An endogenous clock regulates the temporal expression of genes/mRNAs that are involved in the circadian output pathway. In the green alga Chlamydomonas reinhardtii, a clock-controlled RNA-binding protein (Chlamy 1) was identified recently, which represents an analog of the circadian trans-acting factor CCTR from the phylogenetically diverse alga Gonyaulax polyedra. In order to identify in C. reinhardtii target mRNAs that can be recognized by Chlamy 1, gel mobility-shift assays and UV-crosslinking experiments were carried out, and revealed that this protein interacts specifically with the 3' untranslated regions of several mRNAs and recognizes them all via a common cis-acting element, composed of at least seven UG repeats. By using competition assays, it was found that the affinity of Chlamy 1 is highest for mRNAs whose products are key components of nitrogen and CO2 metabolism. Since the activities of enzymes involved in nitrogen metabolism vary in a temporal pattern that is opposite in phase to that of Chlamy 1 binding activity, the protein may repress the translation of the cognate mRNAs.
Mit dem Fortschreiten der Erkrankung wird in unserem Falle schließlich gerade jenes System, dessen Aktivierung das Virus zunächst bewirkte, auf das schwerste geschädigt, wobei die Defekte in den Zellen, die der Nekrose entgehen, im Cytoplasma wieder besonders auffällig sind. Verlorengegangen ist anscheinend vor allem die Fähigkeit zur Nachbildung von Nucleinsäure, ganz ähnlich wie es auch für die dauergeschädigte motorische Vorderhornzelle bekannt ist. Im ganzen hat die Erkrankung Ähnlichkeit mit den Vorgängen, die sich an Vorderhornzellen während einer Infektion mit Poliomyelitis-"Erregern" abspielen. Beide Virusgruppen besitzen ja enge verwandtschaftliche Beziehungen. Auch bei der Erkrankung der Vorderhornzelle ist der Erkrankungsbeginn durch gesteigerte Aktivität des hier besonders umfangreich ausgebildeten, eiweißproduzierenden Systems ausgezeichnet, während in den Spätstadien wiederum dieses System die schwersten Schäden davonträgt. Res. 11, 1 [1951]. der Poliomyelitisgruppe mithin im Rahmen entsprechender Reaktionen des ZNS. Zur Kennzeichnung des eiweißproduzierenden Systems mag die Vermutung ausgesprochen werden, daß es in seinen cytoplasmatischen Anteilen mit der. von Claude 24 aus Zellhomogenaten isolierten submikroskopischen Granulafraktion identisch ist, über deren Verteilung im Zellplasma allerdings kaum etwas bekannt ist. Ausgezeichnet ist diese Fraktion durch ihren hohen Gehalt an Ribonucleotiden, deren Anteil mit fallender Teilchengröße noch zunimmt. Da Chantrenne 25 gleichzeitig eine Abnahme des Enzymgehaltes findet, werden Angaben über den Wirkungsmechanismus gerade der nucleinsäurereich-sten Einheiten ebenso unsicher wie entsprechende Aussagen über die Wirkung ähnlich großer und ähnlich zusammengesetzter Viruskörper. Es wurden vergleichende morphologische, histochemische Untersuchungen sowie flammeiiphotometrisehe Bestimmungen von Kalium und Natrium an der Muskulatur von saugenden Mäusen und Meerschweinchen durchgeführt, die mit Coxsackie-A-Virus infiziert worden waren. Bei den Säuglingsmäusen wurde aui3er den bekannten destruktiven Skeletmuskelverände-rungen im Verlaufe der Infektion ein Abfall des Muskelkaliums und ein Anstieg des Muskelnatriums festgestellt. Bei den saugenden Meerschweinchen blieben markante morphologische Veränderungen am Skeletmuskelgewebe aus; dagegen fanden sich 2 Tage nach der Virusinfektion ebenfalls signifikante Senkungen der Kaliumwerte und eine Vermehrung des Muskelnatriums.
