ABSTRACT. This study aimed to examine the relationship between skull size and the level of cranial vault suture closure. A total of 50 Myrmecophaga tridactyla Linnaeus, 1758 and 178 Tamandua tetradactyla Linnaeus, 1758 skulls were analyzed in relation to 18 skull dimensions. The skulls were grouped into three levels of suture closure: no sutures closed (level 0), one or all the fallowing sutures closed: interfrontalis, sagitalis and coronalis (level 1) and all sutures closed (level 2). The results indicated that among the 18 variables measured, 17 showed significant differences (p ≤ 0.01) between level 0 and level 1 skulls of T. tetradactyla; as well as between level 0 and level 1, and level 0 and level 2 skulls of M. tridactyla. M. tridactyla level 1 and level 2 had no significant difference among any of the 18 dimensions. The foramen magnum height in both species showed no significant difference (p > 0.05) among any suture categories. In principle, suture closure level and cranial dimensions are related. The specimens with larger cranial dimensions showed greater number of cranial vault sutures closed for both species of anteaters. Tamandua tetradactyla and M. tridactyla specimens with none of the cranial vault suture closed have a foramen magnum height similar to those with cranial vault suture closed.KEYWORDS. Age, anteater, cranial morphology, xenarthra. RESUMO.Morfometria craniana e suturas da abóboda de Myrmecophaga tridactyla e Tamandua tetradactyla. O presente estudo teve como objetivo examinar a relação das dimensões do crânio e o grau de fechamento das suturas da abóboda craniana. Foram analisados 50 crânios de Myrmecophaga tridactyla Linnaeus, 1758 e 178 de Tamandua tetradactyla Linnaeus, 1758 em relação a 18 dimensões do crânio. Os crânios foram divididos em três grupos quanto ao grau de fechamento da sutura: nenhuma sutura fechada (grau 0), uma ou todas das seguintes suturas fechadas: interfrontalis, sagitalis e coronalis (grau 1) e todas as suturas fechadas (grau 2). Os resultados indicaram que das 18 dimensões 17 apresentaram diferença significativa (p ≤ 0,01) entre grau 0 e grau 1 de T. tetradactyla; entre grau 0 e grau 1, e grau 0 e grau 2 de M. tridactyla. Indivíduos grau 1 e grau 2 de M. tridactyla não apresentaram diferença significativa em nenhuma das 18 dimensões. A altura do forame magno nas duas espécies não apresentou diferença significativa (p > 0,05) entre nenhuma das categorias de sutura (grau 0, grau 1 e grau 2). Em princípio, o nível de fechamento da sutura e as dimensões cranianas estão relacionados. Os espécimes com dimensões cranianas maiores apresentaram maior número de suturas de abóbada craniana fechadas para ambas as espécies de tamanduás. Os espécimes de T. tetradactyla e M. tridactyla sem sutura da abóbada craniana fechada possuem altura do forame magno semelhante àquelas com sutura da abóbada craniana fechada.
The aim of this study was to test the use of mechanical and mechanical-enzymatic methods, saline solution (SS), and PBS solution for the manipulation and isolation of mare ovarian preantral follicles (PAFs). The ovaries were subjected to mechanical isolation (mixer) alone or in association with enzymatic digestion (collagenase). Incubation times of 10 and 20 min were employed. In the first group, 4.1 ± 4.9 PAFs were harvested with the mechanical-enzymatic method vs 71.1 ± 19.2 with the mechanical procedure, showing a significant difference between methods; using SS and PBS, these numbers were 35.7 ± 34.3 and 39.6 ± 39.6, respectively, with no significant difference between solutions. In the second group, there was significant difference between methods, with 7.1 ± 10.6 follicles harvested with the mechanical-enzymatic method vs 63.2 ± 22.9 with the mechanical procedure; using SS and PBS, means were 35.5 ± 36.4 and 34.9 ± 31.1, respectively. The mechanical method proved more effective than the mechanical-enzymatic approach. Both SS and PBS can be used as a media for equine PAFs preparation.
Morfometria, células da granulosa e cromatina nuclear de folículos pré-antrais em associação ou não com a vitrificação de tecido ovariano bovino RES UMOBusca-se no presente estudo, avaliar o efeito da vitrificação de fragmentos ovarianos na morfometria, número de células da granulosa e cromatina nuclear de folículos pré-antrais bovinos. Ovários foram obtidos em abatedouro e fragmentos do córtex retirados, enquanto uma parte foi fixada imediatamente e processada para histologia (T 0 -controle). Outros fragmentos foram distribuídos em dois tratamentos: T I -toxicidade e T II -vitrificação; no T I , os fragmentos foram expostos apenas aos crioprotetores e fixados em Carnoy, e no T II , usou-se a associação dos crioprotetores DMSO e etilenoglicol acrescido de sacarose. Os resultados indicaram que folículos primordiais, quando vitrificados ou expostos aos crioprotetores, não apresentam variações morfométricas e na quantidade de células da granulosa (p>0,05), exceção feita aos folículos primários (p<0,05) quando comparados com o T 0 . A análise da morfologia da cromatina nuclear de folículos vitrificados ou apenas expostos aos crioprotetores, mostrou diferenças significativas (p<0,05) quando comparados com o T 0 ; conclui-se, então, que a vitrificação causou alterações morfológicas de folículos bovinos inclusos em tecido ovariano. Ovarian was obtained at a local slaughterhouse and cortex fragments were removed. One third were immediately fixed and processed for histology (T 0 -control). The remaining fragments were divided in two treatments, T I -toxicity test and T II -vitrification. In T I the fragments were exposed to only cryoprotectants and fixed in Carnoy and in T II were utilized DMSO and ethylene glycol supplemented with sucrose. The results indicated that the primordial follicles when vitrified or exposed to cryprotectants did not show variation in morphometry and amount of cells of the granulosa (p>0.05), exception to primary follicles (p<0.05) when compared to T 0 . The analysis of the nuclear chromatin of follicles vitrified or cryprotectants exposed showed significant difference (p<0.05) when compared to T 0 with the control treatment, particularly in T II . In conclusion the vitrification technique affected negatively the morphology of bovine follicles enclosed in ovarian tissue.
RESUMOA criopreservação de folículos ovarianos pré-antrais pode ajudar na conservação de muitas espécies domésticas e selvagens. Neste trabalho, objetivou-se verificar o efeito do etileno glicol, em diferentes concentrações, na morfometria e número de células da granulosa de folículos pré-antrais inclusos em tecido ovariano bovino. O teste de toxicidade foi realizado com fragmentos ovarianos expostos ao etileno glicol em concentrações de 10, 20 ou 40%. O tecido foi analisado por técnica histológica clássica. Os folículos primordiais expostos à concentração de 40% apresentaram redução do diâmetro folicular e ovocitário quando comparados ao grupo controle (sem exposição), 10% e 20% (P<0,05) e redução do número de células da granulosa quando comparados ao grupo controle (P<0,05). Com relação aos folículos primários, nenhum efeito foi observado (P>0,05). Esses resultados sugerem que folículos primários são mais resistentes aos efeitos do etileno glicol quando comparados aos primordiais. ABSTRACTThe cryopreservation of the ovarian preantral follicles could help the conservation of several domestic and wild animal species. The objective of this study was to verify the effect of different concentrations of ethylene glycol on the morphometry and number of granulosa cells of the preantral ovarian follicles. A toxicity test was conducted with strips of ovarian cortex using ethylene glycol (10, 20 or 40%). Tissue analysis was done using classical histology techniques. The primordial follicles expose at a concentration of 40% showed reduction of the follicular diameter and oocyte when compared to the control (non exposing), 10 and 20% groups (P<0.05) and reduction in the number of granulosa cells compared to the control group (P<0.05). Regarding the primary follicles no alteration could be observed (P>0.05). These results suggest that primary follicles are more resistant to the effect of ethylene glycol when compared to primordial ones.
Introdução: Melitina é um peptídeo formado por 26 aminoácidos e principal composto da peçonha produzida por abelhas do gênero Apis. Estudos pré-clínicos têm demonstrado uma capacidade antineoplásica do composto, evidenciando seu efeito sobre linhagens celulares de câncer. O câncer de pulmão é o câncer que mais mata no mundo, sendo que 90% dos casos da doença estão relacionados a um histórico tabagista. O diagnóstico tardio muitas vezes devido à falta de sintomas clínicos compromete diretamente o tratamento e o prognóstico, gerando uma baixa taxa de sobrevida. Neste sentido, a melitina surge como uma abordagem no tratamento de câncer de pulmão. Objetivo: Realizar uma revisão integrativa da literatura sobre as pesquisas publicadas nos últimos cinco anos sobre o efeito da melitina em linhagens de células tumorais pulmonares humanas. Resultados: Foram encontradas 15 publicações, 10 foram excluídas por não responderem diretamente ao objetivo proposto ou por se tratarem de artigos duplicados e cinco foram escolhidas para compor esta revisão. Discussão: Os artigos avaliados relatam o efeito anticancerígeno da melitina sobre linhagens celulares de câncer de pulmão in vitro e in vivo. Conclusão: Devido sua atividade citotóxica a melitina é uma candidata no desenvolvimento de novos tratamentos de câncer.
