Taraxastanon‐(3) (I) reagiert mit Äthylformiat in Gegenwart von Na‐äthanolat unter Stickstoff bei nachfolgender Hydrolyse zum Hydroxymethylenderivat (II), das im Gemisch mit den tautomeren Formen (III) und (IV) vorliegt.
Die aus ′Iv‐Taraxasterol (Ia), Taraxasterol (IIa) bzw. Taraxastanol (IIIa) durch CrO3‐Oxidation erhältlichen Ketone (Ib), (IIb) und (IIIb) werden in ihre Oxime (Ic), (IIc) bzw. (IIIc) übergeführt.
Ausgehend von den epimeren Aminotaraxastanen (Ia) bzw. (IIa) werden die Derivate (Ib) und (IIb) [Umsetzung mit Chlorameisensäure‐äthylester], (Ic) und (IIc) [Li‐alanat‐Reduktion von (Ib) bzw. (IIb)], (Id) und (IId) [nachfolgende Acetylierung], (Ie) und (IIe) [Eschweiler‐Clarke‐Methylierung] sowie (If) und (IIf) [nachfolgende Quaternisierung mit Methyljodid] dargestellt.
