằng phương pháp mô tả cắt ngang ở 93 người bệnh được phẫu thuật mổ lấy thai đã được chẩn đoán xác định rau cài răng lược. Kết quả: rách bàng quang là 14,0%, có mối liên quan giữa thời gian phẫu thuật, thái độ xử trí tử cung và phân độ rau cài răng lược với tỷ lệ rách bàng quang (p < 0,05).
Nghiên cứu này nhằm đánh giá thực trạng và đề xuất giải pháp nâng cao hiệu quả hoạt động của Chi nhánh Văn phòng Đăng ký đất đai (CNVPĐKĐĐ) thành phố Ninh Bình. Tiến hành điều tra 90 người sử dụng đất (NSDĐ) đến làm việc tại CNVPĐKĐĐ theo phương pháp chọn mẫu ngẫu nhiên. Sử dụng thang đo 5 mức của Likert để đánh giá hoạt động của CNVPĐKĐĐ. Kết quả nghiên cứu cho thấy CNVPĐKĐĐ thành phố Ninh Bình còn thiếu trang thiết bị, máy móc; trụ sở VPĐKĐĐ còn chật; chưa cập nhật hồ sơ thường xuyên. NSDĐ đánh giá hoạt động của CNVPĐKĐĐ ở mức khá cao với 5/9 tiêu chí được đánh giá ở mức rất cao, 4/9 tiêu chí được đánh giá ở mức cao. Cán bộ công chức, viên chức đánh giá hoạt động của CNVPĐKĐĐ ở mức khá cao với 5/8 tiêu chí ở mức rất cao, 3/9 tiêu chí ở mức cao. Để nâng cao hiệu quả hoạt động của CNVPĐKĐĐ, cần thực hiện các giải pháp sau: tăng cường công tác quản lý nhà nước về đất đai, đẩy mạnh ứng dụng công nghệ thông tin và hoàn thiện cơ sở vật chất, nâng cao chất lượng nguồn nhân lực, tăng cường quản lý cập nhật và chỉnh lý hồ sơ địa chính (HSĐC), tăng cường tuyên truyền và phổ biến kiến thức pháp luật.
